Qualidade da castanha-do-brasil do comércio de Rio Branco, Acre 36

Este trabalho teve como objetivo avaliar a qualidade de castanhas-do-brasil beneficiadas e comercializadas em Rio Branco, Acre. Foram analisadas amostras das três marcas de castanha encontradas no mercado local quanto às variáveis: atividade de água, teor de umidade, contagem total de fungos filamentosos, quantificação de Aspergillus flavus e de A. parasiticus, bem como quantificação de aflatoxinas B1, B2, G1 e G2. As castanhas do comércio se encontravam com um teor de umidade e atividade de água adequados, o que pode ter sido responsável pela baixa contaminação por fungos e por aflatoxinas. Quanto a estas micotoxinas, as amostras estão de acordo com o recomendado pela Anvisa, podendo ser esta uma consequência da grande divulgação no Estado do uso de Boas...

Ação fungitóxica do óleo essencial de Tanaecium nocturnum (Barb. Rodr.) Bur. e K. Shum sobre o Aspergillus flavus isolado da castanha-do-Brasil (Bertholletia excelsa) 36

Neste trabalho, avaliou-se a capacidade fungitóxica do óleo essencial de folhas frescas de Tanaecium nocturnum sobre o Aspergillus flavus isolado da castanha-do-brasil, por meio das técnicas de contato e fumigação. Pelos resultados dos bioensaios realizados até 10 dias de incubação, verificou-se que a inibição total do crescimento micelial ocorreu quando se utilizou o óleo essencial nas concentrações de 782 ppm (técnica de contato) e 1000 ppm (técnica de fumigação). Em ambas as técnicas, o óleo essencial inibiu a esporulação a partir da concentração de 500 ppm. Biotransformation of sucrose into 5-hydroxy-2-hydroxymethyl-γ-pirone by Aspergillus flavus 42

The sucrose hydrolysis and the preference of consumption of glucose instead of fructose were investigated for the production of 5-hydroxy-2-hydroxymethyl-γ-pyrone (HHMP) in the presence of Aspergillus flavus IOC 3974 cultivated in liquid Czapeck medium. Standardized 0.5g of pellets were transferred as inoculum into twelve conical flasks of 250 ml containing 100 ml of medium with different sucrose concentration, which was kept at 120 rpm and 28"C for 16 days without pH adjustment. Aliquots of 500μl of the broth culture were withdrawn at 24 h intervals and analyzed. The major yield of HHMP was 26g l-1 in 120g l-1 of sucrose. Mycelial growth and amylase production by a mycotoxigenic strain of Fusarium moniliforme and Aspergillus flavus were evaluated in a culture medium containing starch, glycerol, wheat bran or corn. With emphasis on corn, different fractions composed by germ, degermed seed, starch, milky stage corn and the respective starch or supernatant fraction were analyzed for F. moniliforme growth . The medium composed of milky stage corn supernatant promoted the best mycelial growth (p<0.05), and it was used to prepare amylase production medium in the next step. ABSTRACT The in vitro susceptibility of 105 clinical and environmental strains of Aspergillus fumigatus and Aspergillus flavus to antifungal drugs, such as amphotericin B, azoles, and echinocandins was evaluated by the broth microdilution method proposed by the European Committee on Antimicrobial Susceptibility Testing (EUCAST). Following the EUCAST-proposed breakpoints, 20% and 25% of the clinical and environmental isolates of A. fumigatus, respectively, were found to be resistant to itraconazole (Minimal Inhibitory Concentration, MIC > 2.0 mg/L). Biflavonoids inhibit the production of aflatoxin by Aspergillus flavus 56

The biflavonoids 6,6"-bigenkwanin, amenthoflavone, 7,7"-dimethoxyagastisflavone and tetradimethoxybigenkwanin isolated from Ouratea species were tested for inhibitory activity on Aspergillus flavus cultures. Suspensions of Aspergillus flavus spores were inoculated into 50 ml of YES medium at different biflavonoid concentrations: 5 and 10 µg/ml for 6,6"-bigenkwanin, amenthoflavone and 7,7"-dimethoxyagastisflavone, and 5, 10, 15 and 20 µg/ml for tetradimethoxybigenkwanin. The four biflavonoids showed inhibitory activity on aflatoxin B1 and B2 production (P<0.001), but did not inhibit fungal growth at the concentration tested (P>0.05). These results show that biflavonoids can be used for the development of agents to control aflatoxin production.

Nutritional changes in powdered red pepper upon in vitro infection of Aspergillus flavus 58

Quantitative losses in various biochemical constituents like capsaicin, carotenes, ascorbic acid, polyphenols, mineral matter, sugars (soluble and insoluble), protein and fat were estimated after the successful growth of Aspergillus flavus for 30 days on powdered red pepper. The fungal biomass was measured by ergosterol content and Aflatoxin B1 by HPLC. Amongst the various nutritional constituents evaluated for nutritional losses and changes the highest nutritional loss was reported in total carotenoids (88.55%) followed by total sugars (85.5%). Copper induction and differential expression of laccase in Aspergillus flavus 58

Aspergillus flavus was isolated from soil and exhibited laccase activity under both constitutive and copper induced conditions. Spiking the medium with 1 mM copper sulfate resulted in an increase in the activity which reached 51.84 U/mL, a distinctive protein band was detected at 60 kDa. The extracellular enzyme was purified 81 fold using gel filtration chromatography and resulted in two different laccase fractions L1 and L2, the latter had a higher enzymatic activity which reached 79.57 U/mL and specific activity of 64.17 U/μg protein. The analysis of the spectrum of the L2 fraction showed a shoulder at 330 nm which is characteristic for T2/T3 copper centers; both copper and zinc were detected suggesting that this is an unconventional white laccase....

Production of aflatoxins by Aspergillus flavus and of fumonisins by Fusarium species isolated from Brazilian sorghum 58

Fifty-nine Aspergillus flavus and 35 Fusarium verticillioides strains, isolated from freshly harvested (10) and stored (130) Brazilian sorghum samples, were tested regarding their ability to produce aflatoxins (coconut milk agar) and fumonisins (rice culture), respectively. Aflatoxins B1 and B2 were detected by TLC, and fumonisins B1 and B2 were analyzed by HPLC. Thirty-eight (64.4%) A. flavus strains produced detectable levels of aflatoxins at concentrations ranging from 12.00 to 3282.50 µg/kg (AFB1 + AFB2), while thirty two (91%) F. verticillioides strains produced FB1 at concentrations ranging from 0.12 to 5.38 µg/g. The effect of inoculation of Aspergillus flavus, Fusarium verticillioides, and Penicillium sp. in Dystrophic Red Latosol (DRL) and Eutroferric Red Latosol (ERL) soils with or without glucose on the total carbohydrate content and the dehydrogenase and amylase activities was studied. The fungal growth and spore production in culture medium with and without glucose were also evaluated. A completely randomized design with factorial arrangement was used. Mycoflora and aflatoxin/fumonisin production by fungal isolates from freshly harvested corn hybrids 58

The mycoflora of 3 hybrids of freshly harvested corn grains collected from three regions of the state of São Paulo, Brazil (Assis, Capão Bonito and Ribeirão Preto) was investigated. A total of 66 samples were analyzed focusing on the influence of abiotic factors (moisture content, water activity, temperature and rainfall) on both the prevalence of Aspergillus flavus and Fusarium moniliforme, and the ability of these genera isolates to produce aflatoxins and fumonisins, respectively. Time course of virus-like particles (VLPs) double-stranded rna accumulation in toxigenic and non-toxigenic strains of Aspergillus flavus 58

Two strains of Aspergillus flavus, non-toxigenic NRRL 6550 and toxigenic NRRL 5940, were studied over a period of 44 days, in order to detect the presence of virus-like particles (VLPs) by means of electron microscopy (EM) and nucleic acids electrophoresis. Only the toxigenic strain contained VLPs, presenting three-segmented dsRNA. An increase in VLPs number was observed during the exponential phase of fungal growth, up to day 12 of culture; after this, higher levels of aflatoxin production in toxigenic NRRL 5940 mycelia occurred in parallel with decreased VLPs replication.

Activity of the aqueous extract from Polymnia sonchifolia leaves on growth and production of aflatoxin B1 by Aspergillus flavus 58

The aqueous extract from Polymnia sonchifolia leaves (AE) was tested for inhibitory activity on aflatoxin B1(AFB1) production and growth of Aspergillus flavus. The cytotoxicity of AE on Vero cells was also performed. Suspensions of A. flavus spores were inoculated into 50 mL of YES medium together with different concentrations of the AE. The aflatoxin B1 was extracted, analyzed by thin layer chromatography and quantified by photodensitometry. All the concentrations of AE induced inhibition of AFB1 production. The aqueous extract showed in vitro cytotoxicity to Vero cells only at concentrations above 500 µg/mL.

As cascas de amendoim (Arachis hypogaea L.) são de grande importância para confecção de cama de frangos, de gado de leite e como fonte de fibras para ruminantes, portanto a elucidação dos mecanismos de contaminação por fungos toxigênicos e por micotoxinas em amendoim é imprescindível, especialmente para que medidas preventivas possam ser tomadas. Abstract Aflatoxins are mutagenic, carcinogenic, and teratogenic mycotoxins. The objective of this work was to study the presence of aflatoxigenic Aspergillus in commercial Bulgur wheat in the city of Maringá, Paraná, Brazil. Thirty samples of commercial Bulgur wheat, acquired in the period of August 2011 to January 2012, were evaluated. The enumeration analysis showed that samples had up to 273.3 CFU of molds and 133.3 CFU of aflatoxigenic Aspergillus per gram of wheat. Toxigenic potential of Aspergillus flavus tested in different culture conditions 116

The objective of the present work was to evaluate the capacity of three isolates of Aspergillus flavus to produce aflatoxin under different culture conditions. This experiment was based on a 2³ factorial design, in which the independent variables were temperature (20-40 °C), incubation time (7-21 days), and the pH (2.0-6.0) in two different synthetic media. The optimal conditions were applied to non-aflatoxigenic isolates previously tested in coconut agar. Aflatoxin B1 was extracted directly from the synthetic cultures with chloroform. Thin Layer Chromatography (TLC) and Photographic Photometry were utilized to identify and quantify the compounds.
