ABSTRACT Brazil nut is a very important nontimber forest product in the Amazon region. Propagation of this tree still represents a challenge due to slow and uneven seed germination. In this context, plant growth-promoting bacteria can facilitate the process of propagation. The aims of this study were to isolate and characterize endophytic bacteria from the roots of Brazil nut trees in native terra firme forest and cultivation areas in northern Brazil, and to identify mechanisms by which bacteria act in plant growth promotion. Overall, 90 bacterial isolates were obtained from the roots of Brazil nut trees in monoculture, agroforestry and native forest areas by using different semisolid media. The isolates were characterized by sequencing the 16S rRNA gene.... |

ABSTRACT To maximize the contribution of biological nitrogen fixation in common bean, it is necessary to use bacterial strains that are more adapted, competitive, and efficient in the symbiotic process. In this regard, the aim of this study was to evaluate the agronomic efficiency (AE) of three bacterial strains isolated from acid soils with high Al content from the Amazon region in an Argissolo Vermelho Distrófico típico soil (Typic Rhodustults - USDA Classification) from the municipality of Formiga, MG, Brazil. We compared their AE to that of the reference strain CIAT 899T and of two controls without inoculation (one without and another with 80 kg ha-1 of N-urea). The results indicated that inoculation with the strains UFLA 02-100 and UFLA 02-127... |

ABSTRACT Maize plants can establish beneficial associations with plant growth-promoting bacteria. However, few studies have been conducted on the characterization and inoculation of these bacteria in the Amazon region. This study aimed to characterize endophytic bacteria isolated from maize in the Amazon region and to assess their capacity to promote plant growth. Fifty-five bacterial isolates were obtained from maize grown in two types of ecosystems, i.e., a cerrado (savanna) and a forest area. The isolates were characterized by the presence of the nifH gene, their ability to synthesize indole-3-acetic acid (IAA) and solubilize calcium phosphate (CaHPO4), and 16S rRNA partial gene sequencing. Twenty-four bacteria contained the nifH gene, of which seven... |

The aim of this study was to evaluate the effects of foliar application of molybdenum and small dose of nitrogen at planting on common bean production when using seed inoculation with rhizobia. Two field experiments were conducted, applying a randomized block design and a factorial arrangement (3x2x2) + 1, with three replicates. Treatments consisted of inoculation with Rhizobium spp. (strain CIAT 899T, strain UFLA 02–100, and uninoculated), foliar application of molybdenum (0 and 80 g ha-1), and fertilization with N-urea at planting (0 and 20 kg ha-1), with an additional N treatment of 40 applied at planting and 40 kg ha-1 applied as topdressing. The foliar application of molybdenum did not favor the nodulation, but its effect on yield was dependent on the... |

ABSTRACT Although the use of inoculants containing rhizobia is encouraged, there are no official recommendations for inoculation of snap bean. In this respect, the aim of this study was to evaluate the agronomic performance of Rhizobium strains in symbiosis with snap bean cultivars with different growth habits and crop cycles. The experiment was carried out in pots with soil in a greenhouse in the spring-summer season in Lavras, state of Minas Gerais, Brazil. A completely randomized experimental design was used, with 4 replications, in a 4 × 7 factorial arrangement involving four snap bean cultivars and seven treatments – five strains of Rhizobium (UFLA 02-100, UFLA 02-127, UFLA 04-173, CIAT 899, and PRF 81), plus two controls without inoculation (with and... |

ABSTRACT Cowpea is a very important crop to Brazilian Semi-Arid mainly small family-based farmers. Rhizobia inoculation is a practice, easy to use, and cheap technology that increases cowpea productivity. The aim of this study was to evaluate the efficiency of two new rhizobia isolates in greenhouse and field as well as classify them taxonomically. To bacterial identification the 16S rRNA gene of ESA 17 and ESA 18 isolates were sequenced. The greenhouse test was conducted with pots containing 3 L of soil and the bacterial isolates evaluated were ESA 17, ESA 18, BR 3267 or BR 3262 strains. A field experiment was implemented on a Vertisol in Juazeiro, Bahia State, to evaluate the cowpea growth and productivity. In this experiment, the peat-based inoculants... |

ABSTRACT Studies in the Amazon indicate a wide diversity of rhizobia with the ability for biological nitrogen fixation (BNF), which could expand the number of strains approved for cowpea. Thus, the aim of this field study was to evaluate the agronomic performance in cowpea of the several strains isolated from the soils of the Brazilian states Acre and Rondônia, and to compare them withstrains approved by the Ministry of Agriculture (MAPA) and withnon-inoculated controls (without and with mineral nitrogen fertilizer). The inoculants performed well. Though less effective than the other strains, the UFLA 03-36 strain also was prominent with respect to grain yield. Because of the positive response of the UFLA 03-129 strain, which led to yield increases greater... |

Estimates of the rates of heterotrophic nitrogen fixation (HNF) and denitrification by acetylene reduction and acetylene inhibition, respectively, were determined for the roots and culms of wild rice, Oryza glumaepatula, in Lake Batata, a lake severely impacted by the dumping of bauxite tailings into the lake basin. Two stations were established in different parts of the lake: one in a natural area and other in an impacted area of the lake. Plant assays were performed during the principle hydroperiods of dry (December) and flood (July) to determine both: a) Actual rates of HNF and denitrification, and b) potential rates of HNF after amending with glucose potential rates of denitrification after amending with glucose, glucose + nitrate, or nitrate alone.... |

Azospirillum brasilense is a nitrogen-fixing bacterium associated with important agricultural crops such as rice, wheat and maize. The expression of genes responsible for nitrogen fixation (nif genes) in this bacterium is dependent on the transcriptional activator NifA. This protein contains three structural domains: the N-terminal domain is responsible for the negative control by fixed nitrogen; the central domain interacts with the RNA polymerase σ54 co-factor and the C-terminal domain is involved in DNA binding. The central and C-terminal domains are linked by the interdomain linker (IDL). A conserved four-cysteine motif encompassing the end of the central domain and the IDL is probably involved in the oxygen-sensitivity of NifA. In the present study,... |

NifA is the transcriptional activator of the nif genes in Proteobacteria. It is usually regulated by nitrogen and oxygen, allowing biological nitrogen fixation to occur under appropriate conditions. NifA proteins have a typical three-domain structure, including a regulatory N-terminal GAF domain, which is involved in control by fixed nitrogen and not strictly required for activity, a catalytic AAA+ central domain, which catalyzes open complex formation, and a C-terminal domain involved in DNA-binding. In Herbaspirillum seropedicae, a β-proteobacterium capable of colonizing Graminae of agricultural importance, NifA regulation by ammonium involves its N-terminal GAF domain and the signal transduction protein GlnK. When the GAF domain is removed, the protein... |

Abstract The strain BR 3262 was isolated from nodule of cowpea (Vigna unguiculata L. Walp) growing in soil of the Atlantic Forest area in Brazil and it is reported as an efficient nitrogen fixing bacterium associated to cowpea. Firstly, this strain was assigned as Bradyrhizobium elkanii, however, recently a more detailed genetic and molecular characterization has indicated it could be a Bradyrhizobium pachyrhizi species. We report here the draft genome sequence of B. pachyrhizi strain BR 3262, an elite bacterium used as inoculant for cowpea. The whole genome with 116 scaffolds, 8,965,178 bp and 63.8% of C+G content for BR 3262 was obtained using Illumina MiSeq sequencing technology. Annotation was added by the RAST prokaryotic genome annotation service and... |

This study evaluated 26 pigeonpea rhizobial isolates according to their cultural characteristics, intrinsic antibiotic resistance, salt and temperature tolerance, carbon source utilization and amylolytic activity. The cultural characterization showed that the majority of them presented the ability to acidify the YMA. Among the 27 isolates evaluated, 25 were able to grow when incubated at 42° C and 11 showed tolerance to 3% (w/v) of NaCl in YMA medium. The patterns of carbon sources utilization was very diverse among the isolates. It was observed the capacity of three strains to metabolize all the carbon sources evaluated and a total of 42% of the bacterial isolates was able to grow in the culture medium supplemented with at least, six carbon sources. The... |
