Influence of growth media and temperature on bacterial adhesion to polystyrene surfaces 52
Zeraik,Ana Eliza; Nitschke,Marcia.
Bacterial adhesion to inert surfaces is a complex process influenced by environmental conditions. In this work, the influence of growth medium and temperature on the adhesion of Pseudomonas aeruginosa, Serratia marcescens, Staphylococcus aureus, Micrococcus luteus and Listeria monocytogenes to polystyrene surfaces was studied. Most bacteria demonstrated the highest adhesion when cultured in TSYEA, except S. marcescens, which showed to be positively influenced by the pigment production, favored in poor nutrient media (lactose and peptone agar). P. aeruginosa adhesion to polystyrene increased at low temperatures whatever the medium used. The culture medium influenced the surface properties of the bacteria as assessed by the MATS test.

Pseudomonas Aeruginosa: Virulence Factors and Antibiotic Resistance Genes 52
Rocha,Antônio José; Barsottini,Mario Ramos de Oliveira; Rocha,Renan Rhonalty; Laurindo,Maria Vitória; Moraes,Francisco Leandro Laurindo de; Rocha,Soraya Lília da.
Abstract In this review, we explore some aspects of Pseudomonas aeruginosa virulence factors that are related to disease development in healthy organisms and resistance to antibiotics. This pathogen is one of the most clinically and epidemiologically important bacteria in Brazil, being the major cause of opportunistic infections. Among the virulence factors, biofilm formation acting of manner different in the organism. Furthermore, we review several P. aeruginosa genes that act in antimicrobial resistance, such as β-lactamases against β-lactamers. The resistance to pied-lactamases in P. aeruginosa is associated to resistance to the broad-spectrum cephalosporin. On the other hand, there is a group of synthetic broad-spectrum antibiotics acting on DNA...

Biofilms: microbes and disease 55
Aparna,Madhu Sharma; Yadav,Sarita.
Bacteria that attach to surface aggregate in a hydrated polymeric matrix of their own synthesis to form biofilms. These represent microbial societies with their own defense and communication system. Transitioning from acute to chronic infection is frequently associated with biofilm formation.Bacteria in biofilms are innately more resistant to antimicrobial agents. The presence of indwelling medical devices increases the risk for biofilm formation and subsequent infection. The current antibiotic therapies are of limited effectiveness in resolving biofilms infection. This review attempts to discuss the stages in biofilm formation, their pathogenic mechanisms, effect of antimicrobial agents, detection and eradication of the biofilms.

Architecture and relevance of several strongly adhered biofilms over a polyester imide (PEI) surface 58
Soncini-Júnior,Giancarlo; Franchetti,Sandra Mara Martins; Marconato,José Carlos.
A microscopical study was undertaken to analyze the eventual fungi adhesion over a polyester imide surface in enameled copper wires. Scanning electron microscopy allowed to observe in these adhered biofilms, a high amount of pigments, hyphae and its enzymatic arsenal probably acting in the surface of the polymer. Due to the highly aromatic nature of this polyester and the residual phenol derivates still present in the reticulated polymer, an antifungal activity could be expected, but no significant changes were noted in fungal growing and in the adhesion process. Additionally, the wires showed completely failure in all insulation properties. The studies aimed to understand and evaluate the great fungi potential that could be explored in biodeterioration...

Newly-synthesized chalcones-inhibition of adherence and biofilm formation of methicillin-resistant Staphylococcus aureus 58
Bozic,Dragana D.; Milenkovic,Marina; Ivkovic,Branka; Cirkovic,Ivana.
Biofilm formation and adherence of bacteria to host tissue are one of the most important virulence factors of methicillin-resistant strains of Staphylococcus aureus (MRSA). The number of resistant strains is seriously increasing during the past years and bacteria have become resistant, not only to methicillin, but also to other commonly used antistaphylococcal antibiotics. There is a great need for discovering a novel antimicrobial agent for the treatment of staphylococcal infections. One of the most promising groups of compounds appears to be chalcones. In present study we evaluated the in vitro effect of three newly synthesized chalcones: 1,3-Bis-(2-hydroxy-phenyl)-propenone, 3-(3Hydroxy-phenyl)-1-(2-hydroxy-phenyl)-propenone and...

Expression of virulence genes by Listeria monocytogenes J0161 in natural environment 58
Tirumalai,Prem Saran; Prakash,Soam.
Majority of studies concerning the gene expression of Listeria monocytogenes have been done on pure culture states. Our objective was to study L.monocytogenes in a co-cultured state and to understand if microbes in their natural state of existence are different in their expression than that of the purely cultured lab grown forms. For a long period discussions have been on the expression of prfA, (which is a virulence gene regulator) in a mammalian host and its role in causing the switch from a saprophytic to pathogenic form of L.monocytogenes. We, in this paper for the first time report the expression of prfA and other virulence genes by L.monocytogenes under different extracellular conditions, and also as a pure culture biofilms, that is different from...

Effect of alcohols on filamentation, growth, viability and biofilm development in Candida albicans 58
Chauhan,Nitin M; Shinde,Ravikumar B; Karuppayil,S. Mohan.
In this study we report the potential of alcohols as morphogenetic regulators in Candida albicans. All the alcohols tested influenced various modes of growth like planktonic as well as biofilm forms. Viability was affected at high concentrations. Among the alcohols, the response of C. albicans to amyl alcohol (pentanol) was noteworthy. Amyl alcohol at a concentration 0.5% which was not inhibitory to growth and viability specifically inhibited morphogenetic switching from yeast to hyphal forms. It also inhibited normal biofilm development favoring yeast dominated biofilms. Based on this study we hypothesize that alcohols produced under anaerobic conditions may not favor biofilm development and support dissemination of yeast cells. Since anaerobic conditions...

Growth, viability and architecture of biofilms of Listeria monocytogenes formed on abiotic surfaces 58
Reis-Teixeira,Fernanda Barbosa dos; Alves,Virgínia Farias; Martinis,Elaine Cristina Pereira de.
Abstract The pathogenic bacterium Listeria monocytogenes can persist in food processing plants for many years, even when appropriate hygienic measures are in place, with potential for contaminating ready-to-eat products and, its ability to form biofilms on abiotic surfaces certainly contributes for the environmental persistence. In this research, L. monocytogenes was grown in biofilms up 8 days attached to stainless steel and glass surfaces, contributing for advancing the knowledge on architecture of mature biofilms, since many literature studies carried out on this topic considered only early stages of cell adhesion. In this study, biofilm populations of two strains of L. monocytogenes (serotypes 1/2a and 4b) on stainless steel coupons and glass were...

Melo,Poliana de Castro; Ferreira,Luciano Menezes; Nader Filho,Antônio; Zafalon,Luiz Francisco; Vicente,Hinig Isa Godoy; Souza,Viviane de.
Biofilm formation is considered to be a selective advantage for Staphylococcus aureus mastitis isolates by facilitating bacterial persistence in the udder. It requires attachment to mammary epithelium, proliferation and accumulation of cells in multilayers. The objective of this study was to determine the sensitivity and specificity of three techniques for the detection of S. aureus biofilm-positive strains. Two phenotypic tests, including growth on microtitre plates and Congo red agar, were compared with a PCR technique using 94 S. aureus strains obtained from cows with subclinical mastitis from two farms in the state of São Paulo. These strains were characterised by in vitro slime production on Congo red agar, biofilm formation on microtitre plates and...

Effect of modified clays on the structure and functional properties of biofilms produced with zein 116
Tavares,Luciana Lopes; Almeida,Crislene Barbosa de; Caruso,Ícaro Putinhon; Cornélio,Marinônio Lopes; Lopes Filho,José Francisco.
The purpose of this study was to evaluate changes in the structure and some functional properties of biofilms added with modified clays (Cloisite® 15A and Cloisite® 30B) prepared by the casting method. The analysis of the microstructure of the films, scanning electron microscopy (SEM), Optical microscopy (MO), and Infrared Spectroscopy (FTIR) indicated that the addition of clay in the films resulted in the formation of a heterogeneous microstructure, microcomposite or tactoid. Due to the formation of a microcomposite structure, functional properties of the films added with both clays such as opacity, solubility, and permeability to water vapor (PVA), were not better than those of the control film. Thus, it was concluded that although it is possible to...
