Micropropagated banana plants induced by gamma irradiation and resistant to the root-knot nematode reproduction CIGR Journal
Taha, Rania Abdel-Ghaffar; El-Nagdi, Wafaa M. A.; Kamel, Hedaya Ahmed.
Micropropagation of banana plants has been used successfully in commerce. Banana plantation has a great interest in Egypt due to suitable climate. However, some problems might thwart the plantation progress like the root-knot nematode; Meloidogyne incognita infection which can affect acclimatization and plantation of in vitro plants. Thus, our investigation aimed to produce nematode resistant in vitro plants using gamma radiation (γ). Banana in vitro plantlets were exposed to gamma radiation at several doses then replanted in vitro. Sensitivity of banana plants to irradiation was determined. Acclimatized plants were infected with M. incognita in greenhouse to see their capability for tolerance. Results indicated that irradiated plants could manage the...

Ionizing radiation quarantine treatments Anais da SEB
Hallman,Guy J..
Irradiation is a viable quarantine disinfestation treatment which has been studied for 40 years although it has received very little commercial use. Two principal obstacles to commercial application, 1) the fact that insects are not killed immediately, and 2) consumer opposition to irradiation, have been allayed to some extent, but the remaining impediment to large-scale commercial use is development of approved protocols by government regulatory agencies in importing countries. The United States Department of Agriculture has taken the first step by allowing for the movement of a number of quarantined fruits from Hawaii to the rest of the country following irradiation treatment. The measures of efficacy of irradiation disinfestation treatments should be...

VICCINI,LYDERSON FACIO; SARAIVA,LUIZ SÉRGIO; ALMEIDA FILHO,JOSÉ DE; CRUZ,COSME DAMIÃO; ANDRADE,ROGÉRIO ALVAREZ DE.
The identification of chemical substances which increase the efficiency of radiation is important to make easier the obtention of plants with structural chromosome aberrations which may be used in an alternative program for hybrid maize production. The present work was carried out to investigate the effect of the chemical substance metronidazole in maize seedlings submitted to gamma radiation. Several treatments were done, soaking the seeds in solutions with varied concentrations of the active substance combined with solution filtration and gamma radiation. On the third day of the experiment, germination percentage, root and stem lengths were evaluated. At a high concentration (1,250 mg/50 mL) metronidazole behaved as a radiosensibilizer in the presence of...

Effect of gamma radiation on lipids by the TBARS and NMR BABT
Silva,Adriana Cristina de Oliveira; Mársico,Eliane Teixeira; Jesus,Edgar Francisco Oliveira de; Guimarães,Carlos Frederico; Cortez,Marco Antonio Sloboda.
The aim of this work was to study the effect of gamma radiation on lipids by TBARS and NMR. The samples of raw whole milk were subjected to gamma radiation from Co60 in doses of 1, 2 and 3 kGy and the production of rancidity was studied through Nuclear Magnetic Resonance (NMR) and Thiobarbituric Acid Test (2-TBARS). The TBARS values increased according to the intensity of the radiation dose applied at the samples, demonstrating correlation between the radiation dose and the production of lipid oxidation. This was confirmed by NMR with the formation of peaks of aldehydes and ketones that were small and similar in the doses of 1 and 2 kGy. In the dose of 3 kGy, the total degradation of milk fat was observed. A correlation between the NMR and 2-TBA was...

Changes in electrophoretic profiles of Ipomoea batatas (sweet potato) induced by gamma radiation BABT
Lage,Celso Luiz Salgueiro; Vasconcellos,Alexandre Guimarães; Silva,Nina Claudia Barboza da; Esquibel,Maria Apparecida.
The ability of nodal segments of Ipomoea batatas to differentiate shoots and roots was evaluated after gamma irradiation. Shoot differentiation was less sensitive to irradiation than roots. However, at 90 Gy, no shoot was able to regenerate a new plant; in contrast 76 % of the roots from irradiated nodal segments continued to grow. The gamma radiation also induced changes in electrophoretic profiles of peroxidases of storage roots. Plants originated from irradiated storage roots presented changes in leaf peroxidase profiles very similar to those produced by leaves directly irradiated. The peroxidase profile of absorbent roots from irradiated storage roots was different from that obtained from directly irradiated absorbent roots.

Effects of 60Co gamma radiation on crotamine BJMBR
Boni-Mitake,M.; Costa,H.; Spencer,P.J.; Vassilieff,V.S.; Rogero,J.R..
Ionizing radiation can change the molecular structure and affect the biological properties of biomolecules. This has been employed to attenuate animal toxins. Crotamine is a strongly basic polypeptide (pI 10.3) from Crotalus durissus terrificus venom composed of 42 amino acid residues. It induces skeletal muscle spasms leading to a spastic paralysis of hind limbs in mice. The objective of the present study was to carry out a biochemical study and a toxic activity assay on native and irradiated crotamine. Crotamine was purified from C.d. terrificus venom by Sephadex G-100 gel filtration followed by ion-exchange chromatography, and irradiated at 2 mg/ml in 0.15 M NaCl with 2.0 kGy gamma radiation emitted by a 60Co source. The native and irradiated toxins...

Effect of gamma radiation on the inactivation of aflatoxin B1 in food and feed crops BJM
Ghanem,I.; Orfi,M.; Shamma,M..
Samples of food crops (peanut, peeled pistachio, unpeeled pistachio, rice, and corn) and feed (barley, bran, corn) were autoclave-sterilized, and inoculated with 10(6) of spore suspension of an isolate of Aspergillus flavus fungus known to produce aflatoxin B1 (AFB1) . Following a 10-day period of incubation at 27 C to allow for fungal growth, food and feed samples were irradiated with gamma radiation at the doses 4, 6, and 10 kGy. Results indicated that degradation of AFB1 was positively correlated with the increase in the applied dose of gamma ray for each tested sample. At a dose of 10 kGy percentages of AFB1 degradation reached highest values at 58.6, 68.8, 84.6, 81.1 and 87.8% for peanuts, peeled pistachios, unpeeled pistachios, corn and rice samples,...

Inactivation of Escherichia coli O157:H7 in hamburgers by gamma irradiation BJM
Chirinos,Rodolfo R.O.; Vizeu,Dirceu M.; Destro,Maria Teresa; Franco,Bernadette D.G.M.; Landgraf,Mariza.
Escherichia coli O157:H7 causes bloody diarrhoea, haemorrhagic colitis and life-threatening complications like haemolytic uremic syndrome and thrombotic thrombocitopenic purpura. Among foods associated with outbreaks caused by this pathogen, hamburger is the most common one. The aim of this research was to determine the radiation dose to reduce the population of E. coli O157:H7 in hamburgers to non-detectable levels in order to render a safer product. Hamburgers, inoculated with Escherichia coli O157:H7, were exposed to gamma radiation (60Co) treatment, with doses ranging from 0 to 0.7 kGy. The average temperature during the process was 5.6ºC. Non-inoculated hamburgers were submitted to sensory evaluation after being exposed to irradiation doses of 0.8 kGy...

Calibration curves by 60Co with low dose rate are different in terms of dose estimation – a comparative study Genet. Mol. Biol.
Mendes,Mariana Esposito; Mendonça,Julyanne Conceição Goes de; Hwang,Suy; Giorgio,Marina Di; Lima,Fabiana Farias de; Santos,Neide.
Abstract Biological dosimetry aims to estimate individual absorbed doses due ionizing radiation exposure. The dicentric chromosomes are considered the most specific biomarker for dose estimation. This study aimed to compare calibration curves for linear low energy transfer (LET) radiation built from low dose rates and whether they vary in terms of dose estimation. For that we did a search in the literature of all calibration curves produced with low dose rates and we simulated the dose estimation from pre-established dicentric’s frequencies. The information on methodologies and cytogenetic results of each study were analyzed. As expected dose rate influence β coefficients, especially at higher doses. However, we have seen that some doses were not...

Effect of ionizing radiation onVibrio bacteria in Crassostrea virginica (American oyster) ArchiMer
Eberiel, D; Dooley, K; Langton, L; Malleth, J.
The presence of Vibrio bacteria in raw shellfish poses a significant health hazard to the consuming public. These human pathogens are natural, seasonal components of the microbial flora of shellfish which can result in primary septicemic or diarrheal fatal infections. Existing shellfish depuration neither targets nor eliminates the Vibrio bacteria. For this reason, additional purification processes are necessary. In this study, the effect of low dose gamma radiation on the inactivation ofVibrio cholerae (01 and non 01) and vulnificus is investigated. Log decrement (D10) values ranging only to a high of 0.09 KGy in the Vibrio strains support the hypothesis that food irradiation technology can provide an additional effective and efficient shellfish...

Mutagenesis in Lupinus polyphyllus a tool for producing spring variants? JBAG
Fatta,N.; Mascarini,A; Lopez,S..
Lupinus polyphyllus remains vegetative during the first year, stores cold during winter and is induced to blossoming in the second year. Inflorescences are attractive and varied coloured terminal clusters. In Buenos Aires the historical average winter temperature is 11.5 ºC and the number of hours of cold are not enough for L. polyphyllus to flower so there is no commercial offer in pots. The line of work chosen to produce genetic variability in L. polyphyllus, was induced mutagenesis with the purpose of rescuing genetic variants that meet the required cold naturally in Buenos Aires and capable of flowering and producing viable seeds. Seeds harvested in Bariloche were sown on september 4, 2003. 600 seedlings received gamma radiation at a rate of 50 Gy and...
