Influence of sediment composition on PAH toxicity using zebrafish (Danio rerio) and Japanese medaka (Oryzias latipes) embryo-larval assays ArchiMer

Due to hydrophobic and persistent properties, polycyclic aromatic hydrocarbons (PAHs) have a high capacity to accumulate in sediment. Sediment quality criteria, for the assessment of habitat quality and risk for aquatic life, include understanding the fate and effects of PAHs. In the context of European regulation (REACH and Water Framework Directive), the first objective was to assess the influence of sediment composition on the toxicity of two model PAHs, benzo[a]pyrene and fluoranthene using 10-day zebrafish embryo-larval assay. This procedure was undertaken with an artificial sediment in order to limit natural sediment variability. Developmental toxicity of PAH mixtures in fish early life stages. Part I: adverse effects in rainbow trout ArchiMer

A new gravel-contact assay using rainbow trout, Oncorhynchus mykiss, embryos was developed to assess the toxicity of polycyclic aromatic hydrocarbons (PAHs) and other hydrophobic compounds. Environmentally realistic exposure conditions were mimicked with a direct exposure of eyed rainbow trout embryos incubated onto chemical-spiked gravels until hatching at 10 °C. Dwarf eelgrasses (Zostera noltei) populations have decreased since 2005 in Arcachon Bay (southwest France). Various stressors have been pointed out, however the role of xenobiotics like pesticides or copper (Cu) and of parameters like water temperature warming have not yet been explored. To determine their impact, Z. noltei individuals were collected in a pollution-free site and transferred to the laboratory in seawater microcosms. The release of polycyclic aromatic hydrocarbons (PAHs) into the environment has increased very substantially over the last decades leading to high concentrations in sediments of contaminated areas. Pesticides represent a major proportion of the chemical pollutants detected in French coastal waters and hence a significant environmental risk with regards to marine organisms. Application of POCIS for the monitoring of pesticides, pharmaceuticals and alkylphenols in marine water ArchiMer

With the new European legislation (the Water Framework Directive, 2000/60/EC), the decision makers responsible for the assessment of the environmental contamination require simple monitoring tools. Passive sampling methods seem to be promising tools for the sampling of polar organic compounds in water, especially in marine waters where important dilutions and low concentrations are usual. They are taking into account the temporal variability and integrating the contamination over period of time. This work presents a study, on a large scale, of the contamination of French Mediterranean coastal waters realized in partnership with the Water Agency Rhône Méditerranée Corse (AERMC), and Coastal Water Quality Offices (CQEL).

Long-term disruption of growth, reproduction, and behavior after embryonic exposure of zebrafish to PAH-spiked sediment ArchiMer

A natural sediment spiked with three individual polycyclic aromatic hydrocarbons (PAHs; pyrene, phenanthrene and benzo[a]pyrene) was used to expose zebrafish embryos and larvae during 4 days. The total PAH concentration was 4.4 μg g−1 which is in the range found in sediment from contaminated areas. Quantification of metabolites in the larvae after exposure confirmed the actual contamination of the larvae and indicated an active metabolism especially for pyrene and benzo[a]pyrene. After a transfer in a clean medium, the larvae were reared to adulthood and evaluated for survival, growth, reproduction, and behavior. While they had previously presented a hight stability, unlike in many coastal sites worldwide, eelgrass beds (Zostera noltii and Zostera marina) of Arcachon Bay have declined sharply since the 2000s. Various causes have been invoked to explain this phenomenon: climate anomalies, disease, over-grazing by herbivorous birds, contamination by herbicides, mechanical destruction during clam fishing. Ce projet a permis de mettre en évidence le potentiel des techniques d'échantillonnage passif (DGT, POCIS, SBSE) à "grande échelle" (18 masses d'eau côtières: 6 récifales, 10 "mer ouverte" et 2 portuaires) au cours de 2 campagnes réalisées en période sèche et humide (post cyclonique). L'étude montre l'opérationnalité de ces systèmes en terme de réduction des coûts et de rapidité d'obtention des résultats, mais aussi en terme de facilité de mise en œuvre (par du personnel préalablement formé). Ces techniques ont pu être transférées aux acteurs locaux en charge de la surveillance de l'état de la contamination chimique des eaux. To better understand the effects of PAH mixture toxicity on fish early life stages, this study compared the developmental toxicity of three PAH complex mixtures. These mixtures were extracted from a PAH-contaminated sediment (Seine estuary, France) and two oils (Arabian Light and Erika). L'objectif principal de cette première étude était d'évaluer, dans le contexte guyanais, l'opérationnalité de techniques d'échantillonnage et de faire une évaluation préliminaire de la contamination chimique de différentes masses d'eaux guyanaises (2 sites en eau de transition et 1 site en eau côtière). Deux types d'échantillonneurs passifs ont été mis en oeuvre: DGT (pour les métaux traces) et SBSE (pour les composés organiques hydrophobes). Les molécules hydrophiles ont été recherchées directement dans l'eau prélevée.
