Genetic diversity in rosewood saplings (Aniba rosaeodora ducke, Lauraceae): an ecological approach Acta Amazonica
Santos,Ronaldo Pereira; Spironello,Wilson Roberto; Sampaio,Paulo de Tarso Barbosa.
This article takes an ecological approach to the genetic diversity of Rosewood (Aniba rosaeodora Ducke) in a central Amazonian terra firme forest north of Manaus. Planted Rosewood setting, under partial shaded canopy, were assessed in terms of fruiting production, frugivory, and seed dispersal. Using RAPD molecular analysis procedures, the influence of the spatial distribution of adult trees on the genetic diversity (polymorphism) of saplings was assessed with genetic samples from 34 reproductive trees and 60 saplings. The density and distribution patterns the reproductive trees did not modify the sapling"s diversity (1.86%, AMOVA). Two types of adult tree dispersion were identified; i) clumped and ii) more widely dispersed. Polymorphism (77.5%) and gene...

PONTES,Lígia Cristine Gonçalves; MOURA,Elisa Ferreira; MOURA,Mônika Fecury; RODRIGUES,Simone de Miranda; OLIVEIRA,Maria do Socorro Padilha de; CARVALHO,José Edmar Urano de; THERRIER,Josette.
ABSTRACT The bacurizeiro (Platonia insignis Mart.) is a tree native to the Amazon whose fruit is much used in the gastronomy in the North and Northeast regions of Brazil. Due to its great economic potential for these regions, the species is being conserved in germplasm banks to support genetic breeding programs. The aim of this work was the molecular characterization of P. insignis accessions belonging to the germplasm bank of the Embrapa Eastern Amazon research unit using ISSR (Inter Simple Sequence Repeat) markers. Seventy-eight accessions of P. insignis belonging to 16 progenies were sampled in two different localities on Marajó Island, state of Pará, Brazil. Among the 16 progenies, seven were collected in Soure and nine in Salvaterra. The 78 accessions...

Pollen and seed dispersal of Brazil nut trees in the southwestern Brazilian Amazon Acta Amazonica
MARTINS,Karina; SANTOS,Raifanny da Silva Oliveira dos; CAMPOS,Tatiana de; WADT,Lúcia Helena de Oliveira.
ABSTRACT Pollen and seed dispersal patterns greatly influence the spatial distribution of plant genetic diversity. Microsatellite-based parentage analysis provides accurate estimates of contemporary gene dispersal. Although most tropical trees have been shown to exhibit widespread pollen dispersal, few studies have estimated contemporary gene dispersal after seedling establishment. Bertholletia excelsa (Lecythidaceae) is pollinated by large-bodied bees, while previous seed-tracking experiments suggest their seeds are mainly dispersed across very short distances by scatter-hoarding rodents, who primarily act as seed predators. Here we used parentage analysis to provide contemporary estimates of pollen and seed dispersal in B. excelsa recruits. We examined...

SYSTEMATIC CONSIDERATIONS ON TWO SPECIES OF NECTARIVOROUS BATS (Anoura caudifer AND A. geoffroyi) BASED ON BARCODING SEQUENCES Acta biol.Colomb.
VARGAS-ARBOLEDA,Andrés; CUADRADO-RÍOS,Sebastian; MANTILLA-MELUK,Hugo.
ABSTRACT As a product of recent revisions of the nectivore bat genus Anoura (Glossophaginae: Anourina), new species have been described, and several taxa have been elevated to the specific level in the so-called Anoura caudifer and A. geoffroyi species complexes in the last four decades. The genus has a wide distribution across regions of high ecological complexity, including geographic features, such as the Andes and the Isthmus of Panamá, previously identified as natural barriers that restrict gene flow in other phyllostomid bat genera. Up to date, no analyses have been conducted to investigate the contribution of biogeographic factors on the genetic divergence of the genus. In this work, we analyze barcoding DNA sequences (cytochrome oxidase subunit I,...

Genetic differentiation of the neotropical tree species Protium spruceanum (Benth.) Engler (Burseraceae) between fragments and vegetation corridors in Brazilian Atlantic forest Acta Botanica
Vieira,Fábio de Almeida; Carvalho,Dulcinéia de.
We studied patterns of genetic differentiation in a connected landscape with an interesting history of human habitat conversion that began two centuries ago, during the Brazilian colonization period. In the fragments of Brazilian Atlantic seasonal forest and corridors of secondary forest, Protium spruceanum is an abundant native, mass-flowering/insect-pollinated and bird-dispersed tree. Genetic diversity was analyzed from 230 individuals in five fragments (1 to 11.8 ha) and four corridors (460 to 1000 m length) using allozyme loci. We did not find evidence of inbreeding within fragments or corridors, but the proportion of heterozygotes (<IMG SRC="/img/revistas/abb/v23n4/a28simb1.gif" WIDTH=287 HEIGHT=96>) were significantly higher in fragments than...

Genetic structure of Dicksonia sellowiana Hook. (Dicksoniaceae) reveals clinal distribution along the latitudinal gradient of the Atlantic Forest Acta Botanica
Fagundes,Bruna Saviatto; Poersch,Maria Augusta; Santos,Jaqueline dos; Gaglioti,André Luiz; Labiak,Paulo Henrique; Muschner,Valeria Cunha.
ABSTRACT Dicksonia sellowiana is the only species of the genus occurring in Brazil. Its distribution is restricted to humid areas of the Atlantic Forest biome. The distribution pattern of biodiversity in this biome is known to have been influenced by historical and environmental factors, although the pattern for ferns remains unknown. This study is first to describe the genetic structure of D. sellowiana along the latitudinal gradient of the Atlantic Forest biome. We use microsatellite markers to estimate genetic diversity and structure for 267 individuals representing 14 populations of D. sellowiana from the Atlantic Forest. The results (Ho, He, Fst, Fis, distance genetic) support the hypothesis of a pattern of biodiversity discontinuity. We found greater...

Miranda,Kássia Marques Corrêa; Guimarães,Rejane Araújo; Silva,Marcos José da; Oliveira,Patrícia Rasteiro Ordiale; Ribeiro,Thania Gonçalves; Mendes,Thainara Policarpo; Telles,Mariana Pires de Campos; Soares,Thannya Nascimento.
ABSTRACT The genus Manihot Mill. contains about 120 species of which about 104 occur in Brazil. We tested the cross-amplification of ten microsatellite markers developed for Manihot esculenta in 15 species of Manihot endemic to the Brazilian Cerrado. We also evaluated the genetic diversity of Manihot irwinii, M. orbicularis, and M. purpureocostata. Ten pairs of primers were amplified among 14 species of Manihot. The percentage of polymorphic loci per species varied from 70 to 100 %. Nine markers showed amplification and polymorphism when evaluated on polyacrylamide gel. The markers were combined to form three sets for multiplex genotyping for genetic diversity analysis, and showed 51, 75, and 75 alleles in M. irwinii, M. orbicularis, and M....

ABSTRACT Understanding the genetic structure and diversity of plants is fundamental to their conservation and permits their sustainable use by local communities. The genus Maytenus (Celastraceae) is composed of plants possessing pharmacological and antioxidant properties. However, the genetic and economic properties of the species M. dasyclada, a typical species of Araucaria forests in Brazil and Uruguay, have been little studied. In this work, the genetic structure and diversity of natural populations of M. dasyclada located in unprotected and preserved forest remnants were investigated using RAPD and isozymes markers. The results demonstrated that in areas of preservation, populations of M. dasyclada possess a relatively high degree of polymorphism and...

Pati,Prakash G; Datta,Subhojit; Agbagwa,Ikechukwu O; Singh,Indra Prakash; Soren,Khela Ram; Das,Alok; Choudhary,Arbind Kumar; Chaturvedi,Sushil Kumar.
Resistance gene analog (RGA)-anchored amplified fragment length polymorphism (AFLP-RGA) marker system was used in order to evaluate genetic relationships among 22 pigeon pea genotypes with varied responses to Fusarium wilt and sterility mosaic disease. Five AFLP-RGA primer combinations (E-CAG/wlrk-S, M-GTG/wlrk-S, M-GTG/wlrk-AS, E-CAT/S1-INV and E-CAG/wlrk-AS) produced 173 scorable fragments, of which 157 (90.7%) were polymorphic, with an average of 31.4 fragments per primer combination. The polymorphism rates obtained with the five primers were 83.3%, 92.0%, 92.3%, 93.0% and 93.1%, respectively. Mean polymorphic information content (PIC) values ranged from 0.24 (with E-CAT/S1-INV) to 0.30 (with E-CAG/wlrk-AS), whereas resolving power (RP) values varied...

Morphological and genetic perspectives of hybridization in two contact zones of closely related species of Petunia (Solanaceae) in southern Brazil Acta Botanica
Teixeira,Marcelo C.; Turchetto,Caroline; Hartke,Sara; Schnitzler,Carolina K.; Freitas,Loreta B..
ABSTRACT Interspecific hybridization has several consequences for parental species, from blurring species limits to the emergence of new taxa. Petunia axillaris and P. exserta occur in sympatry in southernmost Brazil and naturally hybridize despite their different pollination syndromes and habitat requirements. We employed genetic and morphological analyses to characterize two contact zones between the species with the aim of determining the effect of interspecific hybridization. Microsatellite loci and a morphometric evaluation of the corolla shape were used to classify individuals based on their origin as pure parental or hybrids. Corolla color was used to classify individuals a priori (white, red or intermediate, for P. axillaris, P. exserta or hybrid,...

Genetic diversity in Calibrachoa pygmaea (Solanaceae): A hawkmoth-pollinated nightshade from the Pampas Acta Botanica
Mäder,Geraldo; Backes,Alice; Reck-Kortmann,Maikel; Freitas,Loreta B..
ABSTRACT Calibrachoa pygmaea is a unique species of Calibrachoa, especially concerning its flower morphology and the environment where it occurs. The species is self-incompatible and is narrowly distributed in wet and flooded fields of the Pampas region. We characterize the genetic diversity of the species based on traditional plastid markers and newly developed nuclear microsatellites to identify drivers that guide its evolution. Our results identified markers that are informative and useful for studying the population structure of C. pygmaea, as well as that of other species of Calibrachoa. Both marker sets were congruent in developing conclusions regarding the evolutionary scenario of C. pygmaea, and revealed that the genetic variability and population...

Hierarchical genetic clusters for phenotypic analysis Agronomy
Matta, Luiza Barbosa da; Tomé, Lívia Gracielle Oliveira; Salgado, Caio Césio; Cruz, Cosme Damião; Silva, Letícia de Faria.
Methods to obtain phenotypic information were evaluated to help breeders choosing the best methodology for analysis of genetic diversity in backcross populations. Phenotypes were simulated for 13 characteristics generated in 10 populations with 100 individuals each. Genotypic information was generated from 100 loci of which 20 were taken at random to determine the characteristics expressing two alleles. Dissimilarity measures were calculated, and genetic diversity was analyzed through hierarchical clustering and graphic projection of the distances. A backcross was performed from the two most divergent populations. A set of characteristics with variable heritability was taken into account. The environmental effect was simulated assuming . For hierarchical...

Genetic similarities among four species of the Plectranthus (L’Hér.) genus - DOI: 10.4025/actascibiolsci.v32i1.880 Biological Sciences
Bandeira, Juliana Magalhães; UFPel; Bianchi, Valmor João; UFPel; Rubin, Sílvia; UFPel; Peters, José Antonio; UFPel; Braga, Eugenia Jacira Bolacel; UFPel.
The Plectranthus genus comprises several species generally referred to as boldo, which are highly used in popular medicine due to their anti-dyspeptic, analgesic and digestion-stimulating properties. The amount of natural active principles can vary greatly in the related genotypes, which may lead to the inappropriate use of these plants. This work aimed to analyze the interspecific diversity among four species of the Plectranthus genus (P. grandis, P. barbatus, P. neochilus and P. amboinicus) and the intraspecific diversity of P. barbatus collected from different places in southern Brazil, by means of the RAPD technique. A higher genetic similarity was observed between the P. neochilus and P. amboinicus species (80%), followed by P. grandis and P. barbatus...
