ABSTRACT The bacurizeiro (Platonia insignis Mart.) is a tree native to the Amazon whose fruit is much used in the gastronomy in the North and Northeast regions of Brazil. Due to its great economic potential for these regions, the species is being conserved in germplasm banks to support genetic breeding programs. The aim of this work was the molecular characterization of P. insignis accessions belonging to the germplasm bank of the Embrapa Eastern Amazon research unit using ISSR (Inter Simple Sequence Repeat) markers. Developing an in vitro optimized protocol to sweet potato landraces conservation Agronomy

This study aimed to develop a protocol for the in vitro conservation of 30 accessions of sweet potato collected in the northern region of Rio de Janeiro State, Brazil, aiming to extend the period between subcultures for this species. An in vitro minimal growth experiment was established with accession UENF 1931 in a factorial arrangement consisting of four concentrations of mineral salts in MS medium and four concentrations of sucrose, at two temperatures. ABSTRACT: Adoption of resistant cultivars is the primary measure used to control anthracnose stalk rot. The goal of this study was to identify maize-resistant genotypes to anthracnose stalk rot, which are similar to the hybrid 2B710. Experiments were performed at Embrapa Maize and Sorghum experimental fields in Brazil. The first experimental trial evaluated 234 maize lines as well as two commercials hybrids, BRS1010 (susceptible) and 2B710 (resistant). ABSTRACT Cryopreservation is a technique used to preserve genetic material at ultra-low temperatures in cryogenic germplasm banks. In order to enable the cryopreservation of a seed, it is necessary to study its appropriate freezing temperature and its water content, because seeds reach maximum physiological quality with high water contents, and they tolerate drying when they are orthodox seeds, but they may not tolerate cryopreservation. Development and characterization of polymorphic microsatellite markers for the sweet orange (Citrus sinensis L. Osbeck) Genet. Mol. Biol.

The aims of this study were to develop simple sequence repeat markers (SSRs or microsatellite markers) in citrus and to evaluate the efficiency of these markers for characterization of sweet orange. We developed SSRs from a genomic library of 'Pêra IAC' sweet orange enriched for AG/TC, GT/CA, TCA/AGT and AAC/TTG sequence repeats. We selected 279 sequences from which 171 primer pairs were designed of which 113 with the best banding patterns were selected. Characterization of sweet orange microsatellite loci revealed that AG/TC was the most abundant (69%) microsatellite class isolated, followed by GT/CA (15.9%), TCA/AGT (8%) and AAC/TTG (6.2%). Jacaranda decurrens (Bignoniaceae) is an endemic species of the Cerrado with validated antitumoral activity. The genetic diversity of six populations of J. decurrens located in the State of São Paulo was determined in this study by using molecular markers for randomly amplified polymorphic DNA (RAPD) and amplified fragment length polymorphism (AFLP). Following optimization of the amplification reaction, 10 selected primers generated 78 reproducible RAPD fragments that were mostly (69.2%) polymorphic. A core collection of the common bean (Phaseolus vulgaris L.), representing genetic diversity in the entire Mexican holding, is kept at the INIFAP (Instituto Nacional de Investigaciones Forestales, Agricolas y Pecuarias, Mexico) Germplasm Bank. Visando o melhoramento genetico da sacaca, foi instalado um Banco Ativo de Germoplasma (BAG) na Embrapa Amazonia Ocidental. Estao sendo feitas coletas e multiplicacao vegetativa de acesso de sacaca de diversos locais da Amazonia. O BAG-Sacaca visa nao somente a conservacao do germoplasma, mas o estudo da variabilidade genetica entre acesso, para selecao de genotipos superiores para producao de biomassa (folhas), e com maior potencial de producao de oleos essenciais. Resumo: Esta publicação tem por objetivo descrever a história, a composição da Coleção de trabalho de Microrganismos Patogênicos a Caprinos e Ovinos, e de que forma ela tem conservado, caracterizado, documentado e utilizado seus isolados. A coleção foi criada em 2012 e atualmente é composta por 141 cepas bacterianas e seis cepas virais, pertencentes às espécies Corynebacterium pseudotuberculosis, Staphylococcus aureus e ao gênero Lentivirus. Esses isolados são utilizados para produção de antígenos para imunodiagnósticos, em diagnósticos moleculares e na avaliação de resistência a antimicrobianos e ação de anti-sépticos e antimicrobianos. Por considerar o melhoramento genetico linha basica e prioritaria paraa cultura do cupuacu (Theobroma grandiflorum), o CPAA tem em seus objetivos a obtencao de materiais produtivos, com frutos de bom tamanho e boa qualidade, resistentes a pragas e doencas rusticas, com amplo periodo de colheita, com variedades precoces, medianas e tardias. A metodologia de trabalho desenvolvida consta de selecao, coleta e introducaode germoplasma, caracterizacao e avaliacao do germoplasma introduzido e sua utilizacao no processo de melhoramento. Resistance to Fusarium dry root rot disease in cassava accessions PAB

The objective of this work was to identify sources of resistance to dry root rot induced by Fusarium sp. in cassava accessions. A macroconidial suspension (20 µL) of 11 Fusarium sp. isolates was inoculated in cassava roots, from 353 acessions plus seven commercial varieties. Ten days after inoculation, the total area colonized by the pathogen on the root pulp was evaluated by digital image analysis. Cluster analysis revealed the presence of five groups regarding resistance. The root lesion areas ranged from 18.28 to 1,096.07 mm² for the accessions BGM 1518 and BGM 556, respectively. The genotypes BGM 1042, BGM 1552, BGM 1586, BGM 1598, and BGM 1692 present the best agronomical traits.

In vitro conservation of Piper aduncum and Piper hispidinervum under slow-growth conditions PAB

The objective of this work was to evaluate in vitro storage of Piper aduncum and P. hispidinervum under slow-growth conditions. Shoots were stored at low temperatures (10, 20 and 25°C), and the culture medium was supplemented with osmotic agents (sucrose and mannitol - at 1, 2 and 3%) and abiscisic acid - ABA (0, 0.5, 1.0, 2.0 and 3.0 mg L-1). After six-months of storage, shoots were evaluated for survival and regrowth. Low temperature at 20ºC was effective for the in vitro conservation of P. aduncum and P. hispidinervum shoots. In vitro cultures maintained at 20ºC on MS medium showed 100% survival with slow-growth shoots. Variability of ricin content in mature seeds of castor bean PAB

The objective of this work was to evaluate ricin concentration in castor bean seeds (Ricinus communis) of 20 accessions from the Banco de Germoplasma de Mamoneira of the Embrapa Algodão, Campina Grande, PB, Brazil, using the Enzyme Linked Immunosorbent Assay. Significant differences were observed among accessions. BRA 3271 had the highest ricin concentration in seeds (32.18 ng µg-1), and BRS Paraguaçu had the lowest (3.53 ng µg-1). There is the possibility of selecting genotypes with different ricin concentrations, which can be used according on the interest of the breeding programs.

Germplasm characterization of three jabuticaba tree species Rev. Bras. Frutic.

The purpose of this study was to characterize cultivated genotypes of three jabuticaba species (Plinia cauliflora, P. trunciflora, and P. jaboticaba). Phenology and fruit growth, as well as leaf, flower and fruit traits were evaluated. Variability in all traits was observed among genotypes of the three jabuticaba species. The trait peduncle size is indicated for differentiation of the three species under study. The leaf and fruit sizes of the genotypes P. trunciflora 3, P. trunciflora 4, P. trunciflora 5 and P. jaboticaba 1 differ from those described in the literature for these species, indicating the formation of ecotypes. Jabuticaba fruit skin contains high anthocyanin and flavonoid concentrations, with potential use in food and pharmaceutical industries.
