Since 2008, the emergent virus OsHV 1μvar has provoked massive mortality events in Crassostrea gigas spat and juveniles in France. Since 2012, mortality driven by the pathogenic bacteria Vibrio aestuarianus has stricken market sized adults. A hypothesis to explain the sudden increase in mortality observed in France since 2012 is that selective pressure due to recurrent viral infections could have led to a higher susceptibility of adults to Vibrio infection. Whole-genome amplification: a useful approach to characterize new genes in unculturable protozoan parasites such as Bonamia exitiosa ArchiMer

Bonamia exitiosa is an intracellular parasite (Haplosporidia) that has been associated with mass mortalities in oyster populations in the Southern hemisphere. This parasite was recently detected in the Northern hemisphere including Europe. Some representatives of the Bonamia genus have not been well categorized yet due to the lack of genomic information. In the present work, we have applied Whole-Genome Amplification (WGA) technique in order to characterize the actin gene in the unculturable protozoan B. exitiosa. First description of French V. tubiashii strains pathogenic to mollusk: I. Characterization of isolates and detection during mortality events ArchiMer

Nine dominant bacterial isolates were obtained from different batches of Crassostrea gigas spat experiencing high mortality rates in a French experimental hatchery/nursery in 2007. Using phenotypic analysis combined with multilocus sequence analysis, the isolates were shown to be genetically close to the Vibrio tubiashii type strain. Development of a duplex Taqman real-time PCR assay for rapid identification of Vibrio splendidus -related and V. aestuarianus strains from bacterial cultures ArchiMer

To enable the rapid and accurate identification of Vibrio splendidus-related and V. aestuarianus strains associated with Pacific cupped oyster Crassostrea gigas mortality, we developed a duplex Taqman real-time PCR assay and evaluated its efficacy. This technique proved to be rapid, sensitive, and specific and will be particularly valuable for epidemiologic studies.

En 2009, La caractérisation du niveau de ploïdie des naissains captés en 2008 dans les deux principaux bassins de captage que sont Marennes Oléron et Arcachon a été réalisée dans le cadre du réseau biovigilance. Les échantillons des naissains sauvages issus de ce captage naturel ont été prélevés sur 3 sites dans chaque bassin et pour un total analysé de 1121 animaux. En tenant compte de notre échantillonnage, nos résultats de suivi de ploïdie concluent à l’absence d’animaux polyploïdes, triploïdes ou tétraploïdes, captés dans les deux bassins prospectés. Experimental ostreid herpesvirus 1 infection of the Pacific oyster Crassostrea gigas: Kinetics of virus DNA detection by q-PCR in seawater and in oyster samples ArchiMer

Herpes- and herpes-like viruses are known to infect a wide range of bivalve mollusc species throughout the world. Abnormal summer mortalities associated to the detection of ostreid herpesvirus 1 (OsHV-1) have been currently reported in France among larvae and spat of the Pacific cupped oyster Crassostrea gigas. In the present work, we have developed an experimental protocol of horizontal transmission based on the cohabitation between healthy and experimentally infected oysters. Through a cohabitation trial, the kinetics of OsHV-1 detection in different oyster organs and seawater samples were investigated and characterized for the first time using real time quantitative PCR.

Since 2014 in France, in Charente maritime and Vendée, farmed and stocks of blue mussels experienced strong mortality events. In this context, the « MORBLEU » research project (DPMA-Ifremer agreement) was initiated to explore potentially aggravating factors, associated or correlated with mortality of mussels. Here, the work is to study biotic factors in mussels environment, looking for describe diversity and evolution of microbial communities (water column) before, during and after mortality events using NGS technology (metabarcoding) for environmental DNA analysis. Several strains, one disease: experimental investigation of Vibrio aestuarianus infection parameters in the Pacific oyster, Crassostrea gigas ArchiMer

This study investigated oyster infection dynamics by different strains of Vibrio aestuarianus isolated before and after the apparent re-emergence of this pathogen observed in France in 2011. We conducted experiments to compare minimal infective dose, lethal dose 50 and bacterial shedding for six V. aestuarianus strains. Whatever the strain used, mortality was induced in juvenile oysters by intramuscular injection and reached 90–100% of mortality within 5 days. Massive mortality outbreaks, mostly affecting spat Crassostrea gigas, have been reported in France since 2008. Disease investigations revealed that most of the mortality events are related to the detection of the Ostreid herpesvirus 1 (OsHV-1 μVar). Oyster diseases caused by pathogenic vibrios pose a major challenge to the sustainability of oyster farming. In France since 2012 a disease affecting specifically adult oysters has been associated with the presence of Vibrio aestuarianus. Here, by combining genome comparison, phylogenetic analyses and high throughput infections of strains isolated before or during the recent outbreaks, we show that virulent strains cluster into two V. aestuarianus lineages independently of the sampling dates. The bacterial lethal dose was not different between strains isolated before or after 2012. Hence the emergence of a new highly virulent clonal strain is unlikely.
