Background: Cyclamen persicum is an economically important ornamental crop that is propagated exclusively through seeds as vegetative propagation using cuttings is not possible. Therefore a micropropagation method through somatic embryogenesis is of high interest; however the method suffers from low reliability concerning quality and quantity of the produced plantlets. A crucial step of the protocol is the removal of plant growth regulators (PGRs) that triggers embryo development. In order to get a better insight in this crucial step of the propagation process, a gene expression analysis has been set up using five different genes of glutathione S-transferases (GST) as these are known to be auxin responsive as well as stress reactive. Results: One out of... |

The objective of this work was the optimization of the conditions of in vitro culture for callus production of Silybum marianum (L.) Gaertn. (Asteraceae). Sections of cotyledons, previously disinfected by washing successively with ethanol 70º, NaClO (10% w/v) and Tween 20 (0.05% v/v) and rinsing with sterile distilled water, were used as explants. For its initial culture, B5 medium supplemented with BA and 2,4-D solidified with phytagel was used, and a 63% survival was achieved. To obtain callus, two solid media were assayed (S1 and S2) using B5 medium supplemented with growth regulators (BA and 2,4-D or NAA and BA, respectively). The calli were grown at 25ºC during 45 days in darkness. Growth kinetics was studied using S1 medium... |

The aim of this work was to explore the possibility of obtaining transgenic plants of onion varieties cultivated in Argentina, starting from calli induced from mature zygotic embryos, using two strains of Agrobacterium tumefaciens as transfection vectors. Mature embryos from three varieties of 'Valenciana' onion, Torrentina, Cobriza INTA and Grano de oro were in vitro cultivated for callus induction. After three to four months an average of 57.4% success for the three varieties was reached. Transformation was carried out with Agl1 or LBA 4404 A. tumefaciens strains, both carrying a binary vector containing the marker gene gus a and the selection gene nptII. Selection was done in callus induction medium containing 10 mgL-1 geneticin during three... |

Somatic embryogenesis, which is still the method of choice for tissue culture, regeneration and transformation of maize, is largely considered highly genotype-dependent. The Hi II, a highly embryogenic genotype, has been extensively used in transformation protocols. However, this is not an inbred line; instead, it has a proportion of the undesirable A-188 background, and the progeny segregates for phenotypic characteristics and shows poor agronomic performance. In an effort to identify genotypes that combine a high somatic embryogenic response and good agronomic performance, we evaluated 48 advanced inbred lines developed at INTA. Callus development and somatic embryogenesis capacity were measured in 200 immature embryos per line. Embryogenic capacity [EC... |

A callus induction and plant regeneration protocol was developed from leaf and petiole explants of the endemic Astragalus nezaketae. Explants were cultured on Murashige and Skoog medium (MS) supplemented with different plant growth regulators (PGRs) [?-naphthaleneacetic acid (NAA), benzyladenine (BA), 2,4-dichlorophenoxyacetic acid (2,4-D), kinetin (Kin), thidiazuron (TDZ)]. The combinations and concentrations of PGRs were shown significant variations for the frequency of callus formation, appearence of callus and the potential of callus differentiation. NAA x BA have been found highly affective in callusing and plant regeneration. Other PGRs have not resulted in callus differentiation for shoot formation. The highest number of shoots (6/explants) was... |

A plant regeneration method with cell suspension cultures of banana, and the effect of biobalistic on regeneration potential are described in this report. Somatic embryos of banana were obtained from indirect embryogenesis of male inflorescence of banana cultivar Maçã (AAB group). Part of the calluses formed (40%) showed embryogenic characteristics (nonfriable, compact and yellow color). The cell suspension, originated from embryogenic calluses, contained clusters of small tightly packed cells with dense cytoplasms, relatively large nuclei and very dense nucleoli. After four months of culture, somatic embryos started to regenerate. The maximum number of regenerated plants was observed between 45 and 60 days after embryo formation. In the first experiment,... |

Most of the plant regeneration processes in citrus, through tissue culture, involve indirect somatic embryogenesis. The optimization of these processes is important for the development of in vitro plant improvement and micropropagation studies. Studies to evaluate the effect of different carbohydrates in somatic embryogenesis were conducted using calli from 'Ponkan' mandarin (Citrus reticulata, Blanco), 'Cravo' mandarin (C. reticulata), 'Itaboraí' sweet orange (C. sinensis L. Osbeck.), 'Valencia' sweet orange (C. sinensis) and 'Kinnow' mandarin (C. nobilis Loureiro x C. deliciosa Tenore). The culture medium used was MT supplemented with sucrose, galactose, glucose, maltose or lactose with the following concentrations of 18, 37, 75, 110, and 150 mM. The... |

Campomanesia adamantium is a native plant species of Brazilian Cerrado with diverse economic potential and great medicinal importance. Its sexual propagation is impaired by the recalcitrance of its seeds, which prevents effective and profitable propagation. With the purpose of establishing commercial crops and minimizing the extractive use of vegetal resources, the aim of the present study was to induce embryogenic calli in nodal segments of gabirobeira, and to determine and characterize their embryogenic phase through the establishment of a growth curve based on cellular characteristics. Calli were induced using nodal segments inoculated in WPM culture medium without the addition of hormones (control) and with different concentrations of 2,4-D, IAA, IBA,... |

Callus growth and plant regeneration from long-term callus cultures were studied in two elite clones of Asparagus officinalis cv. Argenteuil, to establish a suitable protocol for a prospective in vitro selection program. Callus initiation and growth was evaluated on MS medium with 3% sucrose, 0.9% agar, 1 mg.l-1 kinetin, and three levels of 2,4-D. The highest callus relative growth was obtained on medium with 1.5 mg.l-1 2,4-D and 1 mg.l-1 kinetin. Shoot primordia (SP) induction from >18-months-old calluses was evaluated on several media; the highest percentage of SP induction (89%) and average number of SP per callus (8.6) were obtained with clone ‘265' on MS medium with 5 mg.l-1 2iP, 1 mg.l-1 IAA, 3% sucrose and 0.9% agar. The highest... |

BACKGROUND: The aim of the present work was to examine the role of UV-C irradiation on the production of secondary metabolites (total phenolic, total flavanols, total flavonols, catechin, ferulic acid and trans-resveratrol in phenolic compounds and α-, β-, γ- δ-tocopherols) in callus cultures. Studies on the effects of UV-C treatment on callus culture are seldom and generally focused on UV-B. However UV-C radiation play an important role in accumule secondary metabolites. RESULTS: In this study, callus cultures from Öküzgözü grape cultivar were initiated from leaf petiole explants. Calli formed after 6 weeks on the medium supplemented with 0.5 mg L-1 benzylaminopurine (BA), 0.5 mg L-1 indole acetic acid (IAA) on B5... |

A method for the isolation of coupled mitochondria from the callus of Araucaria angustifolia is described for the first time. Mitochondria were isolated from embryogenic callus of A. angustifolia. They were metabolically active, able to sustain oxidative phosphorylation as shown by respiratory control ratio values, which were about 2.4 when respiring on succinate as substrate. Oxygen uptake experiments, using freeze-thawed disrupted mitochondria, showed the presence of alternative rotenone-insensitive NAD(P)H dehydrogenases, which were stimulated by Ca2+. The procedure now described for the isolation of A. angustifolia mitochondria is an important new tool, allowing the investigation of mitochondrial bioenergetics and metabolism and physiology of plants. |
