In the present study, the subcellular localization of the host red blood cell (RBC) membrane components, the alpha 2-3-linked sialic acid (SA) residues and the lipid bilayer, was observed during the asexual growth of Babesia bovis using two erythrocyte probes, the SA-specific lectin (MALII) and the lipophilic fluorescent (PKH2) probes, respectively. In confocal laser scanning microscopy with MALII, the SA residues on the surface of parasitized RBCs appeared to accumulate into the intracellular parasites as the parasites matured as well as to remain on the surface of extracellular parasites. Throughout the world, infections with the Babesia and Theileria parasites often result in economically significant clinical disease in cattle. We conducted a longitudinal survey of Babesia and Theileria infections in cattle from the Polonnaruwa (n?=?75; dry zone) and Nuwara Eliya (n?=?161; wet zone) districts of Sri Lanka. DNA from blood samples collected in June, September, and December 2014 and March 2015 was screened for Babesia bovis, Babesia bigemina, Theileria annulata and Theileria orientalis using specific polymerase chain reactions (PCRs). An enzyme-linked immunosorbent assay (ELISA) based on the recombinant p23 of Cryptosporidium parvum was established for the detection of antibodies against C. parvum in cattle. The sensitivity and specificity of the ELISA were evaluated with the standard indirect fluorescent antibody test (IFAT) using sporozoites as antigens. Of 77 bovine sera collected from China, 20 (26.0%) were identified as positive by the IFAT. The same samples were tested with the ELISA. The optical densities at 415 nm were compared to the IFAT results and statistically analyzed to designate a provisional cut-off point. As a result, the cut-off point was concluded to be 0.08, which was considered to be the best condition in the light of its sensitivity (80%) and specificity (73.7%).... |

Large numbers of cyclophosphamide- or carrageenan-pretreated immunosuppressed adult ddY mice died within 10 days after inoculation with several species of opportunistic bacteria, including P. aeruginosa, K. pneumoniae, E. coli, S. aureus and methicillin resistant S. aureus. When immunosuppressed mice were administered bacteria in combination with two 100 μg/mouse intramuscular doses of a newly synthesized peptide, Obiopeptide-1 (OP-1), survival rates increased significantly. A heterologous prime-boost immunization regime with priming DNA followed by recombinant vaccinia virus expressing relevant antigens has been shown to induce effective immune responses against several infectious pathogens. In this study, we constructed a recombinant plasmid and vaccinia virus, both of which expressed P50 of Babesia gibsoni, to investigate the immunogenicity and protective efficacy of a heterologous prime-boost immunization against canine babesiosis. We examined the detailed cellular localization of the rhoptry-associated protein-1 (RAP-1) of Babesia bovis merozoites by the post-embedding method of immuno-electron microscopy (IEM) using the anti-RAP-1 monoclonal antibody (mAb) 1C1 in order to substantiate the result in our previous report by an indirect immunofluorescent antibody test and the pre-embedding method of IEM. RAP-1 slightly distributed only in cytoplasm of merozoites, especially around the nucleus, but not in the rhoptry organelle at an early stage after invasion into a red blood cell.
