Screening common bean germplasm for resistance to genetically diverse Sclerotinia sclerotiorum isolates from Argentina Agronomy
Abán, Carla Luciana; Taboada, Gisel Maria; Casalderrey, Norma Beatriz; Maggio, Maria Elisa; Chocobar, Mario Osvaldo; Spedaletti, Yamila Andrea; Gonzalez, Mónica Alicia Aparicio; Vizgarra, Oscar Vicéforo; Galván, Marta Zulema.
White mold caused by Sclerotinia sclerotiorum (Lib.) de Bary is a devastating disease that affects the common bean (Phaseolus vulgaris. L) crop worldwide. In Argentina, white mold has been detected in all bean production areas, reaching seed yield and quality losses up to 100% on susceptible common bean cultivars under favorable weather conditions. The aim of this study was to screen the physiological resistance of 20 common bean accessions to five genetically distinct isolates of S. sclerotiorum collected from the main common bean growing area of Argentina, using the greenhouse straw test. The white mold reaction was scored at 7, 14, and 21 days post-inoculation using a 1 (no disease symptoms) to 9 (severely diseased or dead plants) scale and the area...

Evaluation of soybean breeding lineages to new sources of root-knot nematode resistance Ciência e Agrotecnologia
Hamawaki,Osvaldo Toshiyuki; Hamawaki,Raphael Lemes; Nogueira,Ana Paula Oliveira; Glasenapp,Jacqueline Siqueira; Hamawaki,Cristiane Divina Lemes; Silva,Carolina Oliveira da.
ABSTRACT In Brazil, as the soybean crop expands the losses caused by nematodes as well as its incidence in producing areas have increased. Therefore, this work aimed to develop a non-genetically modified soybean which combines desirable traits as the root-knot nematode resistance, early-maturity, high seed yielding, and high oil and protein contents. Consequently, seeds (F2) derived from the double-crossing between (Guarani x Sambaiba) x (M Soy 6101 x Conquista) were bulked to grow the F3 generation. Following generations were advanced by the SSD method of breeding until F6. Thus, plants showing superior characteristics were planted separately to perform progeny tests and then advanced breeding lines were thoroughly tested and compared to standards in...

Protective effect of an antimicrobial peptide from Mytilus edulis chilensis expressed in Nicotiana tabacum L. Electron. J. Biotechnol.
Arenas,Gloria; Marshall,Sergio H.; Espinoza,Valeria; Ramírez,Ingrid; Peña-Cortés,Hugo.
A "defensin-like" antibacterial peptide from Mytilus edulis chilensis, was sub-cloned into a binary vector for expression in plant tissues. The resulting new clone was electroporated into A. tumefaciens to transform tobacco plants. The presence of the construct in transgenic tobacco lines was demonstrated through RT-PCR, Northern and Western blots. Transformed positive plants were selected and grown for challenging. Tobacco leaves were infiltrated with Pseudomonassyringae pv. syringae and visual lesions determined at different times post-exposure. Of seven plants exposed, four gave variable protection up to seven days post-infection while one of them appears to be fully protected. These results suggest that defensin-like antimicrobial peptides from...

Asian soybean rust (ASR), caused by the phytopathogenic fungi Phakopsora pachyrhizi, has caused large reductions in soybean (Glycine max) yield in most locations in Brazil where it has occurred since it was first reported in May 2001. Primary efforts to combat the disease involve the development of resistant cultivars, and four dominant major genes (Rpp1, Rpp2, Rpp3 and Rpp4) controlling resistance to ASR have been reported in the literature. To develop new long-lasting soybean ASR resistance genes, we used field experiments to assess ASR leaf lesion type in 11 soybean genotypes (BR01-18437, BRS 184, BRS 231, BRS 232, BRSGO Chapadões, DM 339, Embrapa 48, PI 200487, PI 230970, PI 459025-A and PI 200526) and the 55 F2 generations derived from their...

Differential expression of genes identified from Poncirus trifoliata tissue inoculated with CTV through EST analysis and in silico hybridization Genet. Mol. Biol.
Cristofani-Yaly,Mariângela; Berger,Irving J.; Targon,Maria Luisa P.N.; Takita,Marco A.; Dorta,Sílvia de O.; Freitas-Astúa,Juliana; Souza,Alessandra A. de; Boscariol-Camargo,Raquel L.; Reis,Marcelo S.; Machado,Marcos A..
Citrus is the most important fruit crop in Brazil and Citrus tristeza virus (CTV) is considered one of the most important pathogens of citrus. Most citrus species and varieties are susceptible to CTV infection. However, Poncirus trifoliata, a close relative of citrus, is resistant to the virus. In order to better understand the responses of citrus plants to the infection of CTV, we constructed expressed sequence tag (EST) libraries with tissues collected from Poncirus trifoliata plants, inoculated or not with Citrus tristeza virus at 90 days after inoculation, grafted on Rangpur lime rootstocks. We generated 17,867 sequence tags from Poncirus trifoliata inoculated (8,926 reads) and not (8,941 reads) with a severe CTV isolate. A total of 2,782 TCs...

An AFLP marker linked to the Pm-1 gene that confers resistance to Podosphaera xanthii race 1 in Cucumis melo Genet. Mol. Biol.
Teixeira,Ana Paula Matoso; Barreto,Fatima Aparecida da Silva; Camargo,Luis Eduardo Aranha.
Brazil produced 330,000 metric tons of melons in 2005, principally in the Northeast region where one of the most important melon pathogens is the powdery mildew fungus Podosphaera xanthii. The disease is controlled mainly by incorporating single dominant resistance genes into commercial hybrids. We report on linkage analysis of the Pm-1 resistance gene, introgressed from the AF125Pm-1 Cantalupensis Charentais-type breeding line into the yellow-fleshed melon (Group Inodorus) breeding line AF426-S by backcrossing to produce the resistant line AF426-R, and the amplified fragment length polymorphism (AFLP) marker M75/H35_155 reported to be polymorphic between AF426-S and AF426-R. Segregation analysis of M75/H35_155 using a backcross population of 143 plants...

Reaction of Capsicum peppers commercialized in the Federal District to bacterial wilt Horticultura Brasileira
Rossato,Maurício; Santiago,Thais R.; Lopes,Carlos Alberto.
ABSTRACT In Brazil, the bacterial pathogens Ralstonia solanacearum and R. pseudosolanacearum cause substantial losses by inducing bacterial wilt on several solanaceous crops; R. pseudosolanacearum is the main species associated with peppers (Capsicum sp.). To verify the bacterial wilt reaction on Capsicum peppers commercialized in the Federal District (DF), fruits of several genotypes within this genus were collected from six different fairs distributed in the satellite cities of Gama, Sobradinho and Guará. Seedlings with four true leaves derived from seeds extracted from such fruits were root inoculated with 108 CFU/mL with a representative isolate of R. pseudosolanacearum (race 1, biovar 3, phylotype I, sequevar 18). The evaluated species were: Capsicum...

Sources of resistance against the Pepper yellow mosaic virus in chili pepper Horticultura Brasileira
Bento,Cíntia dos S; Rodrigues,Rosana; Zerbini Júnior,Francisco Murilo; Sudré,Cláudia P.
The Pepper yellow mosaic virus (PepYMV) naturally infects chili and sweet pepper, as well as tomato plants in Brazil, leading to severe losses. This work reports the reaction to the PepYMV of 127 Capsicum spp. accessions, aiming at identifying resistance sources useful in breeding programs. The experiment was carried out in a completely randomized design, with eight replications, in greenhouse conditions. Plants were protected with an insect-proof screen to avoid virus dissemination by aphids. Leaves of Nicotiana debneyi infected with the PepYMV were used as the inoculum source. Plants were inoculated with three to four fully expanded leaves. A second inoculation was done 48 hours later to avoid escapes. Only the youngest fully expanded leaf was...
