Soccol,Carlos Ricardo; Pollom,Teresinha E. V.; Fendrich,Ricardo Cancio; Prochmann,Fernando Alberto; Mohan,Radijiskumar; Blaskowski,Marita Maciel Moreira; Melo,André Luiz de Almeida; Carvalho,Cláudio Jose Barros de; Soccol,Vanete Thomaz.
Aedes aegypti is the vector of Dengue disease, responsible for 20,000 deaths/year worldwide. Bacillus thuringiensis var israelensis - Bti releases selective and effective toxins (crystal proteins) against A. aegypti larvae. We present a low cost bioprocess for toxin production, accomplished by a selected Brazilian strain Bti (BR-LPB01) and employment of low cost substrates. Soybean meal and sugarcane molasses lead to high toxic effectiveness after 2L bioreactor fermentation (LD50=26ng/mL), near to the reference strain IPS82 (LD50=17.3 ng/mL). The pH ranged between 5.8 and 7.0 during the exponential growth period and between 7.0 and 8.4 during the stationary phase, with low activity. Thus, control of foam and pH 7.0 were started and proved to be crucial for...

Biopigments from Monascus: strains selection, citrinin production and color stability BABT
Carvalho,Júlio Cesar de; Oishi,Bruno Oliva; Pandey,Ashok; Soccol,Carlos Ricardo.
Fungi form the genus Monascus are a promising source for natural color additives. However, before effectively applying Monascus to foods, it is important to select strains which produce large amounts of biopigments but little or no citrinin, a mycotoxin usually also produced by these fungi. Also, color stability of these pigments should be properly investigated. In order to compare Monascus strains for biopigment production in solid substrate fermentation (SSF), 4 strains (NRRL 1991, NRRL 2897, CCT 3802 and LPB 31) were cultivated over PDA in Petri dishes, and compared for radial growth velocity. Also, these strains were cultivated over cooked rice, and compared in relation to their capacity to produce biopigments and citrinin. The results showed that the...

Simple models for the continuous aerobic biodegradation of phenol in a packed bed reactor BABT
Gerrard,Andrew Mark; Páca Júnior,Jan; Kostecková,Alena; Páca,Jan; Stiborová,Marie; Soccol,Carlos Ricardo.
This paper proposes the use of a preliminary, phenol removal step to reduce peak loads arriving at a conventional effluent plant. A packed bed reactor (PBR) using polyurethane foam, porous glass and also cocoa fibres as the inert support material was used. Experiments have been carried out where the flow-rates, plus inlet and outlet phenol concentrations were measured. A simple, plug-flow model is proposed to represent the results. Zero, first order, Monod and inhibited kinetics rate equations were evaluated. It was found that the Monod model gave the best fit to the experimental data and allowed linear graphs to be plotted. The Monod saturation constant, K, is approximately 50 g m-3, and ka is around 900 s-1.

Respirometry kinetics of phenol oxidation by Comamonas testosteroni Pb50 under various conditions of nutritional stress BABT
Paca,Jan; Kosteckova,Alena; Pacova,Leona; Prell,Ales; Halecky,Martin; Paca Jr.,Jan; Stiborova,Marie; Kozliak,Evguenii; Soccol,Carlos Ricardo.
Kinetics of phenol biodegradation using suspended biomass of Comamonas testosteroni Pb50 (monoculture) was measured under conditions of nutrient abundance, limitation, and prolonged cell starvation in a fed-batch reactor, with phenol being the sole carbon and energy source. The pre-washed cells were applied for measurement of the phenol and oxygen uptake rates at varied starting phenol concentrations with the kinetic parameters calculated using the Haldane model. The results revealed that nutrient limitation significantly suppressed the maximum value of exogenous respiration rate while the endogenous respiration rate, affinity and tolerance to phenol increased. By contrast, cell starvation resulted in a drop of both the exogenous and endogenous respiration...

Hernández-Carbajal,Gerardo; Rutiaga-Quiñones,Olga Miriam; Pérez-Silva,Araceli; Saucedo-Castañeda,Gerardo; Medeiros,Adriane; Soccol,Carlos Ricardo; Soto-Cruz,Nicolás Óscar.
In this work, fifty yeast strains, isolated from the spontaneous alcoholic fermentation of Agave duranguensis to produce mezcal, were tested using the double coupling system. These yeasts were from the genera Pichia, Torulaspora, Saccharomyces, Kluyveromyces, Deckera, Hanseniaspora, and Candida. P. fermentans ITD00165 was the best isoamyl acetate producer, yielding 0.38 g/L of ester after incubation for 24 h, while K. marxianus ITD00211 produced 0.32 g/L of ester. Thus P. fermentans TD00165 could be considered as an excellent choice for use in optimization studies of the culture medium and bioreactor operating conditions to develop a process for biotechnological production of isoamyl acetate.

Recent developments in microbial oils production: a possible alternative to vegetable oils for biodiesel without competition with human food? BABT
Christophe,Gwendoline; Kumar,Vinod; Nouaille,Régis; Gaudet,Geneviève; Fontanille,Pierre; Pandey,Ashok; Soccol,Carlos Ricardo; Larroche,Christian.
Since centuries vegetable oils are consumed as human food but it also finds applications in biodiesel production which is attracting more attention. But due to being in competition with food it could not be sustainable and leads the need to search for alternative. Nowdays microbes-derived oils (single cell oils) seem to be alternatives for biodiesel production due to their similar composition to that of vegetable oils. However, the cold flow properties of the biodiesel produced from microbial oils are unacceptable and have to be modified by an efficient transesterification. Glycerol which is by product of transesterification can be valorised into some more useful products so that it can also be utilised along with biodiesel to simplify the downstream...

Thermal characterization of partially hydrolyzed cassava (Manihot esculenta) starch granules BABT
Lacerda,Luiz Gustavo; Azevedo,Jayme Augusto Menegassi; Carvalho Filho,Marco Aurélio da Silva; Demiate,Ivo Mottin; Schnitzler,Egon; Vandenberghe,Luciana Porto de Souza; Soccol,Carlos Ricardo.
Cassava starch, partially hydrolyzed by fungal á-amylase, was characterized using thermal analysis, light microscopy and X-ray diffraction. Thermal degradation was initiated at lower degradation temperatures after enzymatic treatment and the DSC (Differential scanning calorimetry) analysis showed almost similar range of gelatinization temperature, but the enthalpies of gelatinization were quite increased for the partially hydrolyzed starch granules. The results suggested that the partial degradation of the starch granules was concentrated in the amorphous regions.

Starch nutritional fractions as well as thermal properties and other analysis are essential for food and industrial application. Cassava bagasse is an important agro-industrial residue and its starch content was evaluated using two alternative methods. Thermal characterization and microscopy analyses helped to understand how hydrolysis digests starchy fraction of cassava bagasse. The melting point of cassava starch occurred at 169.2ºC. Regarding TG analyses, after moisture content, there were observed two main mass losses for all samples. Results suggest hydrolysis carried out using enzyme is less effective in order to convert total starch content in cassava bagasse. However, using sulfuric acid, fibers are affected by analyses conditions.
