Additive transcriptomic variation associated with reproductive traits suggest local adaptation in a recently settled population of the Pacific oyster, Crassostrea gigas 5
Sussarellu, Rossana; Huvet, Arnaud; Lapegue, Sylvie; Quillen, Virgile; Lelong, Christophe; Cornette, Florence; Jensen, Lasse Fast; Bierne, Nicolas; Boudry, Pierre.
Background Originating from Northeast Asia, the Pacific oyster Crassostrea gigas has been introduced into a large number of countries for aquaculture purpose. Following introduction, the Pacific oyster has turned into an invasive species in an increasing number of coastal areas, notably recently in Northern Europe. Methods To explore potential adaptation of reproductive traits in populations with different histories, we set up a common garden experiment based on the comparison of progenies from two populations of Pacific oyster sampled in France and Denmark and their hybrids. Sex ratio, condition index and microarray gene expression in gonads, were analyzed in each progeny (n = 60). Results A female-biased sex-ratio and a higher condition index were...

Ropert, Michel; Blin, J.l.; Cornette, Florence; Pacary, S; Pien, S; Leganeur, E; Kopp, Joel; Richard, O.
Since 1998, a growth and quality monitoring study of reared molluscs has been developped as part of a close collaboration between the Shellfish Research Laboratory of Normandy (IFREMER Port-en-Bessin, Calvados) and the SMEL (Syndicat Mixte d'Expansion du Littoral) Experimental Centre (Blainville, Manche). REMONOR (REseau MOllusque de NORmandie) is based on the experience gained through the national monitoring network REMORA, which protocol has been entirely applied. Adults and spat of Pacific oysters Crassostrea gigas were placed in 13 stations all along the oysters rearing areas of Normandy. These oysters had the same origin and history (i.e. age and weight) until they were dispatched on the 13 stations. 4 essential criteria were used to monitor rearing...

Multiplex PCR sets of novel microsatellite loci for the great scallop Pecten maximus and their application in parentage assignment 5
Morvezen, Romain; Cornette, Florence; Charrier, Gregory; Guinand, Bruno; Lapegue, Sylvie; Boudry, Pierre; Laroche, Jean.
We report the isolation, development and multiplex optimisation of 12 new microsatellite loci for the great scallop, Pecten maximus. Diversity was moderate to high, with number of alleles ranging from 4 to 20 and observed heterozygosity between 0.28 and 0.88. Progeny produced in a commercial hatchery was used to test locus power for parentage assignment. The percentage of offspring that was unambiguously assigned to a unique pair of parents was 97% (software package CERVUS-COLONY). Parentage assignment revealed that 22% of the studied progeny resulted from unplanned crosses. Effective population size of the study progeny was also estimated. Our study illustrates the power of microsatellites for the genetic monitoring of hatchery-produced great scallops.

Population genomics shed light on the demographic and adaptive histories of European invasion in the Pacific oyster, Crassostrea gigas 5
Rohfritsch, Audrey; Bierne, Nicolas; Boudry, Pierre; Heurtebise, Serge; Cornette, Florence; Lapegue, Sylvie.
Crassostrea gigas originated from the Pacific coast of Asia, but was introduced into several European countries in the early 1970s. Natural populations have now spread across the length of the western seaboard of Europe. To elucidate the demographic and selective processes at play during this rapid expansion, genome-scan analysis was performed on different populations. High diversities and low differentiation were observed overall, but significant genetic differentiation was found among newly established populations and between the newly established northern group and a nearly panmictic group composed of southern European populations and a population from Japan. Loss of genetic diversity was also seen in the north, likely caused by founder events during...

Genetic Characterization of Cupped Oyster Resources in Europe Using Informative Single Nucleotide Polymorphism (SNP) Panels 5
Lapegue, Sylvie; Heurtebise, Serge; Cornette, Florence; Guichoux, Erwan; Gagnaire, Pierre-alexandre.
The Pacific oyster, Crassostrea gigas, was voluntarily introduced from Japan and British Columbia into Europe in the early 1970s, mainly to replace the Portuguese oyster, Crassostrea angulata, in the French shellfish industry, following a severe disease outbreak. Since then, the two species have been in contact in southern Europe and, therefore, have the potential to exchange genes. Recent evolutionary genomic works have provided empirical evidence that C. gigas and C. angulata exhibit partial reproductive isolation. Although hybridization occurs in nature, the rate of interspecific gene flow varies across the genome, resulting in highly heterogeneous genome divergence. Taking this biological property into account is important to characterize genetic...

GenoPopTaille. Moving stock assessment into the future 5
Marandel, Florianne; Biais, Gerard; Bidault, Adeline; Charrier, Grégory; Cornette, Florence; Lapegue, Sylvie; Laroche, Jean; Le Cam, Sabrina; Stephan, Eric; Trenkel, Verena; Lorance, Pascal.
At the ecosystem level, sustainable exploitation of fisheries resources depends not only on the status of target species but also on that of bycatch species, some of which are even more sensitive to exploitation. This is the case for a number of elasmobranchs (skates, rays and sharks) species whose abundance declined during the 20th century. Further, the biology of elamobranchs is still poorly known and traditional fisheries stock assessment methods using fisheries catches and scientific survey data for estimating abundance are expensive or even inapplicable due to the small numbers observed. The GenoPopTaille project attempts to apply to the case of the thornback ray (Raja clavata) recent genetic-based methods for absolute population abundance estimation...

Chromosome loss in tetraploid families of the Pacific oyster Crassostrea gigas 5
Mccombie, Helen; Lapegue, Sylvie; Cornette, Florence; Ledu, Christophe; Boudry, Pierre.
Alterations of chromosome number have been observed in the somatic tissue of Crassostrea gigas diploids and artificial polyploids bred in this species. Tetraploid animals with lower chromosome counts in some or all of their somatic tissue are considered undesirable as parents either for commercial triploids (produced via a cross with diploids) or for tetraploid breeding because (1) they may confer the tendency to lose chromosomes and revert to lower ploidy levels to their offspring or (2) their offspring could have a lower ploidy directly because of potential links between somatic and gametic chromosome loss. The present study evaluated the phenomenon in bi-parental tetraploid families bred from parents of differing ploidy quality, including a...

Cornette, Florence; Grouhel, Stephanie; Sharbel, Tim; Boudry, Pierre; Goulletquer, Philippe; Lapegue, Sylvie.
Oocyte activation in Mytilus edulis by Crassostrea gigas spermatozoa 5
Mccombie, Helen; Cornette, Florence; Beaumont, Andy; Boudry, Pierre.
The development of hatchery production techniques for blue mussels is receiving increased research attention in Europe as it could offer an alternative source of spat for the industry ('Blue Seed' Project, Kamermans ICSR 2007). Among the necessary techniques are those of controlled spawning and fertilization, and techniques of triploidisation that can offer increased seasonal availability, yield and/or decreased growing time in the subsequent production stages. In several experiments, Mytilus edulis oocytes, obtained by thermally induced spawning, were fertilized with Mytilus edulis spermatozoa obtained in the same way or by stripping. Parallel treatments examined the effect of mixing Mytilus edulis oocytes with spermatozoa stripped from the Pacific...

Lapegue, Sylvie; Heurtebise, Serge; Cornette, Florence; Brizard, Raphael; Sharbel, Tim; Boudry, Pierre.

Application of high-throughput sequencing to population differentiation in the cupped oysters Crassostrea angulata/C. gigas 5
Gagnaire, Pierre-alexandre; Cornette, Florence; Heurtebise, Serge; Auge, M. T.; Bierne, Nicolas; Lapegue, Sylvie.
The cupped oysters Crassostrea angulata and Crassostrea gigas have been sequentially introduced in Europe from their native Pacific area for aquaculture purposes. Ongoing viral epidemic disease is responsible for large losses in oyster aquaculture production throughout Europe. In parallel with current efforts in developing selection programs for disease resistance, it becomes critically important to characterize genetic diversity patterns in both native and introduced areas. With the advent of next generation sequencing techniques, the power to resolve the fine-scale genetic structure at a genome-wide scale opens to new perspectives towards genetic resource management. Here, we present a genome-wide screen of genetic variation that combines SNP genotyping...

Boudry, Pierre; Heurtebise, Serge; Collet, Bertrand; Cornette, Florence; Ledu, Christophe; Phelipot, Pascal; Gerard, Andre.
The cupped oyster strain bank was created at Ifremer at La Tremblade in 1992. It was funded by the Poitou-Charentes regional council. Its goals were: 1. to study the intra- and interspecific genetic differentiation within the genus Crassostrea. 2. to study acclimatization potential of foreign oysters in French waters in order to identify species that could replace Crassostrea gigas in case of a major disease outbreak in C. gigas or in case of a need or desire to diversify shellfish production. 3. to study the possibility of interspecific hybridisation in the genus Crassostrea and hybrid performance.

The European flat oyster Ostrea edulis has been considered as the traditional French oyster for a very long time and has now nearly disappeared from our farms, as a consequence of the two successive epizootic of the Marteilia refringens parasite and Bonamia ostreae. The actual quasi monoculture of Crassostrea gigas has made necessary the search for a diversification in oyster culture, including the search for a new start of the indigenous flat oyster culture. The Ifremer has been conducting parallel works since 1985 in order to better understand the biology of the parasite as well as to try and select different lines of flat oyster resistant to Bonamia ostreae.

Boudry, Pierre; Naciri, Yamama; Collet, Bertrand; Cornette, Florence; Heurtebise, Serge; Ledu, Christophe; Chollet, Bruno; Phelipot, Pascal; Gerard, Andre.

Analysis of genome-wide differentiation between native and introduced populations of the cupped oysters Crassostrea gigas and Crassostrea angulata 5
Gagnaire, Pierre-alexandre; Lamy, Jean-baptiste; Cornette, Florence; Heurtebise, Serge; Degremont, Lionel; Flahauw, Emilie; Boudry, Pierre; Bierne, Nicolas; Lapegue, Sylvie.
The Pacific cupped oyster is genetically subdivided into two sister taxa, Crassostrea gigas and C. angulata, which are in contact in the north-western Pacific. The nature and origin of their genetic and taxonomic differentiation remains controversial due the lack of known reproductive barriers and the high degree of morphologic similarity. In particular, whether the presence of ecological and/or intrinsic isolating mechanisms contributes to species divergence is unknown. The recent co-introduction of both taxa into Europe offers a unique opportunity to test how genetic differentiation is maintained under new environmental and demographic conditions. We generated a pseudo-chromosome assembly of the Pacific oyster genome using a combination of BAC-end...

A comparison of growth and biochemical composition of Mytilus galloprovincialis (Lmk.) and Mytilus edulis (L.) on the West coast of Cotentin, Normandy, France 1999-2000 5
Kopp, Joel; Cornette, Florence; Simonne, Charlotte.
The blue mussel, Mytilus edulis has been reared along the Normandy coast line since the 1960s. The gonadal cycle of this mussel species shows a sharp decrease in meat quality during the winter period after spawning. This decline in meat quality is so severe that sales have to be suspended from December to July. Another species of mussel, Mytilus galloprovincialis, which is reared in the lagoons along the French shore of the Mediterranean Sea has a different spawning cycle. An experiment was undertaken to study the meat quality of M. Galloprovincialis throughout the year after the spat were transferred from the Mediterranean Sea to the Normandy coast. This species showed an immediate adaptation to the gonadal cycle of M. edulis. Despite suggestions from...

REseau MOllusques des Rendements Aquacoles huitre creuse (REMORA) ; Résultats des stations nationales. Année 2000 5
Fleury, Pierre-gildas; Cornette, Florence; Claude, Serge; Palvadeau, Hubert; Robert, Stéphane; D'Amico, Florence; Le Gall, Patrik; Vercelli, Catherine.
The monitoring network IFREMER /REMORA is assessing, on a yearly basis, mortality, growth and quality criteria of two oyster batches ("juveniles" = one-year old ; and "adults" = two-years old), distributed among various stations of the main French oyster areas. Samplings are carried out in March, June, September and December. This network is a tool for trend estimations (geographic and chronological, on the national scale) ; it must not be considered as an accurate estimation of professional oyster ongrowings. With the exception of Spring mortalities due to poor initial quality of adult oysters and to difficulties in the counting of juveniles, the year 2000 appeared to be a year of low mortality. 2000 is a year of poor growth (61.5 g in average weight in...

The regional network REMONOR showed 2001's characteristics of growth and quality of oysters (Crassostrea gigas) in the three rearing areas of Normandy. Compared to year 2000, two additional stations along the East Coast have been settled. The summer adults mortalities have been registered between 0 % and 25 %. The average for all the rearing areas reached 6 %. Highest rates have been noticed in the Baie des Veys and the East Coast. The juvenile's mortality occurred during the same season, in summer. This year 2001 is included in lower growth years in all Normandy. This year, the West Coast growth rate appeared to be quit unusual. For the first time growth rates were in the average in the northern area that usually shows very bad performance. We can...

At the end of its third year, the regional network for monitoring REMONOR, confirms the particularities of oyster from Normandy in matter of growth and quality. Results from 2 000 are globally very close of those from 1998. Mortality of adults reached 14% and juvenile 6% which represent a low death rate. For this year the most blow area is the Baie des Veys with 31% death rate at Géfosse. Observed growth this year had been weaker than the past two years despite the spring and automnal growth. At spring, the higher recorded growth rate on the West coast with 0.42% (regional average : 0.29%). This seasonal trend seems more and more obvious every year. Regarding the classification of the special oysters, despite the change of the standard, the Baie des Veys...
