New Caledonian shrimp farming feasibility studies started in the mid-seventies. Today ca. 2000 tons of Litopenaeus stylirostris, introduced in 1978, are semi-intensively produced each year. Since fifteen years, production has been impacted by two seasonal vibriosis, the “Summer” syndrome caused by Vibrio nigripulchritudo in relation to pond bottom and water column quality, and the “Winter Syndrome” caused by Vibrio penaeicida in relation to drastic temperature drops . With the ambition of elucidating processes possibly leading to mortality outbreaks in ponds, Ifremer has been conducting a research program since 2003 under the DESANS project (2003-2006) and the present DEDUCTION project, equally funded by the South and North Provinces and by the Government...

Genetic improvement strategy in small aquaculture industries : the new caledonian shrimp experience ArchiMer
Goyard, Emmanuel; Goarant, Cyrille; Brun, Pierre; Herlin, Jose; Pham, Dominique; Beliaeff, Benoit; Harache, Yves; Loubersac, Lionel; Patrois, Jacques.
Shrimp farming in New Caledonia relies on the culture of a domesticated strain of Litopenaeus stylirostris introduced from Mexico at a time when genetic principles were of little or no consideration. Since then, advances in agriculture and for some aquatic species of importance led caledonian shrimp farmers to reconsider the appropriateness of a genetic improvement strategy adapted to local biotechnical and economical constraints. This questioning involves many different and interrelated aspects: scientific and technologic (genetics, biosecurity, quarantine, ), economic and organizational (financing, diffusion of genetic improvement) and pedagogic (awareness of farmers). Local institutions, producers associations, research centre and sanitary services...

Salinité et confort physiologique - Application pratique en élevage larvaire ArchiMer
Pham, Dominique; Mailliez, Jean-rene; Peignon, Jean-marie; Broutoi, Francis; Marteau, Anne-laure; Wabete, Nelly.
The caledonian blue shrimp Litopenaeus stylirostris is the second largest export commodity in New Caledonia. This industry is based on 19 production farms but also four hatcheries, two feed production units and two packing plants. The maximum production was reached in 2005 with 2400 tons. Supply of juveniles by hatchery is the bottle neck of the production chain because this species is not present in the natural environment. Recent problems of larval survival led some scientists to work on eco-physiology to propose adaptations of larval rearing protocol. Salinity of the rearing environment is taken as an example to study the influence of abiotic parameters on the animal physiology according to its development stage. The influence of salinity on survival...

Grow-out of sandfish Holothuria scabra in ponds shows that co-culture with shrimp Litopenaeus stylirostris is not viable ArchiMer
Bell, J; Agudo, N; Purcell, S; Blazer, P; Simutoga, M; Pham, Dominique; Della Patrona, Luc.
We examined the potential for producing the large numbers of sandfish (Holothuria scabra) needed for restocking programmes by co-culturing juveniles with the shrimp Litopenaeus stylirostris in earthen ponds. Our experiments in hapas within shrimp ponds were designed to detect any deleterious effects of sandfish on shrimp, and vice versa. These experiments showed that a high stocking density of juvenile sandfish had no significant effects on growth and survival of shrimp. However, survival and growth of sandfish reared with shrimp for 3 weeks were significantly lower than for sandfish reared alone. Increased stocking density of shrimp also had a significant negative effect on survival and/or growth of sandfish. A grow-out trial of juvenile sandfish in...

Selection and characterization of potential probiotic bacteria for Litopenaeus stylirostris shrimp hatcheries in New Caledonia ArchiMer
Pham, Dominique; Ansquer, Dominique; Chevalier, Anne; Dauga, Clement; Peyramale, Aude; Wabete, Nelly; Labreuche, Yannick.
In New Caledonia, shrimp hatcheries are confronted with mass mortality in the larval stages, a phenomenon poorly understood as no specific causative agent has been identified. This has resulted in an excessive use of prophylactic antibiotics, although their adverse effects in aquaculture are notorious. The present work was thus aimed at selecting potential probiotic strains for penaeid hatcheries. From a pool of more than 400 marine bacterial isolates sampled from the local marine environment seven strains exhibited in vitro antagonistic activity towards Vibrio harveyi. These isolates were characterized both phenotypically and genotypically using a biochemical approach and 16S rDNA sequencing. Six out of these seven strains were found to belong to the...

Salinity tolerance, ontogeny of osmoregulation and zootechnical improvement in the larval rearing of the Caledonian Blue Shrimp, Litopenaeus stylirostris (Decapoda, Penaeidae) ArchiMer
Pham, Dominique; Charmantier, Guy; Wabete, Nelly; Boulo, Viviane; Broutoi, Francis; Mailliez, Jean-rene; Peignon, Jean-marie; Charmantier-daures, Mireille.
The ontogeny of osmoregulationwas investigated in Litopenaeus stylirostris by studying salinity tolerance and osmoregulatory capacity. Shrimp at different larval and postlarval stages were exposed to various salinities and survival was monitored for 24 h. Survival rates exceeded 80% at salinity over 25 ppt (750 mOsm.kg(-1)) at all the stages. At salinities below to 25 ppt, salinity tolerance was higher in nauplii and zoeae than in mysis larvae. Postlarval stages were able to withstand lower salinities, e.g. 6.0 ppt (176 mOsm.kg(-1)) at PL9 stage, but they were more sensitive than larvae to salinities over 35 ppt (1035 mOsm.kg(-1)). Zoea and mysis larvae slightly hyper-regulated at all tested salinities. After metamorphosis, postlarvae progressively...

Live preys in shrimp culture: nutritional and sanitary considerations on the use of Artemia in New Caledonia ArchiMer
Pham, Dominique; Wabete, Nelly; Maillez, Jean-rene; Broutoi, Francis; Chim, Liet.
In aquaculture, Artemia nauplii are commonly used as live prey; and represent 40 % of the total aquaculture feed demands in the early stages. However, the production of Brine shrimp is very unstable from one year to another and in the late 90’s, only 20 % of the world aquaculture need was covered. An insufficient offer leads to higher prices of the Artemia cysts and consequently, the price of shrimp juveniles from hatchery. In New Caledonia, marine aquaculture is mainly based on the blue shrimp Litopenaeus stylirostris. From Zoea 3 to 10 days old post-larvae (P10), the shrimp are mainly fed with live preys. Eleven kilos of Artemia cyst are necessary for producing one million of P10. From a sanitary point of view, Artemia can be a virus carrier and Vibrio...

Residual genetic variability in domesticated populations of the Pacific blue shrimp (Litopenaeus stylirostris) of New Caledonia, French Polynesia and Hawaii and some management recommendations ArchiMer
Goyard, Emmanuel; Arnaud, Sophie; Vonau, Vincent; Bishoff, Vincent; Mouchel, Olivier; Pham, Dominique; Wyban, Jim; Boudry, Pierre.
The Latin American shrimp Litopenaeus stylirostris was introduced in three different Pacific islands (Tahiti, New Caledonia via Tahiti, and Hawaii) and hatchery-propagated for 7-25 generations to develop shrimp farming based on these domesticated stocks. Three microsatellite markers have been used in an attempt to assess the genetic bases of the populations available to start a selective breeding program. The comparison of eight hatchery stocks (five New Caledonian, two Hawaiian and one Tahitian stocks) and one wild Ecuadorian population showed a much lower variability in the domesticated stocks than in the wild population, especially in New Caledonia and Tahiti (2-3.7 vs. 14-27 alleles per locus; 20-60% vs. 90% expected heterozygosity). The Tahitian and...

Probiotic P-acidilactici application in shrimp Litopenaeus stylirostris culture subject to vibriosis in New Caledonia ArchiMer
Castex, Mathieu; Chim, Liet; Pham, Dominique; Lemaire, Pierrette; Wabete, Nelly; Nicolas, Jean-louis; Schmidely, P; Mariojouls, C.
We studied the effects of a lactic acid bacterium, Pediococcus acidilactici (strain MA 18/5M, CNCM), as a dietary probiotic on growth performance and some nutritional and microbiological aspects of the shrimp Litopenaeus stylirostris. Experimental shrimp culture was carried out over 10 weeks, using floating cages of 14 m(2) each set in earthen ponds, in a New Caledonia farm affected by "Summer syndrome", a septicaemic vibriosis caused by Vibrio nigripulchritudo. The experiment design testing probiotic-coated pellets against control was replicated in two earthen ponds. High mortalities, characteristic of Summer syndrome, were observed during the first 2 weeks of the trial. The probiotic improved production in the treated cages from both ponds with increases...

Ontogeny of osmoregulation in the Pacific blue shrimp, Litopenaeus stylirostris (decapoda, penaeidae): Deciphering the role of the Na+/K+-ATPase ArchiMer
Pham, Dominique; Charmantier, Guy; Boulo, Viviane; Wabete, Nelly; Ansquer, Dominique; Dauga, Clement; Grousset, Evelyse; Labreuche, Yannick; Charmantier-daures, Mireille.
The role of the main ion transporting enzyme Na +/K + − ATPase in osmoregulation processes was investigated in Litopenaeus stylirostris. The development and localization of the osmoregulation sites were studied during ontogenesis by immunodetection of Na+ K+-ATPase using monoclonal antibodies and transmission electron microscopy (TEM). Osmoregulation sites were identified as the pleurae and branchiostegites in the zoeae and mysis stages. In the subsequent post-metamorphic stages the osmoregulatory function was mainly located in the epipodites and branchiostegites and osmotic regulation was later detected in the gills. The presence of ionocytes and microvilli in these tissues confirmed their role in ionic processes. The complete open reading frame of the...

Caractéristiques des Artemia d’origine russe ArchiMer
Pham, Dominique.
New Artemia strain from Russia are now available on the market. They have been evaluated by comparison with Artemia from Great Salt Lake which are routinely used at the LAC hatchery,. Five criteria were compared and all of them were in favour of the American strain with 191 833 nauplii per gram while the Russian ones only gave 102 667 nii per gram. Another important criteria when using Artemia was hatching time which turned out to be longer for the Russian.

Amélioration génétique pour la résistance au Syndrome 93 : bilan de 5 générations de sélection expérimentale ArchiMer
Goyard, Emmanuel; Ansquer, Dominique; Brun, Pierre; De Decker, Sophie; Dufour, Robert; Goarant, Cyril; Patrois, Jacques; Peignon, Jean; Pham, Dominique.
Selective breeding of shrimp for viral resistance is established for species like L. vannamei and L. stylirostris. However, selection for bacterial infection resistance is not documented. In New Caledonia, "Syndrome 93" is a vibriosis that affects cultured shrimps during the cool season with survival from 15 to 30%. Current practises for breeders rearing at low densities do not improve the resistance of the offsprings mainly because selection pressures for "Syndrome 93" are low. Even after the fifth generation, survival improvement is not significant.

Evaluation of floating cages as an experimental tool for marine shrimp culture studies under practical earthen pond condition ArchiMer
Castex, Mathieu; Chim, Liet; Lemaire, Pierrette; Wabete, Nelly; Pham, Dominique.
Scientific and technical studies carried out under semi-intensive farming conditions, with large earthen ponds (Fig 1), to assess the effects of treatments on zootechnical results are highly complex due to the many logistical, economic and/or experimental constraints involved. In our study, we evaluated the use of floatingcages (Paquotte et al., 1998) immersed in earthen ponds as an experimental tool in order to design future powerful and economical expe-riments under semi-intensive farming conditions.

Reproduction of the hawaiian strain of pacific blue shrimp Litopenaeus stylirostris in New Caledonia. ArchiMer
Pham, Dominique; Patrois, Jacques; Goyard, Emmanuel; Maillez, Jean-rene; Broutoi, Francis; Dufour, Robert; Peignon, Jean-marie; Brun, Pierre; Lambert, Christian; Pita, Etienne.
The Pacific blue shrimp Litopenaeus stylirostris was introduced in New Caledonia thirty years ago. Because of its high inbreeding, a SPF strain domesticated in Hawaii, genetically differentiated from the Caledonian strain, had to be imported. The two strains’ reproductive performances were compared at different periods and cross breedings were assessed. The average results show that Caledonian animals give twice as many nauplii than the Hawaiian animals. The best cross breeding is obtained with Caledonian male x Hawaiian female. A possible explanation is the later age or weight of maturity for the Hawaiian strain compared to the Caledonian strain, difference which could come from the geographical origins and/or the zootechnical and climatic domestication...

Floating cages as an experimental tool for shrimp culture studies: first attempts to check their reliability ArchiMer
Pham, Dominique; Castex, Mathieu; Wabete, Nelly; Lemaire, Pierrette; Brun, Pierre.
The first aim of this study was to evaluate on a technical basis the rearing of shrimps in floating cages set up in earthen ponds. Shrimps reared in and outside the cages presented comparable growths and survival rates. Then it was concluded that shrimp culture in floating cages did not present zootechnical biases comparing to pond culture. The second objective of this study was to assess the present method as a statistical point of view. We found that for expected differences of 20% to the control mean, 3 and 6 floating cages per treatment will be reasonable to determined statistical differences for growth parameters and survival. Furthermore, we showed a significant pond effect as regard as survival and growth between both sets of cages. This results...

Contribution à l'amélioration des survies et performances de reproduction de L. stylirostris en saison fraîche en Nouvelle-Calédonie ArchiMer
Pham, Dominique; Wabete, Nelly; Lemaire, Pierrette; Mailliez, Jean-rene; Broutoi, Francis; Chim, Liet.
Since 1993, the New Caledonian shrimp industry has been faced with cold season mortality outbreaks of multifactorial origin. This pathology called “Syndrome 93” affects as well animals in grow-out ponds as broodstock after their transfer in maturation room, with up to 35% of mortality in 48 hours. From literature data and physiological studies on Litopenaeus stylirostris, several combinations of three abiotic parameters (temperature, salinity and feeding) have been tested during the breeders stocking period. This research allowed us to lower stress factors impact and to propose a transfer protocol which leads to an optimal physiological comfort of the animal. Results showed that survival rate is improved and spawning rates are twice higher. These findings...

Evaluation of floating cages as an experimental tool for marine shrimp culture studies under practical earthen pond conditions ArchiMer
Chim, Liet; Castex, Mathieu; Pham, Dominique; Brun, Pierre; Lemaire, Pierrette; Wabete, Nelly; Schmidely, P; Mariojouls, C.
The New Caledonia blue shrimp Litopenaeus stylirostris is commercially produced under semi-intensive rearing conditions. The size of the farming earthen ponds (5–10 ha) and the subsequent production constraints make it difficult to use them as experimental units for random experiments. Moreover, since every pond has its own characteristics, ponds' effects cannot be ruled out, thus making it hard to define true replicates. In order to design future experiments under conditions resembling those used in production, we evaluated the possible use of floating cages as experimental units with the aim of assessing treatment effects with a reasonable statistical power. To this end, two sets of floating cages were placed respectively in two different ponds in a...
