We report the isolation, development and multiplex optimisation of 12 new microsatellite loci for the great scallop, Pecten maximus. Diversity was moderate to high, with number of alleles ranging from 4 to 20 and observed heterozygosity between 0.28 and 0.88. Progeny produced in a commercial hatchery was used to test locus power for parentage assignment. The percentage of offspring that was unambiguously assigned to a unique pair of parents was 97% (software package CERVUS-COLONY). Parentage assignment revealed that 22% of the studied progeny resulted from unplanned crosses. Effective population size of the study progeny was also estimated. Our study illustrates the power of microsatellites for the genetic monitoring of hatchery-produced great scallops. |

The objective of this study was to develop an integrative approach in ecotoxicology (from biomarkers to population genetics) to assess the ecological status of fish populations. Flounders (Platichthys flesus) collected after the spawning season in the heavily polluted Seine estuary were compared with the moderately polluted Bay of Douarnenez. The muscle energetic reserves were highly depleted in Seine vs. Douarnenez fish. The Seine fish displaying a reduced capacity to manage the oxidative stress and a higher energetic metabolism. An increase in the content of muscle membrane phospholipids (sphingomyelin, phosphatidylserine, free sterols) was detected in the Seine vs. Douarnenez fish. The data integration allowed to hypothesize relationships between... |

An epidemiological survey was conducted in the Seine estuary and in two smaller and relatively preserved estuaries on the French Atlantic coast in order to estimate the occurrence of liver lesions in European flounder, Platichthys flesus, and also to seek putative risk factors for the recorded pathologies. Four hundred and seventy-eight fish of both sexes and of different size ranges were sampled in the three studied areas, 338 of which in the Seine estuary. All fish were examined for histopathological liver lesions, while DNA adducts and otoliths were analyzed on a subsample. Five categories of hepatic lesions were recorded with the following prevalence for the Seine estuary: 36.7 % inflammations, 8 % parasites (mainly encysted nematodes), 6.5 % foci of... |

At the ecosystem level, sustainable exploitation of fisheries resources depends not only on the status of target species but also on that of bycatch species, some of which are even more sensitive to exploitation. This is the case for a number of elasmobranchs (skates, rays and sharks) species whose abundance declined during the 20th century. Further, the biology of elamobranchs is still poorly known and traditional fisheries stock assessment methods using fisheries catches and scientific survey data for estimating abundance are expensive or even inapplicable due to the small numbers observed. The GenoPopTaille project attempts to apply to the case of the thornback ray (Raja clavata) recent genetic-based methods for absolute population abundance estimation... |

A multibiomarker approach was developed to evaluate the juvenile European flounder responses to a complex mixture of 9 polycyclic aromatic hydrocarbons (PAHs) and 12 polychlorinated biphenyls (PCBs). Exposure was performed through contaminated food pellets displaying: (1) PAH and PCB levels similar to those detected in the heavily polluted Seine estuary, respectively in sediments and in flatfish and (2) ten times these concentrations. Several biomarkers of the immune system (e.g., lysozyme concentration and gene expression of complement component C3 and TNF-receptor), DNA damage (e.g., Comet assay), energetic metabolism (e.g., activity of cytochrome C oxidase), detoxification process (e.g., cytochrome P450 1A1 expression level: CYP1A1; betaine homocysteine... |

Phosphoglucomutase is a key enzyme in glycolysis and has been widely studied in vertebrates and some invertebrates but no molecular information is available in marine invertebrates despite the importance of this marker in ecological and genetical studies. In this work, we isolated a cDNA and the corresponding genomic sequence that encode PGM-2 locus in the Pacific oyster Crassostrea gigas. We used sequences drawn from the database to construct an evolutionary framework for examining the position of mollusc PGM sequences among prokaryotic and eukaryotic homologues and showed that oyster PGM gene organization was closer to vertebrates PGM genes than other invertebrates as previously found in other Lophotrochozoa species. We also investigated PGM mRNA... |

The great scallop Pecten maximus is harvested in several European countries and fisheries targeting this species are severely regulated by fishing quotas. In addition, hatchery-based population enhancement has been developed in some countries to provide alternative or complementary production. The genetic structure of wild populations of P. maximus and the potential impact of aquaculture on the genetic diversity of this species remains poorly documented. In this study, we explored the genetic structure of P. maximus using 12 microsatellite markers, considering 14 populations sampled from Galicia (Spain) to the North of Norway, and one population of Pecten jacobaeus (L., 1758) from the Lion Gulf (Mediterranean Sea). Results indicated a clear differentiation... |

Dinoflagellates of the genus Alexandrium are a major cause of harmful algal blooms (HABs) that have increasingly disrupted coastal ecosystems for the last several decades. Microalgae from the genus Alexandrium are known to produce paralytic shellfish toxins (PST) but also bioactive extracellular compounds (BEC) that can display cytotoxic, allelopathic, ichtyotoxic or haemolytic effects upon marine organisms. The objective of this experimental study was to assess the effects of PST and BEC produced by A. minutum upon juvenile great scallops Pecten maximus. Scallops were exposed for one week to two different strains of A. minutum, the first producing both PST and BEC and the second producing only BEC. Escape response to starfish, daily shell growth,... |

Processes regulating population connectivity are complex, ranging from extrinsic environmental factors to intrinsic individual based features, and are a major force shaping the persistence of fish species and population responses to harvesting and environmental change. Here we developed an integrated assessment of demographic and genetic connectivity of European flounder Platichthys flesus in the northeast Atlantic (from the Norwegian to the Portuguese coast) and Baltic Sea. Specifically, we used a Bayesian infinite mixture model to infer the most likely number of natal sources of individuals based on otolith near core chemical composition. Simultaneously, we characterised genetic connectivity via microsatellite DNA markers, and evaluated how the combined... |

Estuaries are important areas highly vulnerable to anthropogenic pollutions. Therefore, the assessment of estuarine water quality is a major ecological issue. In this study, we sampled juveniles of the European flounder in the “pristine” Canche estuary, and caged them in Canche and in two polluted sites of the Seine estuary, Rouen and Fosse Nord. After one month, the metal and organic pollutants in these sites were assessed, and we evaluated several phenotypic indicators (condition index, RNA/DNA ratios and genotoxicity), and extracted the proteins in fish livers for analysis using a shotgun proteomics approach. The results showed strong modifications in the fish caged in both sites of the Seine estuary, as compared to those caged in Canche. In particular,... |
