Nitrogenous and phosphorous waste production in a flow-through land-based farm of European seabass (Dicentrarchus labrax) ArchiMer
Lemarie, Gilles; Martin, Jean-louis; Dutto, Gilbert; Garidou, C.
A study on nitrogenous and phosphorous waste production in European seabass (Dicentrarchus labrax) was carried out in a flow-through land-based farm. The main objectives were (i) to calculate nitrogenous (N) and phosphorous (P) waste production (particulate and dissolved) from in situ measurements for different rearing ponds with specific fish biomasses, fish sizes and flow rates, (ii) to establish nitrogenous and phosphorous waste production budgets, ratios and equations, and (iii) to compare, for the whole farm, in situ measurements to estimate N and P waste production from waste equations. Waste production was manifest by an increase in concentrations of total ammonia nitrogen, total Kjeldhal nitrogen, particulate nitrogen, dissolved phosphorus and...

Transcriptomic Profiling of Egg Quality in Sea Bass (Dicentrarchus labrax) Sheds Light on Genes Involved in Ubiquitination and Translation ArchiMer
Zarski, Daniel; Thaovi Nguyen,; Le Cam, Aurelie; Montfort, Jerome; Dutto, Gilbert; Vidal, Marie-odile; Fauvel, Christian; Bobe, Julien.
Variable and low egg quality is a major limiting factor for the development of efficient aquaculture production. This stems from limited knowledge on the mechanisms underlying egg quality in cultured fish. Molecular analyses, such as transcriptomic studies, are valuable tools to identify the most important processes modulating egg quality. However, very few studies have been devoted to this aspect so far. Within this study, the microarray-based transcriptomic analysis of eggs (of different quality) of sea bass (Dicentrarchus labrax) was performed. An Agilent oligo microarray experiment was performed on labelled mRNA extracted from 16 batches of eggs (each batch obtained from a different female) of sea bass, in which over 24,000 published probe arrays were...

Effect of chronic ammonia exposure on growth of European seabass (Dicentrarchus labrax) juveniles ArchiMer
Lemarie, Gilles; Dosdat, Antoine; Coves, Denis; Dutto, Gilbert; Gasset, Eric; Person, Jeannine.
Chronic effects of ammonia were studied in juvenile seabass, Dicentrarchus labrax (mean weight= 11 g), exposed for 63 days to eight stable ammonia concentrations, ranging from 0.24 to 0.90 mg l(-1) unionised ammonia nitrogen (UIA-N), respectively, from 6.1 to 22.3 mg l(-1) total ammonia nitrogen (TA-N). Temperature (21.8 degreesC), pH (8.0), salinity (37.0 ppt), and oxygen concentration (over 80% saturation at the outlet) were maintained constant. Fish were fed using a self-feeder device, and they were starved during the last 8 days. Mortality of 28.9 and 42.6% occur-red within the first 8 days at the two highest UIA-N concentrations, respectively, 0.90 and 0.88 mg l(-1). From days 0 to 55, a 1.8-fold increase in weight gain was observed under the 0.90-mg...

Effect of chronic exposure to ammonia on growth, food utilisation and metabolism of the European sea bass (Dicentrarchus labrax) ArchiMer
Dosdat, Antoine; Person, Jeannine; Coves, Denis; Dutto, Gilbert; Gasset, Eric; Le Roux, Annick; Lemarie, Gilles.
The chronic effects of exposing sea bass (average initial weight 100 g) to ammonia in water at 22 degreesC were first evaluated over a 61-day period (period 1, P1) during which nine different groups were submitted to nine ambient ammonia levels ranging from 0.014 to 0.493 mg 1(-1) NH3-N (0.53-16.11 mg 1(-1) total ammonia nitrogen (TA-N)) and fed using self-feeders. At the end of P1, the fish were starved for 10 days (P2). Their recovery capacity was tested over 43 days (P3) after which the exogenous ammonia supply was stopped in all treatments and the fish were allowed to feed. After 20 days of exposure a highly significant effect of ammonia was evident from the decrease in feeding activity, voluntary feed intake (VFI) and specific growth rate (SGR), and...

Long-term monitoring of individual fish triggering activity on a self-feeding system: An example using European sea bass (Dicentrarchus labrax) ArchiMer
Coves, Denis; Beauchaud, M; Attia, J; Dutto, Gilbert; Bouchut, C; Begout, Marie-laure.
In two experiments, a computerized on-demand feeding system coupled with a PIT tag monitoring device was used to continuously record the triggering activity by ca. 50 individual sea bass for 55 days (Exp. 1, initial average body weight and coefficient of variation, CV: 299 g, 15%) and 69 days (Exp. 2, 157 g, 13%). Each group was stocked in 1 m(3) tanks and exposed to a water temperature of 21 +/- 1 degrees C and a light regime of 16:8 LD. Only 5% of the triggering activity was not associated with simultaneous PIT tag detection. Although each individual was detected via PIT tag during the experiment, only 67% and 74% of the fish actuated the trigger at least once in Exp. 1 and Exp. 2, respectively. Moreover, only two fish in Exp. 1 and one fish in Exp. 2...

Gamete quality and management for in vitro fertilisation in meagre (Argyrosomus regius) ArchiMer
Ramos-júdez, Sandra; González, Wendy; Dutto, Gilbert; Mylonas, Constantinos C.; Fauvel, Christian; Duncan, Neil.
The aquaculture of meagre (Argyrosomus regius) requires methods for the control of reproduction that enable the production of families from specific individuals for selective breeding programs. We experimentally determined the parameters required for an in vitro fertilisation protocol. A total of 14 females and 5 males (mean ± S.D. weights of 20.45 ± 6.22 and 15.94 ± 2.75 kg, respectively) were used. Selected females had vitellogenic oocytes >550 μm in diameter and males had fluid sperm upon application of abdominal pressure. Both sexes were treated with an injection of 15 μg kg−1 of gonadotropin-releasing hormone agonist (GnRHa) to induce oocyte maturation/ovulation and enhance sperm production. To determine the timing of ovulation and window of high...

No efficiency of the lateral system on nocturnal feeding in the European sea bass (Dicentrarchus labrax L.) ArchiMer
Faucher, Karine; Dutto, Gilbert; Coves, Denis; Aubert, Anne; Lagardere, Jean-paul.
In order to evaluate the effect and consequence of lateral system inactivation on fish nocturnal feeding, the differential growth of groups of European sea bass maintained in different rearing conditions were compared. Whereas some fish with intact lateral system (placebo fish) were placed under a photoperiod of 12-L: 12-D, other placebo fish were kept in the dark. In the same way, fish deprived of lateral system by section of their lateral system nerves and antibiotic treatment were placed under a photoperiod of 12-L: 12-D and the others in the dark. For each of these four rearing conditions, two sets of experiment were realized. Percent mortality, feed rhythm, averaged daily feed demand, specific growth rate and feed efficiency were compared among these...

Effect of chronic exposure to ammonia on alterations of proteins and immunoglobulins in sea bass (Dicentrarchus labrax) serum ArchiMer
Coeurdacier, Jean-luc; Dutto, Gilbert.
The relationship between serum protein, immunoglobulin concentrations and protein molecular weight profile (PMWP) alterations of sea bass (131.3 +/- 4.3 g) reared in sea water with sublethal concentrations of ammonia was studied over two periods totalling 116 days. During the exposure period (62 days) the first group (group 1) lived in sea water with 0.204 mg.L-1 unionized ammonia nitrogen equivalent to 12 % of the lethal concentration for 50 % of a population exposed for 96 h to ammonia (96-h LC50), whilst the second group (group 2) lived in sea water with 0.340 mg.L-1 UIA-N equivalent to 20 % of the 96-h LC50 of ammonia. Then, the two groups were left for a recovery period (54 days) in the same water as the control group (group 0). The determination of...

Muscle bioenergetics of two emblematic Mediterranean fish species: Sardina pilchardus and Sparus aurata ArchiMer
Teulier, Loïc; Thoral, Elisa; Queiros, Quentin; Mckenzie, David; Roussel, Damien; Dutto, Gilbert; Gasset, Eric; Bourjea, Jerome; Saraux, Claire.
We investigated links between swimming behavior and muscle bioenergetics in two emblematic Mediterranean fish species that have very different ecologies and activity levels. European sardines Sardina pilchardus are pelagic, they swim aerobically, school constantly and have high muscle fat content. Gilthead seabream Sparus aurata are bentho-pelagic, they show discontinuous spontaneous swimming patterns and store less fat in their muscle. Estimating the proportion of red and white muscle phenotypes, sardine exhibited a larger proportion of red muscle (~10% of the body mass) compared to gilthead seabream (~5% of the body mass). We firstly studied red and white muscle fiber bioenergetics, using high-resolution respirometers, showing a 4-fold higher oxidation...

Tagging juvenile seabass and sole with telemetry transmitters: medium-term effects on growth ArchiMer
Begout, Marie-laure; Coves, Denis; Dutto, Gilbert; Laffargue, Pascal; Lagardere, Francoise.
The effects of tagging with acoustic transmitters on the growth of juvenile seabass, Dicentrarchus labrax (L.) (initial mean mass +/- SD, 173 g +/- 23.4) in a 47 d tank experiment, and sole. Solea solea (L.) ( 103.2 g +/- 14.8) in a 72 d tank experiment and (104.0 g +/- 18.4) in a 58 d salt marsh mesocosm experiment were examined. Twenty externally tagged seabass grew more slowly than the 20 with surgically implanted tags, which reached the same mass as nine control fish. Movements of the external transmitter's harness caused abrasions of the skin and loss of the tag in 60% of the cases. We thus recommend implanting transmitters for telemetry studies of juvenile seabass weighing between 120 and 214 g and carrying a tag that represents 2.2-2.5% of body...

Is total serum protein a good indicator for welfare in reared sea bass (Dicentrarchus labrax)? ArchiMer
Coeurdacier, Jean-luc; Dutto, Gilbert; Gasset, Eric; Blancheton, Jean-paul.
Reared sea bass were submitted to six stressful situations: hyperoxia with or without hypercapnia, increased stocking density in an open flow or recirculating system, transfer to another tank and nodavirus injection. The potentially negative impact of these factors on the lives of sea bass was investigated by measuring 9 water parameters and 19 fish parameters including total serum protein (TSP). TSP has already been used to evaluate stress. The present study investigates data of previous and new experiments, concentrating on the potential use of TSP as a routine indicator to assess welfare in sea bass reared on fish farms. In the current experiment, oxygen and carbon dioxide were seen to affect levels of TSP, but alterations were too erratic to enable...

Small pelagic fish dynamics: a review of mechanisms in the Gulf of Lions ArchiMer
Saraux, Claire; Van Beveren, Elisabeth; Brosset, Pablo; Queiros, Quentin; Bourdeix, Jean-herve; Dutto, Gilbert; Gasset, Eric; Jac, Cyrielle; Bonhommeau, Sylvain; Fromentin, Jean-marc.
Around 2008, an ecosystem shift occurred in the Gulf of Lions, highlighted by considerable changes in biomass and fish mean weight of its two main small pelagic fish stocks (European anchovy, Engraulis encrasicolus; European sardine, Sardina pilchardus). Surprisingly these changes did not appear to be mediated by a decrease in fish recruitment rates (which remained high) or by a high fishing pressure (exploitation rates being extremely low). Here, we review the current knowledge on the population's dynamics and its potential causes. We used an integrative ecosystem approach exploring alternative hypotheses, ranging from bottom-up to top-down control, not forgetting epizootic diseases. First, the study of multiple population characteristics highlighted a...

Food in the Sea: Size Also Matters for Pelagic Fish ArchiMer
Queiros, Quentin; Fromentin, Jean-marc; Gasset, Eric; Dutto, Gilbert; Huiban, Camille; Metral, Luisa; Leclerc, Lina; Schull, Quentin; Mckenzie, David; Saraux, Claire.
Small pelagic fish are key components of marine ecosystems and fisheries worldwide. Despite the absence of recruitment failure and overfishing, pelagic fisheries have been in crisis for a decade in the Western Mediterranean Sea because of a marked decline in sardine size and condition. This situation most probably results from bottom-up control and changes in the plankton community toward smaller plankton. To understand such an unusual phenomenon, we developed an original and innovative experimental approach investigating the mechanisms induced by a reduction in the quantity and size of sardine prey. While experimentations offer the unique opportunity to integrate behavior and ecophysiology in understanding key demographic processes, they remain rarely...

During 84 days, the effects of density on juvenile sea bass (Dicentrarchus labrax) (76 +/- 16 g) were studied in an experimental tank-based flow through system. Performance, stress response and resistance to virus infection were analysed under five stabilized rearing densities: 10, 20, 40, 70 and 100 kg m(-3). Water quality parameters (CO2, total ammonia nitrogen and O-2) were measured and maintained close to the recommended values for farmed sea bass by adjusting water renewal exchange. No significant differences were observed between density treatments, neither on stress response (cortisol) nor susceptibility to nodavirus. With regards to biological performances, the daily feed intake and specific growth rate were significantly lower in fish reared at...

VALDORA 2018. Etude de la perte de poids au jeûne sur deux classes de poids de dorades sauvages, Sparus aurata, maintenues en vivier ArchiMer
Coves, Denis; Dutto, Gilbert; Geoffroy, Thibault.
