O presente trabalho teve como objetivo avaliar os teores de trigonelina, ácidos clorogênicos (5-ACQ) e cafeína em grãos de diferentes cultivares de cafeeiro (Coffea arabica L.). As cultivares avaliadas foram coletadas na Fazenda Experimental da Epamig em Lavras, Estado de Minas Gerais, no ano agrícola de 2005/2006. O delineamento experimental utilizado foi um inteiramente casualizado constando de oito cultivares de café com três repetições. The ratio of nitrogen (N) to potassium (K) is important in the production of specialty coffees because the relative amounts of N and K can either suppress the formation or increase the concentration of compounds that are essential to the flavor and aroma of specialty coffees. The purpose of this study was to evaluate the effect of different N:K ratios (w/w) and K doses on the cup quality of coffee. The concentrations of chemical compounds essentials to achieve good flavor and aroma, the N and K contents, and the caffeine contents of coffee leaves and beans were evaluated in this study. Extracted articular cartilage from the femurs and tibias of 15 Wistar rats at three days old to isolate chondrocytes. This work evaluated the qualitative and quantitative cellular changes induced by treatment with 5-aminouracil (5-AU) and a combination of 5-AU and caffeine in plant cells in relation to DNA damage, repaired damage, and residual damage. As biological material, Allium cepa L. root tips were used, grown in filtered water, in darkness, with aeration at constant temperature of 25 °C ± 0.5. Cell populations were synchronized using 5 mM caffeine in order to study the effects of 5-AU and caffeine/5-AU combined treatment on the DNA content and their incidence in the entrance to mitosis. Checkpoint response to DNA damage involves the activation of DNA repair and G2 lengthening subpathways. The roles of nibrin (NBS1) and the ATM/ATR kinases in the G2 DNA damage checkpoint, evoked by endogenous and radio-induced DNA damage, were analyzed in control, A-T and NBS lymphoblast cell lines. Short-term responses to G2 treatments were evaluated by recording changes in the yield of chromosomal aberrations in the ensuing mitosis, due to G2 checkpoint adaptation, and also in the duration of G2 itself. The role of ATM/ATR in the G2 checkpoint pathway repairing chromosomal aberrations was unveiled by caffeine inhibition of both kinases in G2. Modulation by caffeine of calcium-release microdomains in frog skeletal muscle fibers Biol. Res.

The effects of caffeine on the process of excitation-contraction coupling in amphibian skeletal muscle fibers were investigated using the confocal spot detection technique. This method permits to carefully discriminate between caffeine effects on the primary sources of Ca2+ release at the Z-lines where the triads are located and secondary actions on other potential Ca Release sources. Our results demonstrate that 0.5 mM caffeine potentiates and prolongs localized action-potential evoked Ca2+ transients recorded at the level of the Z-lines, but that 1mM only prolongs them. Characterisation of AC1: a naturally decaffeinated coffee Bragantia

We compared the biochemical characteristics of the beans of a naturally decaffeinated Arabica coffee (AC1) discovered in 2004 with those of the widely grown Brazilian Arabica cultivar "Mundo Novo" (MN). Although we observed differences during fruit development, the contents of amino acids, organic acids, chlorogenic acids, soluble sugars and trigonelline were similar in the ripe fruits of AC1 and MN. AC1 beans accumulated theobromine, and caffeine was almost entirely absent. Tests on the supply of [2-14C] adenine and enzymatic analysis of theobromine synthase and caffeine synthase in the endosperm of AC1 confirmed that, as in the leaves, caffeine synthesis is blocked during the methylation of theobromine to caffeine. Mineral nutrition and caffeine content in coffee leaves Bragantia

The effect of nutrient supply on the caffeine content of coffee (Coffea arabica L.) leaves was investigated. Seeds were germinated in nutrient-agar media lacking N, P, K, Ca, Mg, S, Zn, B or Mo. The control treatment contained all essential nutrients. The caffeine concentration was determined seven months after seed sowing when the seedlings have 3 to 4 pair of leaves. The omission of K induced the highest caffeine content in the leaves (24.5 g.kg-1). Caffeine in the control treatment was 21.9 g.kg -1. Absence of P induced the lowest content, 17.5 g.kg-1.

Spectrophotometric determination of tannins and caffeine in preparations from Paullinia cupana var. sorbilis BABT

The present work was designed to quantify the caffeine and total polyphenols in the extractive solution and the granulated form from the seeds of Paullinia cupana var. sorbilis, by the spectrophotometric method. The method showed linearity for the caffeine and polyphenols in the range of 5-25 µg/ml and 2.4-5.6 µg/ml respectively. The solutions of the semipurified fraction (EPA) and granulated form (GRA) showed linear responses in the range of 0.288-0.672 and 0.4-1.2 µg/ml, respectively. The precision and accuracy were determined for the EPA solution at a concentration of 100 µg/ml. The spectrophotometric method performed well in quantifying the caffeine and total polyphenols.

In this work the author puts in evidence a practical and at the same time rapid method for the determination of the cafein quantity in erva-mate, that may be utilized to perfection in food and industrial analyses or others that not demand scientific rigour. The author presents a description of the method and makes a comparison with the classical method of Grandval-Lajoux, showing that the differences between the one and the other are practically nil. Caffeine formation by suspension cultures of Coffea dewevrei BABT

The low caffeine content in leaves of C. dewevrei (~ 0.5 mg/g) is due to a low biosynthesis associated with a fast degradation. On the other hand, high biosynthesis and low degradation confer a higher content (~ 8 mg/g) in leaves of C. arabica. In this work it was observed that cell cultures of C. dewevrei recovered the ability to synthesize caffeine almost in similar levels of C. arabica cultures. Tracer experiments with labelled carbon dioxide showed a significant accumulation of radioactivity in caffeine and metabolites, indicating an active biosynthesis. Hydroxypropylcellulose controlled release tablet matrix prepared by wet granulation: effect of powder properties and polymer composition BABT

The aim of this study was to attain 100% drug release of caffeine after 24 h from hydroxypropylcellulose (HPC) tablet matrices and to investigate the effect of co-excipient. Physical properties of the powders were evaluated and suggested for a wet granulation process. The tablet containing caffeine was formulated by different weight ratios of hydrophilic polymers. The results of polymer evaluation confirmed that the increase of HPC level with the same drug content significantly decreased the rate of drug release. The presence of co-polymer excipients carboxymethylcellulose (CMC) and polyvinylpyrrolidone (PVP) in the tablet matrix was also investigated. The objective of this study was to evaluate the influence of integral and decaffeinated coffee brews (Coffea arabica L and C. canephora Pierre) on the metabolic parameters of rats fed with hyperlipidemic diet. Thirty male Wistar rats (initial weight of 270 g ± 20 g) were used in the study, which were divided into six groups five each. The treatments were normal diet, hyperlipidemic diet, hyperlipidemic diet associated with integral coffee arabica or canephora brews (7.2 mL/kg/day) and hyperlipidemic diet associated to decaffeinated arabica, or canephora brews, using the same dosage. The influence of caffeine and thymol on the survival, growth and reproduction of Subulina octona (Brugüière, 1789) (Mollusca, Subulinidae) BABT

Subulina octona is a terrestrial snail which serves as an intermediate host for the parasites. It is also an agricultural pest. The aim of this work was to assess, during 120 days, the effects of caffeine and thymol at 2.5 g/L and 5 g/L on the hatchability, survival after hatching, growth and reproduction of S. octona under the laboratory conditions. A total of 240 eggs, 240 juveniles aged 10-day-old, and 240 aged 30-day-old were tested. The results showed that thymol (at 2.5 g/L and 5 g/L) and caffeine (at 5 g/L) acted as ovicides. Stability of phenolic compounds and antioxidant capacity of regular and decaffeinated coffees BABT

This study compared the regular and decaffeinated coffees in relation to antioxidant capacity, levels of some antioxidant molecules and stability of these parameters over a six-month period under different storage conditions. The regular coffee samples analyzed right after the industrial production showed higher antioxidant capacity (ORAC and DPPH), the same levels of phenolic compounds and higher levels of phenolic acids than decaffeinated coffee. Our main aim was to evaluate the effect of coffee intake regarding clinical, biochemical tests and liver biopsy data in treatment naïve patients with chronic hepatitis C. One hundred and thirty-six patients with chronic hepatitis C, diagnosed through liver biopsy, or by means of clinical, ultrasound or endoscopic signs of cirrhosis, were assessed by determination of biochemical tests, metabolic and morphological alterations. Bovine serum albumin potentiates caffeine- or ATP-induced tension in human skinned skeletal muscle fibers BJMBR
Human skinned muscle fibers were used to investigate the effects of bovine serum albumin (BSA) on the tension/pCa relationship and on the functional properties of the Ca2+-release channel of the sarcoplasmic reticulum (SR). In both fast- and slow-type fibers, identified by their tension response to pSr 5.0, BSA (0.7-15 µM) had no effect on the Ca2+ affinity of the contractile proteins and elicited no tension per se in Ca2+-loaded fibers. Controversy still exists concerning the potential ergogenic benefit of caffeine (CAF) for exercise performance. The purpose of this study was to compare the effects of CAF ingestion on endurance performance during exercise on a bicycle ergometer at two different intensities, i.e., approximately 10% below and 10% above the anaerobic threshold (AT).
