Artemia in aquatic toxicology: a review OMA
Persoone, G.; Wells, P.G..
Due to commercial availability of dried cysts from which live test material can be hatched at will, Artemia is used extensively in research and applied toxicology.Despite the extensive literature on dose-effect relationships of chemicals on brine shrimp, it was not until 1980 that an experimental protocol was developed for a simple acute toxicity test with Artemia nauplii, meeting the prerequisites for standardization. The reliability and accuracy of this short-term test were determined during an intercalibration exercise involving 80 laboratories and were found to be quite satisfactory. Consequently, the so-called ARC-test, which is one of the very few standardized marine toxicity tests, is now used routinely at the...

The influence of temperature and salinity on the sensitivity of Artemia nauplii to chemical compounds OMA
Persoone, G.; Van Steertegem, M..
The toxicity of chemicals to aquatic organisms is mostly determined in a set of experimental conditions which are close to optimal for the test species. Because in nature organisms have to cope with fluctuating environmental conditions which can range during the course of the year from optimal to suboptimal, up to the limits of the tolerance range, the conditions under which routine bioassays are normally performed do, however, not reflect the large range of situations occurring in the field. Considering the scarcity of information on the effect of fluctuating environmental conditions on the sensitivity of test species to toxicants, a comparative laboratory study has been undertaken to determine the combined influence of environmental variables on the...

Technological improvements for the cultivation of invertebrates as food for fishes and crustaceans: 1. Devices and methods OMA
Persoone, G.; Sorgeloos, P..
Technological problems can be considered as one of the bottlenecks of research concerned with culturing aquatic invertebrates in the laboratory, as well as the pilot and mass production stages. Oxygenation problems, circulation of the food and sedimentation problems can easily be solved by adapting the shape of the culturing vessels, i. e., by using funnel-shaped bottoms or high, narrow cylinders. In both cases, the food particles, settled on a very small bottle area, can easily be blown back into suspension using an air pump, which re-aerates the medium at the same time. A number of culturing devices for different categories of organisms, including algae are described and commented on. The only power source involved is compressed air which aerates, keeps...

The influence of the formulation of the herbicide paraquat on its toxicity for aquatic organisms OMA
Benijts-Claus, C.; Persoone, G..
Short term bio-assays have been carried out on representatives of different trophic levels of the aquatic food chain. The two wetters, which are included in the commercial formulation "Gramoxone" are very harmful for fish and their combination to paraquat increases the toxicity of the herbicide. Algae and crustaceans on the contrary, are more affected by the herbicide than by the wetters, the toxicity to the latter two groups being much higher than to fish.

Research on the culturing of the brine shrimp Artemia salina L. at the State University of Ghent (Belgium) OMA
Sorgeloos, P.; Baeza-Mesa, M.; Benijts, F.; Persoone, G..
In the framework of research on mariculture at the "Institute for Marine Scientific Research" (IZWO), the "Laboratory for Biological Research in Environmental Pollution" at the State University of Ghent, is focusing on culturing problems of the brine shrimp Artemia salina. The following research topics are discussed: 1. The mechanisms leading to the hatching of Artemia cysts are reviewed and complemented with new data on the sensitivity to light and temperature, and on the reversibility of the development within the cyst. The possible influences of the collecting, sorting, and packaging processes on the quality of the cysts are discussed. 2. Since in the literature much confusion exists concerning the mechanisms...

Determination of the toxicity of four heavy metal compounds and three carcinogens using two marine nematode species, Monhystera microphthalma and Diplolaimelloides bruciei OMA
Bogaert, T.; Samoiloff, M.R.; Persoone, G..
Bioassays with two marine nematode species were used to determine the toxicity of four heavy metal compounds and three carcinogens. The test using the marine species Diplolaimelloides bruciei is more sensitive than the existing nematode test using the freshwater nematode Panagrellus redivivus for mercury chloride and 2-acetamidofluorene, but less sensitive for methylmercury chloride, phenacetin, and 4-aminobiphenyl. The Monhystera microphthalma test is less sensitive than the nematode test with P. revivus for mercury chloride, selenium oxide, and cesium chloride. The range between the concentrations at which no toxic effects are observed and the concentrations at which 100%...

Technological improvements for the cultivation of invertebrates as food for fishes and crustaceans: 2. Hatching and culturing of the brine shrimp, Artemia salina L. OMA
Sorgeloos, P.; Persoone, G..
Although extensive fundamental research has been performed on the brine shrimp, the yields from hatching and culturing this organism which is widely used in mariculture, can be greatly improved. By exposing the immersed cysts to light and by using a new type of hatching cylinder and separator box, greater hatching and separation efficiencies can be obtained. Controlled mass culture of brine shrimp larvae to the adult stage has been extremely difficult until now; the literature on the subject is reviewed and a new technique of high density culturing at the pilot-stage is described.

A simple volumeter for small invertebrates OMA
Persoone, G..
The displacement technique for measuring the volume of plankton catches is well established, and a number of papers have been published describing suitable apparatus for this purpose including those by Frolander (1957), and Yentsch & Hebard (1957). Gnanamuthu (1952) reviews several papers dealing with equipment for measuring the volume of small aquatic animals and discusses his own volumeter. The apparatus here described employs a water column and is, therefore, more convenient to handle than previous designs which used a mercury column. It is inexpensive and simple to construct, and will measure to 0.01 or even 0.005 ccm. We have found it most useful in quickly measuring the volume of polychaetes or oligochaetes, small bivalves, small crustaceans, etc.

The ARC-test: a standardized short-term routine toxicity test with Artemia nauplii: methodology and evaluation OMA
Vanhaecke, P.; Persoone, G..
Considering the need for reliable standardized routine toxicity tests for the marine environment, a short-term bioassay with Artemia nauplii has been developed for routine testing. The major reason for the selection of brine shrimp as test species is the continuous availability of Artemia under the form of dry cysts from which the larvae are hatched very easily. This unique advantage solves one of the major problems of aquatic ecotoxicological tests, namely stock recruitment and/or culturing; the Artemia test can be performed anywhere and at any time from dry biological material available "on the shelf".An extensive study has been performed on various brine shrimp tests published in scientific...

Assessment of ammonia toxicity in contaminated sediments in the upper Scheldt (Belgium): the development and application of toxicity identification evaluation procedures OMA
Van Sprang, P.; Janssen, C.R.; Sabayasachi, M.; Benijts, F.; Persoone, G..
The toxicity of sediment pore waters from 4 sites in the Upper Scheldt war characterized using modified Toxicity Identification Evaluation (TIE) procedures The samples from all locations exhibited acute toxicity to the freshwater crustacean Thamnocephalus platyurus. The IIE approach was conducted using an enlarged battery of fractionation tests. Toxicity was removed or reduced significantly by the cation exchange resins and the air-stripping test at pH 11. Additionally, increased toxicity war observed at higher pH while a reduced toxicity war found at a decreased pH. From these TIE studies, ammonia war identified as the dominant toxic agent.

The use of Artemia nauplii for toxicity test: a critical analysis OMA
Sorgeloos, P.; Remiche-Van der Wielen, C.; Persoone, G..
Because of their easy hatching from dry cysts and their year-round availability, nauplii of the brine shrimp Artemia salina are most convenient test organisms for toxicity studies. The bioassays mentioned in literature have, however, mostly been carried out with larvae of which neither the exact instar stage nor the geographical origin is known. The incubation of the cysts should always be carried out under strictly controlled temperature conditions since both the onset of hatching and the length of the hatching period are temperature-dependent. Moreover, the experiments should only be carried out with populations of nauplii of the same stage of development since it is demonstrated that the second and third instars are...

Protozoa as test organisms in marine ecotoxicology: luxury or necessity? OMA
Dive, D.; Persoone, G..
A critical examination of the utility of protozoan bioassays in marine as well as in freshwater ecotoxicology implies the answer to three basic questions: why, how and when. Arguments in favor of tests with Protozoa are: protozoans combine all biological mechanisms and functions in one single cell; the physiology and biochemistry of several species has been studied extensively; the generation time of Protozoa is very short in comparison to Metazoa; large numbers of organisms can be produced in a small volume; induction of cryptobiotic stages is possible with some species; Protozoa play a significant role in aquatic ecosystems; test methodologies with very sensitive criteria can be worked out. The two basic approaches for bioassay are in...

Three simple culture devices for aquatic invertebrates and fish larvae with continuous recirculation of the medium OMA
Sorgeloos, P.; Persoone, G..
The 3 culturing devices described are based on the principle of the classic air-water lift, which facilitates continuous recirculation and aeration of the cultivation medium. The first device is a "circulation cylinder", convenient for hatching eggs of the brine shrimp Artemia salina and for culturing algae and protozoans. In the second device, an internal basket (i.e., a glass cylinder with plankton-gauze bottom) prevents the animals being carried along in the air-water lift. The third device, analogous to the second, is provided with a lateral waste-drain and a water collector with siphon, which empties regularly into the main culture vessel. The second and third devices proved to be excellent for the cultivation of a variety of...

La pollution dans le port d'Ostende et ses répercussions sur la réproduction de l'huître plate dans le Bassin de Chasse = La pollution dans le port d'Ostende et ses répercussions sur la réproduction de l'huître plate dans le Bassin de Chasse OMA
Persoone, G..
The oyster culture, flourishing along the Belgian coast at the beginning of the century, perished entirely in the course of two World Wars. In 1956, it was taken on again intensively in the Scour Basin of Ostend. If we consider the obtained results with the affination of Flat and Portuguese oyster, this 86 ha closed basin, is of an exceptional productivity. Essays on the understanding of the origin of calcified oysters, done from 1960 until 1966, turned out to be very promising and an integral oyster culture from the larvae up to the adult oyster could be observed. In any case the last few years, eventhough compared to before a normal emission of larvae of Ostrea edulis, there has not been a fixation of the origin. Regular samples of...
