Cancer, Photodynamic Therapy and Porphyrin-Type Derivatives Anais da ABC (AABC)
GOMES,ANA T.P.C.; NEVES,MARIA G.P.M.S.; CAVALEIRO,JOSÉ A.S..
ABSTRACT This review has two parts. The first one gives an approach to interdisciplinary studies against cancer carried out by many scientists using porphyrin-type substrates as photosensitizers in PDT. Intensive studies were performed for almost six decades. The successes really started in 1993 with the first formulation patented under the trade name Photofrin, which was immediately approved in several countries to treat certain types of cancer. Photofrin is still used although certain negative features soon became well known. That has motivated the search for better new photosensitizers. Several ones were developed, evaluated and a few of them had clinical approval. This group includes porphyrin derivatives and pro-drugs (aminolevulinic acid and its...

Studies on the Synthesis of Vitamin D Analogs with Aromatic D-Ring Anais da ABC (AABC)
EDUARDO-CANOSA,SILVINA; SIGÜEIRO,MARÍA MARCO, RITA; MOURIÑO,ANTONIO.
ABSTRACT Herein, we describe our studies on the synthesis of 1α,25-dihydroxyvitamin D3 analogs possessing a benzene ring replacing the natural 5-membered D-ring by the Wittig-Horner and dienyne approaches. A key feature is the synthesis of a Cr(CO)3-complexed previtamin D derivative that enables the construction of vitamin D analogs with aromatic D-ring through a thermal [1,7]-H sigmatropic shift. This study establishes the basis for the design of new vitamin D analogs containing aromatic D-ring, complexed or uncomplexed to Cr(CO)3 type moieties for specific molecular recognition and drug research and development.

Adiponectin serum levels in female dogs bearing mammary gland carcinomas Arq. Bras. Med. Vet. Zootec.
Souza,I.B.R.; Cardoso,C.V.; Pereira,A.A.F.; Poppe,S.C.; Bondan,E.F..

Immunohistochemical and molecular expression of laminin-332 gamma-2 chain in canine mammary tumors Arq. Bras. Med. Vet. Zootec.
Zuccari,D.A.P.C; Castro,R; Jardim,B.V; Mancini,U.M; Polachini,G.M.
Forty-eight cases of canine mammary cancer were investigated to evaluate the immunohistochemical distribution of the γ2 chain of laminin-332. Tumor cells were compared to a pool of normal mammary tissues using quantitative RT-PCR. The western blot was performed in eight tumor samples as complementary test to evaluate protein integrity. Immunohistochemistry experiments showed negative, focal, and weak expression of laminin-332 γ2 in tumors with the worst prognosis. Quantitative PCR revealed downregulation of the gene in 27 (56.2%) of the animals. Out of the 16 dogs with γ2 chain overexpression, seven were still alive. The western blot results showed bands generation of 36, 50, and 98kDa, suggesting degradation of laminin-332 γ2 in malignant tumors. The...

Lipid peroxidation in female dogs bearing mammary gland carcinomas Arq. Bras. Med. Vet. Zootec.
Souza,I.B.; Cardoso,C.V.; Poppe,S.C.; Bondan,E.F..
Nesse...

In vitro effects of 2-methoxyestradiol-bis-sulphamate on cell growth, morphology and cell cycle dynamics in the MCF-7 breast adenocarcinoma cell line Biocell
Vorster,Chris; Joubert,Annie.
In the search for new and improved anticancer therapies, researchers have identified several potentially useful compounds. One of these agents is 2-methoxyestradiol-bis-sulphamate (2ME-BM), a sulphamoylated derivative of 2-methoxyestradiol. The objective of this study was to evaluate 2ME-BM's in vitro efficacy as antiproliferative agent in the MCF-7 breast adenocarcinoma cell line. Light- and fluorescent microscopy showed decreased cell density, increased apoptotic characteristics and significant ultrastructural aberrations indicative of autophagic cell death after 24 hours of exposure at a concentration of 0.4µM. In addition, mitotic indices revealed that 2ME-BM induces a G2M block. The latter was confirmed by flow cytometric analyses where increased...

Mqoco,Thandi; Marais,Sumari; Joubert,Annie.
2-Methoxyestradiol-bis-sulphamate is a bis-sulphamoylated derivative of the naturally occurring 17-beta-estradiol metabolite namely 2-methoxyestradiol. 2-Methoxyestradiol-bis-sulphamate is regarded as a potential anticancer drug with increased antiproliferative activity when compared to 2-methoxyestradiol. The aim of this pilot in vitro study was to determine the influence of 2-methoxyestradiol-bis-sulphamate on cell growth, morphology and possible induction of certain types of cell death in the SNO esophageal carcinoma cell line. A dose-dependent study (0.2-1.0µM) was conducted with an exposure time of 24 hours. Data revealed that 2-methoxyestradiol-bis-sulphamate reduced cell numbers statistically significantly to 74% after exposure to 0.4µM of the drug....

Galectin-8 promotes migration and proliferation and prevents apoptosis in U87 glioblastoma cells Biol. Res.
Metz,Claudia; Döger,Remziye; Riquelme,Elizabeth; Cortés,Priscilla; Holmes,Christopher; Shaughnessy,Ronan; Oyanadel,Claudia; Grabowski,Catalina; González,Alfonso; Soza,Andrea.
BACKGROUND: Glioblastoma is one of the most aggressive cancers of the brain. Malignant traits of glioblastoma cells include elevated migration, proliferation and survival capabilities. Galectins are unconventionally secreted glycan-binding proteins that modulate processes of cell adhesion, migration, proliferation and apoptosis by interacting with beta-galactosides of cell surface glycoproteins and the extracellular matrix. Galectin-8 is one of the galectins highly expressed in glioblastoma cells. It has a unique selectivity for terminally sialylated glycans recently found enhanced in these highly malignant cells. A previous study in glioblastoma cell lines reported that Gal-8 coating a plastic surface stimulates two-dimensional motility. Because in other...

Glucose transporters: expression, regulation and cancer Biol. Res.
MEDINA,RODOLFO A.; OWEN,GARETH I..
Mammalian cells depend on glucose as a major substrate for energy production. Glucose is transported into the cell via facilitative glucose transporters (GLUT) present in all cell types. Many GLUT isoforms have been described and their expression is cell-specific and subject to hormonal and environmental control. The kinetic properties and substrate specificities of the different isoforms are specifically suited to the energy requirements of the particular cell types. Due to the ubiquitousness of these transporters, their differential expression is involved in various disease states such as diabetes, ischemia and cancer. The majority of cancers and isolated cancer cell lines over-express the GLUT family members which are present in the respective tissue of...

The clinical impact of [18F]-FDG Pet during the opening year of a Pet centre BABT
Talbot,Jean-Noël.
We have evaluated the clinical impact of FDG-PET on patient staging and management during the opening year of our PET centre in France. A questionnaire, translation in French of the questionnaire used recently in California, was sent to the referring physician of each of the 476 patients who had at least one routine FDG-PET examination during the year 2000. Of 348 responses (response rate = 73%), the disease was upstaged in 26% of the cases and downstaged in 9%. Inter-modality management changes (change from a scheduled therapeutic modality for a different one) were reported in 37% of the cases and intra-modality changes in 9%. Those modification rates were respectively 38% and 7% in recurrence of colorectal cancer (153 patients), 47% and 7% in lung cancer...

Positron emission tomography (PET) and its recent update PET/CT are very effective diagnostic tools for non-invasive imaging of metabolic or functional disorders in target tissues. The clinical usefulness of fluorodeoxyglucose-(18F) (FDG) has been now widely accepted. Recently, the clinical usefulness of fluoroDOPA-(18F) or FDOPA, an aminoacid labelled with the same positron emitter fluorine-18, has been evaluated and recognised in France and subsequently in several EU countries. FDOPA is diagnostic PET agent, which has been used for decades in imaging the loss of dopaminergic neurons in Parkinson's disease, and more recently to detect, stage and restage neuroendocrine tumours and to search for recurrence of viable glioma tissue. The present article...

Modeling for Taxol® Separation in a simulated moving bed BABT
Cremasco,Marco Aurelio; Starqui,Axel Nicolas.
This work presents an alternative numerical resolution strategy for a model to describe the dynamic of linear adsorption processes involving multicomponent mixture of taxanes with Taxol® (paclitaxel), a powerful anti-cancer agent, and non-identified impurities, in a Simulated Moving Bed (SMB) system. To solve the model, a hybrid method were used. The liquid concentration inside the particles was found analytically and was related with the liquid bed concentration using Duhamel's theorem. The results from simulation were compared with experimental ones from the literature, showing a good agreement, which demonstrated the applicability of the model and of the hybrid resolution proposed.
