Genetic diversity among Brazilian soybean cultivars based on SSR loci and pedigree data BABT
Priolli,Regina Helena Geribello; Pinheiro,José Baldin; Zucchi,Maria Imaculada; Bajay,Miklos Maximiliano; Vello,Natal Antonio.
In this study, simple sequence repeats (SSR) loci and pedigree data were used to investigate the genetic relationship in a group of 168 Brazilian soybean cultivars. Eighteen SSR loci produced an average of 5.06 alleles and a mean gene diversity of 0.58 for the cultivars studied. Genetic distance (GD) was determined using the modified Roger's Wright distance, and a final dendrogram was in agreement with the cultivar pedigree. A distance matrix based on the coefficient of parentage scores was also generated for the cultivars, which ranged from 0 to 1, with a mean of 0.18, whereas SSR-based genetic similarity (1- GD) ranged from 0.01 to 0.90, with a mean of 0.25. Mantel's Z test showed that the similarity matrices generated from both the data sets were low,...

Genetic structure and gene flow in Eugenia dysenterica DC in the Brazilian Cerrado utilizing SSR markers Genet. Mol. Biol.
Zucchi,Maria Imaculada; Brondani,Rosana Pereira Vianello; Pinheiro,José Baldin; Chaves,Lázaro José; Coelho,Alexandre Siqueira Guedes; Vencovsky,Roland.
The ''cagaita tree'' (Eugenia dysenterica) is a plant found widespread in the Brazilian Cerrado. Its fruit is used for popular consumption and for industrial purposes. This study opens a new perspective for the generation of population genetic data and parameters estimates for devising sound collection and conservation procedures for Eugenia dysenterica. A battery of 356 primer pairs developed for Eucalyptus spp. was tested on the ''cagaita tree''. Only 10 primer pairs were found to be transferable between the two species. Using a polyacrilamide gel, an average of 10.4 alleles per locus was detected, in a sample of 116 individuals from 10 natural ''cagaita tree'' populations. Seven polymorphic loci allowed estimation of genetic parameters, including...

Origin, evolution and genome distribution of microsatellites Genet. Mol. Biol.
Oliveira,Eder Jorge; Pádua,Juliano Gomes; Zucchi,Maria Imaculada; Vencovsky,Roland; Vieira,Maria Lúcia Carneiro.
Microsatellites, or simple sequence repeats (SSRs), have been the most widely applied class of molecular markers used in genetic studies, with applications in many fields of genetics including genetic conservation, population genetics, molecular breeding, and paternity testing. This range of applications is due to the fact that microsatellite markers are co-dominant and multi-allelic, are highly reproducible, have high-resolution and are based on the polymerase chain reaction (PCR). When first introduced, the development of microsatellite markers was expensive but now new and efficient methods of repetitive sequence isolation have been reported, which have led to reduced costs and microsatellite-technology has been increasingly applied to several species,...

A wide array of molecular markers has been used to investigate the genetic diversity among common bean species. However, the best combination of markers for studying such diversity among common bean cultivars has yet to be determined. Few reports have examined the genetic diversity of the carioca bean, commercially one of the most important common beans in Brazil. In this study, we examined the usefulness of two molecular marker systems (simple sequence repeats - SSRs and amplified fragment length polymorphisms - AFLPs) for assessing the genetic diversity of carioca beans. The amount of information provided by Roger's modified genetic distance was used to analyze SSR data and Jaccards similarity coefficient was used for AFLP data. Seventy SSRs were...

The genetic structure and mating system of Acrocomia aculeata (Arecaceae) Genet. Mol. Biol.
Abreu,Aluana Gonçalves; Priolli,Regina Helena Geribello; Azevedo-Filho,Joaquim Adelino; Nucci,Stella Maris; Zucchi,Maria Imaculada; Coelho,Ricardo Marques; Colombo,Carlos Augusto.
Acrocomia aculeata is a perennial, fruit-producing palm tree, native to tropical forests. Its fruits have spurred interest because of their significant potential for use in the cosmetic industry and as feedstock for biofuel. In the present study, the genetic structure and mating system in Acrocomia aculeata were analyzed, using eight nuclear microsatellite loci and samples from São Paulo and Minas Gerais states, Brazil. By means of Bayesian analysis, these populations were clustered into two or three groups. A high multilocus outcrossing rate suggests that outcrosses were predominant, although a certain degree of biparental inbreeding also occurred. Thus, although monoecious and self-compatible, there is every indication that A. aculeata bears a mixed...

Genetic divergence of rubber tree estimated by multivariate techniques and microsatellite markers Genet. Mol. Biol.
Gouvêa,Lígia Regina Lima; Rubiano,Luciana Benchimol; Chioratto,Alisson Fernando; Zucchi,Maria Imaculada; Gonçalves,Paulo de Souza.
Genetic diversity of 60 Hevea genotypes, consisting of Asiatic, Amazonian, African and IAC clones, and pertaining to the genetic breeding program of the Agronomic Institute (IAC), Brazil, was estimated. Analyses were based on phenotypic multivariate parameters and microsatellites. Five agronomic descriptors were employed in multivariate procedures, such as Standard Euclidian Distance, Tocher clustering and principal component analysis. Genetic variability among the genotypes was estimated with 68 selected polymorphic SSRs, by way of Modified Rogers Genetic Distance and UPGMA clustering. Structure software in a Bayesian approach was used in discriminating among groups. Genetic diversity was estimated through Nei's statistics. The genotypes were clustered...

Caffeine inheritance in interspecific hybrids of Coffea arabica x Coffea canephora (Gentianales, Rubiaceae) Genet. Mol. Biol.
Priolli,Regina H.G.; Mazzafera,Paulo; Siqueira,Walter J.; Möller,Milene; Zucchi,Maria Imaculada; Ramos,Luis Carlos S.; Gallo,Paulo B.; Colombo,Carlos A..
Caffeine inheritance was investigated in F2 and BC1F1 generations between Coffea arabica var. Bourbon Vermelho (BV) and Coffea canephora var. Robusta 4x (R4x). The caffeine content of seeds and leaves was determined during 2004 and 2005. Microsatellite loci-markers were used to deduce the meiotic pattern of chromosome pairing of tetraploid interspecific hybrids. Genetic analysis indicated that caffeine content in seeds was quantitatively inherited and controlled by genes with additive effects. The estimates of broad-sense heritability of caffeine content in seeds were high for both generations. In coffee leaves, the caffeine content (BSH) from the same populations showed transgressive segregants with enhanced levels and high BSH. Segregation of...

Because of the continuous introduction of germplasm from abroad, some collections have a high number of accessions, making it difficult to explore the genetic variability present in a germplasm bank for conservation and breeding purposes. Therefore, the aim of this study was to quantify and analyze the structure of genetic variability among 500 common bean accessions to construct a core collection. A total of 58 SSRs were used for this purpose. The polymorphism information content (PIC) in the 180 common bean accessions selected to compose the core collection ranged from 0.17 to 0.86, and the discriminatory power (DP) ranged from 0.21 to 0.90. The 500 accessions were clustered into 15 distinct groups and the 180 accessions into four distinct groups in the...

Karasawa,Marines M.G.; Vencovsky,Roland; Silva,Cynthia M.; Zucchi,Maria Imaculada; Oliveira,Giancarlo C.X.; Veasey,Elizabeth A..
Knowledge of the genetic structure and diversity of natural populations is important in developing strategies for in situ and ex situ conservation. We used eight microsatellite loci to estimate genetic structure and investigate within and between population genetic variation in eleven Brazilian wild rice (Oryza glumaepatula) populations. The study showed the following genetic diversity parameters: average number of 3.1 alleles per locus; 77.3% polymorphic loci; 0.091 observed heterozygosity and 0.393 gene diversity. F-statistics detected by microsatellite loci were: F ST = 0.491 (and R ST = 0.608), F IS = 0.780 and F IT = 0.888. No population was in Hardy-Weinberg equilibrium. The estimated apparent outcrossing rate (0.143) indicated a predominance of...

High genetic diversity among and within bitter manioc varieties cultivated in different soil types in Central Amazonia Genet. Mol. Biol.
Alves-Pereira,Alessandro; Peroni,Nivaldo; Cavallari,Marcelo Mattos; Lemes,Maristerra R.; Zucchi,Maria Imaculada; Clement,Charles R..
Abstract Although manioc is well adapted to nutrient-poor Oxisols of Amazonia, ethnobotanical observations show that bitter manioc is also frequently cultivated in the highly fertile soils of the floodplains and Amazonian dark earths (ADE) along the middle Madeira River. Because different sets of varieties are grown in each soil type, and there are agronomic similarities between ADE and floodplain varieties, it was hypothesized that varieties grown in ADE and floodplain were more closely related to each other than either is to varieties grown in Oxisols. We tested this hypothesis evaluating the intra-varietal genetic diversity and the genetic relationships among manioc varieties commonly cultivated in Oxisols, ADE and floodplain soils. Genetic results did...

Genetic instability of sugarcane plants derived from meristem cultures Genet. Mol. Biol.
Zucchi,Maria Imaculada; Arizono,Hideto; Morais,Vicente Alberto; Fungaro,Maria Helena Pelegrinelli; Vieira,Maria Lúcia Carneiro.
The RADP (Random amplified polymorphic DNA) technique was used to detect tissue-culture-induced variations in sugarcane. Plants of the Brazilian variety RB83-5486 propagated via rhizomes and via meristem cultures were studied. The polymorphism rate for 98 RAPD loci was 6.93% when the plants derived from meristems. Besides, in order to evaluate the influence of the number of subcultures on the generation of somaclonal variation, field-grown RB83-5486 plants derived from 10 meristems were studied after five subcultivations. Although different rates of polymorphism were observed, there was no direct association with the stage of subcultivation. The analysis of plants of two sugarcane varieties cultivated in vitro from meristems showed that variety RB83-5486...

Similaridade genética entre raças bovinas brasileiras PAB
Rangel,Priscila Nascimento; Zucchi,Maria Imaculada; Ferreira,Márcio Elias.

Genetic structure and gene flow of Eugenia dysenterica natural populations PAB
Zucchi,Maria Imaculada; Pinheiro,José Baldin; Chaves,Lázaro José; Coelho,Alexandre Siqueira Guedes; Couto,Mansuêmia Alves; Morais,Lizz Kezzy de; Vencovsky,Roland.
This study was carried out to assess the genetic variability of ten "cagaita" tree (Eugenia dysenterica) populations in Southeastern Goiás. Fifty-four randomly amplified polymorphic DNA (RAPD) loci were used to characterize the population genetic variability, using the analysis of molecular variance (AMOVA). A phiST value of 0.2703 was obtained, showing that 27.03% and 72.97% of the genetic variability is present among and within populations, respectively. The Pearson correlation coefficient (r) among the genetic distances matrix (1 - Jaccard similarity index) and the geographic distances were estimated, and a strong positive correlation was detected. Results suggest that these populations are differentiating through a stochastic process, with restricted...

ABSTRACT Some germplasm collections have a high number of accessions, which makes it difficult to explore the genetic variability present in the germplasm bank due to the redundancy and the difficulty of detailed analysis of all conserved accessions. Therefore, our study aimed to analyze the genetic diversity of 153 lima bean (Phaseolus lunatus) accessions for the purpose of constructing a core collection. Eleven SSRs were used for this purpose. The 153 lima bean accessions can be represented by low redundancy using a minimum of 34 accessions, thus representing 22 % of the size of the entire germplasm bank. The core collection had a higher Shannon diversity index and expected heterozygosity (1.906 and 0.811, respectively) than those presented by the entire...
