Background: Cyclamen persicum is an economically important ornamental crop that is propagated exclusively through seeds as vegetative propagation using cuttings is not possible. Therefore a micropropagation method through somatic embryogenesis is of high interest; however the method suffers from low reliability concerning quality and quantity of the produced plantlets. A crucial step of the protocol is the removal of plant growth regulators (PGRs) that triggers embryo development. The objective of this work was the optimization of the conditions of in vitro culture for callus production of Silybum marianum (L.) Gaertn. (Asteraceae). Sections of cotyledons, previously disinfected by washing successively with ethanol 70º, NaClO (10% w/v) and Tween 20 (0.05% v/v) and rinsing with sterile distilled water, were used as explants. For its initial culture, B5 medium supplemented with BA and 2,4-D solidified with phytagel was used, and a 63% survival was achieved. The aim of this work was to explore the possibility of obtaining transgenic plants of onion varieties cultivated in Argentina, starting from calli induced from mature zygotic embryos, using two strains of Agrobacterium tumefaciens as transfection vectors. Mature embryos from three varieties of 'Valenciana' onion, Torrentina, Cobriza INTA and Grano de oro were in vitro cultivated for callus induction. Somatic embryogenesis, which is still the method of choice for tissue culture, regeneration and transformation of maize, is largely considered highly genotype-dependent. The Hi II, a highly embryogenic genotype, has been extensively used in transformation protocols. However, this is not an inbred line; instead, it has a proportion of the undesirable A-188 background, and the progeny segregates for phenotypic characteristics and shows poor agronomic performance. A callus induction and plant regeneration protocol was developed from leaf and petiole explants of the endemic Astragalus nezaketae. Explants were cultured on Murashige and Skoog medium (MS) supplemented with different plant growth regulators (PGRs) [?-naphthaleneacetic acid (NAA), benzyladenine (BA), 2,4-dichlorophenoxyacetic acid (2,4-D), kinetin (Kin), thidiazuron (TDZ)]. The combinations and concentrations of PGRs were shown significant variations for the frequency of callus formation, appearence of callus and the potential of callus differentiation. The superfamily Olivoidea is broadly distributed in the world’s oceans mostly in coastal waters at tropical and subtropical latitudes. It encompasses around 30 Recent genera and 460 species. Two families – Olividae and Olivellidae – are classically recognized within the superfamily. Their shell is very characteristic due to the presence of a modified callused anterior end and a fasciole. Prior to the present work, neither the monophyly of the superfamily nor the relationships among its genera had been tested with molecular phylogenetics. A plant regeneration method with cell suspension cultures of banana, and the effect of biobalistic on regeneration potential are described in this report. Somatic embryos of banana were obtained from indirect embryogenesis of male inflorescence of banana cultivar Maçã (AAB group). Part of the calluses formed (40%) showed embryogenic characteristics (nonfriable, compact and yellow color). The cell suspension, originated from embryogenic calluses, contained clusters of small tightly packed cells with dense cytoplasms, relatively large nuclei and very dense nucleoli. This study aimed to develop a protocol of direct and indirect organogenesis of Phyllanthus tenellus Roxb. Estudou-se os efeitos de auxilias exógenas e ácido bórico, no enraizamento de estacas de lichia (Litchi chinensis Sonn.). As estacas foram uniformizadas, com 25 cm de comprimento e 4 folhas cortadas pela metade. Cerca de 2,5 cm da base das mesmas foi mergulhado nos tratamentos: H2O; Boro 150 µg/ml; IBA 5.000 ppm, IBA 2.000 ppm; IBA 5.000 ppm + Boro 150 µg/ml; IBA 2.000 ppm + Boro 150 µg/ml; NAA 3.000 ppm; NAA 1.500 ppm; NAA 3.000 ppm + Boro 150 µg/ml; NAA 1.500 ppm + Boro 150 µ/g/ml. Curcuma zedoaria Roscoe (zedoary) is a medicinal properties-bearing Zingiberaceae from which rhizomes are commercially exploited. The objective of this work was to establish an in vitro protocol for micropropagation and callogenesis of Curcuma zedoaria Roscoe as alternative to improve plant production, turning economically feasible the exploitation of its secondary metabolites which present medicinal properties. Plant regeneration after long term callus culture in clones of Asparagus officinalis L. Biocell

Callus growth and plant regeneration from long-term callus cultures were studied in two elite clones of Asparagus officinalis cv. Argenteuil, to establish a suitable protocol for a prospective in vitro selection program. Callus initiation and growth was evaluated on MS medium with 3% sucrose, 0.9% agar, 1 mg.l-1 kinetin, and three levels of 2,4-D. The highest callus relative growth was obtained on medium with 1.5 mg.l-1 2,4-D and 1 mg.l-1 kinetin. Normania triphylla is an endemic species from Madeira island (Portugal) extinct in the wild since 1991. The aim of this work was to culture the meristems of this species in vitro and to multiply its shoots in order to preserve this endangered species. The best results in terms of multiplication were obtained in Murashige and Skoog medium supplemented with 10 µM 6‑benzylaminopurine (BAP). The number of shoots, the number of nodes and the number of leaves were the most important in this medium. Improved method for isolation of coupled mitochondria of Araucaria angustifolia (Bert.) O. Kuntze BABT

A method for the isolation of coupled mitochondria from the callus of Araucaria angustifolia is described for the first time. Mitochondria were isolated from embryogenic callus of A. angustifolia. They were metabolically active, able to sustain oxidative phosphorylation as shown by respiratory control ratio values, which were about 2.4 when respiring on succinate as substrate. Oxygen uptake experiments, using freeze-thawed disrupted mitochondria, showed the presence of alternative rotenone-insensitive NAD(P)H dehydrogenases, which were stimulated by Ca2+. The procedure now described for the isolation of A. angustifolia mitochondria is an important new tool, allowing the investigation of mitochondrial bioenergetics and metabolism and physiology of plants.
