Human‐mediated transport creates secondary contacts between genetically differentiated lineages, bringing new opportunities for gene exchange. When similar introductions occur in different places, they provide informally replicated experiments for studying hybridisation. We here examined 4,279 Mytilus mussels, sampled in Europe and genotyped with 77 ancestry‐informative markers. We identified a type of introduced mussels, called “dock mussels,” associated with port habitats and displaying a particular genetic signal of admixture between M. edulis and the Mediterranean lineage of M. galloprovincialis. These mussels exhibit similarities in their ancestry compositions, regardless of the local native genetic backgrounds and the distance separating colonised...

DNA damage in dab (Limanda limanda) and haddock (Melanogrammus aeglefinus) from European seas ArchiMer
Hylland, Ketil; Skei, Bjorn Borge; Brunborg, Gunnar; Lang, Thomas; Gubbins, Matthew J.; Le Goff, Jeremie; Burgeot, Thierry.
Dab (Limanda limanda) and haddock (Melanogrammus aeglefinus) were collected from coastal and offshore locations in the Baltic (dab only), North Sea (haddock from one location only) and Iceland. Blood was analysed for DNA strand breaks using the comet assay and liver samples for DNA adduct concentrations. DNA strand breaks were at background levels in dab from the two Iceland locations and from the Dogger Bank. The highest levels were observed in dab from the Firth of Forth, Ekofisk and the German Bight. Hepatic DNA adducts in dab were highest at Ekofisk, in the Baltic and Dogger Bank, below detection limit in dab from Iceland and low in dab from the Firth of Forth and German Bight. There was large variation in DNA strand breaks between locations and...

L'éthoxyrésorufine-o-déethylase, les adduits à l'ADN et les mocronuclei dans les organismes marins. Application à la surveillance des effets biologiques sur les côtes françaises ArchiMer
Burgeot, Thierry.
Three biomarkers [ethoxyresorufm-o-deethylase (EROD), DNA adducts and micronuclei (MN) were studied for biomonitoring applications along French coasts. EROD was developed at an experirnental stage in two pilot sites, Seine Bay and Fos sur Mer, using the dab (Limanda limanda), dragon et (Callionymus lyra), red mullet (Mullus barbatus) and comber (Serranus hepatus) as target species. Higbest EROD activity was observed in the subestuarial part of Seine Bay and at the stations closest to sources of pollution from Cortiou and Fos sur Mer. The suitable number of fish for detection of a significant difference between two EROD activities was deterrnined as a function of sampling variance and a preselected statistical power. A preliminary study on hepatocytes in a...

Huvet, Arnaud; Royer, J.; Moal, J.; Burgeot, Thierry; Lapègue, Sylvie; Boulo, Viviane; Nicolas, Jean-louis; Lambert, C..

Tributyltin: A Bottom–Up Regulator of the Crangon crangon Population? ArchiMer
Parmentier, Koen F. V.; Verhaegen, Yves; De Witte, Bavo P.; Hoffman, Stefan; Delbare, Daan H. R.; Roose, Patrick M.; Hylland, Ketil D. E.; Burgeot, Thierry; Smagghe, Guy J.; Cooreman, Kris.
The restrictions and the concerted action of the global ban on the use and presence of tributyltin (TBT) in marine applications to protect ecosystems in the marine environment in 2008 was mainly based on the economic impact on shellfish industries and the dramatic extinction of local mollusk populations in the past. In contrast to the vast datasets on effects on mollusks, the knowledge on impacts on species from other taxa remained in the uncertain until almost two decades ago. The assumption on a long-term TBT-mediated pernicious metabolic bottom–up regulation of the crustacean Crangon crangon population was provoked by the outcome of an EU-project ‘Sources, Consumer Exposure and Risks of Organotin Contamination in Seafood.’ This study reported high TBT...

Pollutant effects on Pacific oyster, Crassostrea gigas (Thunberg), hemocytes: Screening of 23 molecules using flow cytometry ArchiMer
Gagnaire, Beatrice; Thomas Guyon, H; Burgeot, Thierry; Renault, Tristan.
The shellfish industry is an important economic activity in France, occurring mostly in estuarine zones subject to pollution due to anthropogenic activities. The harmful effects of pollutants on species inhabiting these estuarine zones are not well known. Among marine species, bivalve mollusks---particularly Pacific oyster, Crassostrea gigas-may serve a model of interest. The species is sedentary and filter-feeding, which favors bioaccumulation of pollutants in their tissues. Oysters may be suitable for studies on disturbance by pollutants of physiological activities, among which defense mechanisms are poorly documented in bivalves. In this study, effects of pollutants on hemocyte functions were monitored in Pacific oyster, C. gigas. Hemocytes were exposed...

Effects of water accommodated fractions of crude oils and diesel on a suite of biomarkers in Atlantic cod (Gadus morhua) ArchiMer
Holth, T. F.; Eidsvoll, D. P.; Farmen, E.; Sanders, M. B.; Martinez-gomez, C.; Budzinski, H.; Burgeot, Thierry; Guilhermino, L.; Hylland, K..
The aim of this study was to characterise concentration- and time-dependent responses in juvenile Atlantic cod (Gadus morhua) following exposure for one and three weeks to the water-soluble fraction (WAF) of three weathered oils: Arabian Light crude oil (ALC), North Sea crude oil (NSC) and ship-diesel. The sum of polycyclic aromatic hydrocarbons (PAH) in water was highest after one week of exposure and within environmentally relevant concentrations. PAH metabolites in bile confirmed exposure to and uptake of PAHs. Hepatic cytochrome P450 1A (CYP1A) gene expression (here and later the phrase gene expression is used synonymously to gene transcription, although it is acknowledged that gene expression is also regulated by, e.g., translation and protein...

Histopathological lesions and DNA adducts in the liver of European flounder (Platichthys flesus) collected in the Seine estuary versus two reference estuarine systems on the French Atlantic coast ArchiMer
Cachot, Jerome; Cherel, Yan; Larcher, Thibaut; Pfohl-leszkowicz, Annie; Laroche, Jean; Quiniou, Louis; Morin, Jocelyne; Schmitz, Julien; Burgeot, Thierry; Pottier, Didier.
An epidemiological survey was conducted in the Seine estuary and in two smaller and relatively preserved estuaries on the French Atlantic coast in order to estimate the occurrence of liver lesions in European flounder, Platichthys flesus, and also to seek putative risk factors for the recorded pathologies. Four hundred and seventy-eight fish of both sexes and of different size ranges were sampled in the three studied areas, 338 of which in the Seine estuary. All fish were examined for histopathological liver lesions, while DNA adducts and otoliths were analyzed on a subsample. Five categories of hepatic lesions were recorded with the following prevalence for the Seine estuary: 36.7 % inflammations, 8 % parasites (mainly encysted nematodes), 6.5 % foci of...

Many maritime countries in Europe have implemented marine environmental monitoring programmes which include the measurement of chemical contaminants and related biological effects. How best to integrate data obtained in these two types of monitoring into meaningful assessments has been the subject of recent efforts by the International Council for Exploration of the Sea (ICES) Expert Groups. Work within these groups has concentrated on defining a core set of chemical and biological endpoints that can be used across maritime areas, defining confounding factors, supporting parameters and protocols for measurement. The framework comprised markers for concentrations of, exposure to and effects from, contaminants. Most importantly, assessment criteria for...

Effects of increasing temperatures on biomarker responses and accumulation of hazardous substances in rope mussels (Mytilus galloprovincialis) from Bizerte lagoon ArchiMer
Kamel, Naouel; Burgeot, Thierry; Banni, Mohamed; Chalghaf, Mohamed; Devin, Simon; Minier, Christophe; Boussetta, Hamadi.
This study examined the influence of increasing temperatures in spring and summer on biochemical biomarkers in Mytilus galloprovincialis mussels sampled from Bizerte lagoon (northern Tunisia). Spatial and seasonal variations in a battery of seven biomarkers were analyzed in relation to environmental parameters (temperature, salinity and pH), physiological status (condition and gonad indexes), stress on stress (SoS) and chemical contaminant levels (heavy metals, PAHs and PCBs) in digestive glands. Integrated Biological Response (IBR) was calculated using seven biomarkers (AChE, BPH, MXR, GST, CAT, MDA and MT). Seasonal variations in biological response were determined during a critical period between spring and summer at two sites, where chemical...

Parental exposure to the herbicide diuron results in oxidative DNA damage to germinal cells of the Pacific oyster Crassostrea gigas ArchiMer
Barranger, Audrey; Heude-berthelin, Clothilde; Rouxel, Julien; Adeline, Beatrice; Benabdelmouna, Abdellah; Burgeot, Thierry; Akcha, Farida.
Chemical pollution by pesticides has been identified as a possible contributing factor to the massive mortality outbreaks observed in Crassostrea gigas for several years. A previous study demonstrated the vertical transmission of DNA damage by subjecting oyster genitors to the herbicide diuron at environmental concentrations during gametogenesis. This trans-generational effect occurs through damage to genitor-exposed gametes, as measured by the comet-assay. The presence of DNA damage in gametes could be linked to the formation of DNA damage in other germ cells. In order to explore this question, the levels and cell distribution of the oxidized base lesion 8-oxodGuo were studied in the gonads of exposed genitors. High-performance liquid chromatography...

Nutrition and reproduction are key parameters in the interaction process leading to Crassostrea gigas oyster summer mortality in France ArchiMer

Complex interactions between oyster, environment and pathogens were observed during the MOREST (2001-2005) project on summer mortality events of Crassostrea gigas oysters in France. The talk will show how nutrition level increasing reproductive intensity generates a risk for pathogen infection under synergistic effect of temperature and stress. Moreover, a genetic component is associated to specific reproductive performances and spawning behaviour. To better understand origin of such resistance or susceptibility to summer mortality, other characteristics as stress susceptibility, energy management and hemocyte activities of oysters from the two phenotypes were compared. This first comparison suggests interactions between stress, reproduction and defence...

A novel "Integrated Biomarker Response" calculation based on reference deviation concept ArchiMer
Sanchez, Wilfried; Burgeot, Thierry; Porcher, Jean-marc.
Multi-biomarker approaches are used to assess ecosystem health and identify impacts of environmental stress on organisms. However, exploration of large datasets by environmental managers represents a major challenge for regulatory application of this tool. Several integrative tools were developed to summarize biomarker responses. The aim of the present paper is to update calculation of the "Integrated Biological Response" (IBR) described by Beliaeff and Burgeot (Environ Toxicol Chem 21:1316-1322, 2002) to avoid weaknesses of this integrative tool. In the present paper, a novel index named "Integrated Biological Responses version 2" based on the reference deviation concept is presented. It allows a clear discrimination of sampling sites as for the IBR, but...

Multigenerational exposure of the microalga Tetraselmis suecica to diuron leads to spontaneous long-term strain adaptation. ArchiMer
Stachowski-haberkorn, Sabine; Jerome, Marc; Rouxel, Julien; Khelifi, Celia; Rince, Maeva; Burgeot, Thierry.
To investigate the ability of microalgae to develop stable, long-term resistance to herbicides, the marine microalga Tetraselmis suecica was exposed to the herbicide diuron (5 µg/L) for a 43-generation exposure period followed by a 12-generation depuration phase. During the first 25 generations, diuron-exposed cultures showed doubling times ranging from 1.95 to 2.6 days, which was 2 to 2.5-fold longer than control cultures. Between generations 25 and 38, during diuron exposure, two out of the three exposed cultures exhibited a spontaneous drop in doubling time. These results provided evidence of culture adaptation to diuron. To assess persistence of the diuron adaptation observed on growth performance, one of the adapted cultures (D3) was maintained for 12...

Isolation and primary culture of gill and digestive gland cells from the common mussel Mytilus edulis ArchiMer
Faucet, Jerome; Maurice, Manuelle; Gagnaire, Beatrice; Renault, Tristan; Burgeot, Thierry.
As the marine mussel Mytilus edulis is commonly used as a sentinel species, it would be useful to develop a primary culture of the target organs most often in contact with the marine environment. This study reports an improved method for dissociating the digestive gland and gills of M. edulis and considers the effect of mussel storage on cell viability and functionality before culture initiation. Viability and enzymatic activities such as those of esterase and peroxidase were monitored by flow cytometry, a sensitive, objective technique allowing large volumes of cells to be counted within a short time. A primary culture of digestive gland showed more than 75% viability after 72 h. Mussels were maintained in an aquarium containing clean, oxygenated seawater...

Genitors of the Pacific oyster Crassostrea gigas were submitted during gametogenesis to a short pulse exposure to the herbicide diuron at a realistic environmental concentration. Histological analysis showed no effect of diuron on gametogenesis course, sex ratio and reproductive effort. A non-significant increase in testosterone and progesterone levels was observed in genitors exposed to the herbicide. At cell level, diuron exposure was shown to modulate the phagocytic activity of circulating hemocytes. The results of a transcriptional analysis showed that diuron affected the expression of genes belonging to functions known to play a major role during oyster gametogenesis such as gene transcription regulation, DNA replication and repair, DNA methylation...
