Effects of nutritional conditions on lipid production by cyanobacteria Anais da ABC (AABC)
CORDEIRO,RAQUEL S.; VAZ,IZABELA C.D.; MAGALHÃES,SÉRGIA M.S.; BARBOSA,FRANCISCO A.R..
ABSTRACT The present study evaluated the effects of the culturing media and the levels of nitrogen and phosphorus on the growth, biomass productivity and lipid production of four species of Microcystis (M. novacekii, M. aeruginosa, M panniformis and M. protocystis). The lipid extract was obtained by refluxing with dichloromethane (Soxhlet). The biomass and biomass productivity yields were maximized with ASM-1 medium treatment enriched with nitrogen and/or phosphorus (0.25-0.65 g/L and 25-50.7 mg/L d-1, respectively). The lipid extract yields from M. panniformis and M. novacekii were inversely correlated with the concentration of nitrogen and directly correlated with the concentration of phosphorus (35.8 % and 31.7 %). The lipid extract yield from M....

Growth capacity of thermotolerant campylobacters in culture media supplemented with pig and cow blood BABT
Tresierra-Ayala,Álvaro; Navas,Manuel; Flores,Josué; Perea,Ramsés; Huanaquiri,Juan; Bendayán,María; Fernández,Heriberto.
In this work 60 thermotolerant Campylobacter strains (37 C. jejuni and 23 C. coli) isolated from the cows, pigs, chickens and ducks (15 strains of each type of animal) were used to establish their growth capacity on media containing cow or swine blood as potential substitutes of sheep or horse blood. The growth capacity was assessed by viable counts on cow and swine blood media, using the modified Miles and Misra method. Campylobacter strains showed better growth in the media supplemented with pig or sheep blood than with cow blood. Thus, the use of pig blood could be a supplement for Campylobacter culture medium, when there was no availability of sheep or horse blood.

Assessment of the metabolism of different strains of Bacillus megaterium BABT
Suarez,Carlos Alberto Galeano; Montano,Inti Doraci Cavalcanti; Nucci,Edson Romano; Iemma,Monica Rosas da Costa; Giordano,Raquel de Lima Camargo; Giordano,Roberto de Campos.
Bacillus megaterium is a promising host for expression of heterologous proteins. This paper reports the nutrient consumption patterns and production of metabolites for three different strains of B. megaterium, ATCC 14945, QMB 1551 and PV 361, which is QMB 1551 with seven constitutive plasmids deleted. 14 h cultivations in agitated flasks were run, for two different media: A (LB plus 10g/L glucose) and B (medium A, with the yeast extract replaced by tryptone). Strains PV361 and QMB 1551 showed higher maximum specific growth rates in medium B, reaching 0.42 h-1 and 0.48 h-1 respectively. The main by-products of the glucose overflow mechanism were acetate and lactate, for all three strains, which had preferential amino acids for substrate: Ala, Asp, Glu, Ser....

Evaluation of gelling agents on anther culture: response of two soybean cultivars BABT
Cardoso,Milena Barcelos; Bodanese-Zanettini,Maria Helena; Mundstock,Elsa Cristina de; Kaltchuk-Santos,Eliane.
Anthers of two soybean cultivars were cultured in B5 long basal culture media gelled with agarose or PhytagelTM. Cytological examinations of the anthers were carried out during the first 45 days of culture to assay the viability and developmental stage of microspores. Frequency of callus formation was recorded at 45 days of culture. The analysis of variance of the microspore viability assay showed significant Cultivar X Gelling Agent X Day of Culture interactions. The frequencies of viable microspores decreased significantly with time of culture, within each cultivar and gelling agent tested. The interaction Day X Cultivar was significant for the frequencies of binucleate symmetrical grains and multinucleate/multicellular structures. The effect of gelling...

Growth of the Ectomycorrhizal Fungus Pisolithus Microcarpus in different nutritional conditions BJM
Rossi,Márcio José; Oliveira,Vetúria L..
The most important plant species employed in reforestation programs depend on ectomycorrhizal fungi for their establishment and growth. The exploitation of this symbiosis to improve forest productivity requires fungal inoculants in a large scale level. To develop such a technology it is necessary to define the optimal composition of the culture medium for each fungus. With these objectives in mind, the effect of the composition of the culture medium on biomass production of the ectomycorrhizal fungus Pisolithus microcarpus (isolate UFSC-Pt116) was studied. The original composition of two culture media, already employed for cultivation of ectomycorrhizal fungi, was submitted to several variations with the C/N ratio as the main variable. A variation of the...

Toxigenic potential of Aspergillus flavus tested in different culture conditions Ciênc. Tecnol. Aliment.
Ritter,Ana Carolina; Hoeltz,Michele; Noll,Isa Beatriz.
The objective of the present work was to evaluate the capacity of three isolates of Aspergillus flavus to produce aflatoxin under different culture conditions. This experiment was based on a 2³ factorial design, in which the independent variables were temperature (20-40 °C), incubation time (7-21 days), and the pH (2.0-6.0) in two different synthetic media. The optimal conditions were applied to non-aflatoxigenic isolates previously tested in coconut agar. Aflatoxin B1 was extracted directly from the synthetic cultures with chloroform. Thin Layer Chromatography (TLC) and Photographic Photometry were utilized to identify and quantify the compounds. Preliminary results showed that YES agar was an alternative medium for detecting the toxigenic potential of...

Culture alternative medium for the growth of extreme halophilic bacteria in fish products Ciênc. Tecnol. Aliment.
Yeannes,María Isabel; Ameztoy,Irene Mabel; Ramirez,Elida Elvia; Felix,Mónica María.
Halotolerant or halophilic (Archaeabacteria) microorganisms can be found in salted and ripening fish products that are not affected by salt. They can be moderate or extremely halophilic bacteria. The extremely halophilic bacteria require between 15-30% of NaCl for growth. The extremely halophilic archaeobacteria may be selectively isolated in different media. The aim of this work was to determine the effectiveness of the Salt-Agar-Milk medium, a medium modified in our laboratory through the addition of MgSO4 and KCl - named SAMm, and its effect on the bacterial growth by means of comparison with other media, with and without milk, determining time of incubation and counting. Two samples of salted fish from local fish salting factories and two laboratory...

Production and characterization of algae extract from Chlamydomonas reinhardtii Electron. J. Biotechnol.
Kightlinger,Weston; Chen,Kai; Pourmir,Azadeh; Crunkleton,Daniel W; Price,Geoffrey L; Johannes,Tyler W.
Background: Algae offer many advantages as biofuel sources including: high growth rates, high lipid content, the ability to grow on non-agricultural land, and the genetic versatility to improve strains rapidly and produce co-products. Research is ongoing to make algae biofuels a more financially attractive energy option; however, it is becoming evident that the economic viability of algae-based fuels may hinge upon high-value co-products. This work evaluated the feasibility of using a co-product, algae extract, as a nutrient source in cell culture media. Results: Algae extract prepared from autolysed Chlamydomonas reinhardtii was found to contain 3.0% protein, 9.2% total carbohydrate, and 3.9% free α-amino acid which is similar to the nutrient...

PEREIRA,GUILLERMO; ALBORNOZ,VERÓNICA; ROMERO,CHRISTIAN; LARA,SEBASTIÁN; SÁNCHEZ-OLATE,MANUEL; RÍOS,DARCY; ATALA,CRISTIAN.
ABSTRACT Orchids require symbiotic fungi and/or specific conditions to germinate. Asymbiotic techniques have been shown successful for orchid germination. In Chile, Chloraea include many endemic, and potentially ornamental, terrestrial orchid species. In this study, individuals of Chloraea crispa, C. gavilu and C. virescens were manually autopollinated. The resulting capsules were sterilized and seeds were aseptically obtained. We evaluated asymbiotic germination in: Agar Water (AW), Knudson C (KC), Banana Culture Media (CMB), Tomato Culture Media (CMT), Malmgren Modified (MM), Murashige and Skoog (MS), and MS modified (MS1/2). Seeds were incubated in the dark at 24±1 ºC for 2 weeks. Then they were put in 16/8 h light/dark cycles for 14 weeks. We...
