Effects of gene therapy on cardiovascular symptoms of lysosomal storage diseases Genet. Mol. Biol.

Abstract Lysosomal storage diseases (LSDs) are inherited conditions caused by impaired lysosomal function and consequent substrate storage, leading to a range of clinical manifestations, including cardiovascular disease. This may lead to significant symptoms and even cardiac failure, which is an important cause of death among patients. Currently available treatments do not completely correct cardiac involvement in the LSDs. Gene therapy has been tested as a therapeutic alternative with promising results for the heart disease. In this review, we present the results of different approaches of gene therapy for LSDs, mainly in animal models, and its effects in the heart, focusing on protocols with cardiac functional analysis.

Thirty hearts from adult individuals, 15 from male individuals and 15 female individuals, from the Anatomy Institute of Severino Sombra University were used. After a paramedian incision in the sternocostal joint, a careful dissection was performed in order to expose and identify the sinoatrial nodal artery (SAN), a right or left coronary artery branch. The domain was registered as right, if the right coronary branch irrigated the SAN. The same was listed for the left coronary artery. Increasingly the use and convenience of electrical appliances in our daily lives are the cause of harmful effects caused by electromagnetic fields (EMF). The aim of this study was to research the effect of EMF on the ultrastructure of the heart in EMF exposed rats. In this study 45 male Sprague Dawley rats ranging in weight between 260 and 280 grams were used. Origin and Development of the Coronary Arteries International Journal of Morphology

The formation of the coronary vasculature is a fundamental event in heart development and involves a series of carefully regulated temporal events that include vasculogenesis and angiogenesis. This review focuses the knowledge concerning the formation of the coronary arteries available so far and some molecular mechanisms involved in this process. Understanding coronary embryogenesis is important for interventions regarding adult cardiovascular diseases as well as those necessary to correct heart congenital defects. The insight of the coronary artery development as a result of ingrowth changed the understanding of several congenital coronary artery variations and anomalies described in gross anatomy.

Destruxin A, a toxic cyclodepsipeptide, was purified from culture filtrates of the entomopathogenic fungus Metarhizium anisopliae (Metsch.) Sorok. by Reverse-Phase HPLC, and assayed for its effects on a larval Manduca sexta (L.) (Lepidoptera: Sphingidae) heart preparation. Destruxin A was found to cause reversible and dose-dependent acceleration of the heart beat. The heart bioassay was highly sensitive to pulse applications of destruxin A at concentrations as low as 35 pmol. Chronic valvular disease: correlation between clinical, electrocardiographic, radiographic and echocardiographic aspects in dogs Arq. Bras. Med. Vet. Zootec.

Echocardiographic aspects of chronic mitral valvular disease were studied and compared to physical, radiographic and electrocardiographic aspects. Seventy dogs were used, and clinical examination, thoracic radiography, electrocardiogram and echocardiogram were performed. Correlations between regurgitation severity with cardiac failure functional class and murmur intensity were observed. The electrocardiogram showed a low sensibility in detecting cardiac chamber enlargement, caused by mitral regurgitation. All the dogs with severe mitral regurgitation showed cardiomegaly according to thoracic radiographies.

ABSTRACT Peccaries are wild mammals belonging to the Tayassu genus that are found almost everywhere in the Americas and have demonstrated great potential as an experimental model for scientific investigations. Twelve healthy adult animals were sedated to perform echocardiographic examinations in B, M and Doppler mode. The variables that exhibited statistically significant correlation coefficients with weight were LVFWd, LVIDd, LVIDs, E wave, A’RV, MAM, and TAPSE. The aim of this study was to investigate the effects of training programs on serum lipid profile and myocardial oxidative stress. Male Wistar rats (2 mo-old) were divided into three groups (n=8): sedentary (S), loadless trained (T) and trained-overload 2% body weight (TL). T and TL were trained through swimming for 9 weeks. T and TL rats had increased myocardial lipoperoxide (TBA) and lipid hydroperoxide (HP), whereas HP was higher in TL than in T animals. Superoxide dismutase (SOD) activities were lowest in TL. Chronic stimulation of sympathetic nervous activity contributes to the development and maintenance of hypertension, leading to left ventricular hypertrophy (LVH), arrhythmias and cardiac death. Moxonidine, an imidazoline antihypertensive compound that preferentially activates imidazoline receptors in brainstem rostroventrolateral medulla, suppresses sympathetic activation and reverses LVH. We have identified imidazoline receptors in the heart atria and ventricles, and shown that atrial I1-receptors are up-regulated in spontaneously hypertensive rats (SHR), and ventricular I1-receptors are up-regulated in hamster and human heart failure. Alkaline phosphatase (ALP) is important in calcification and its expression seems to be associated with the inflammatory process. We investigated the in vitro acute effects of compounds used for the prevention or treatment of cardiovascular diseases on total ALP activity from male Wistar rat heart homogenate. ALP activity was determined by quantifying, at 410 nm, the p-nitrophenol released from p-nitrophenylphosphate (substrate in Tris buffer, pH 10.4). Oxytocin (OT), a nonapeptide, was the first hormone to have its biological activities established and chemical structure determined. It was believed that OT is released from hypothalamic nerve terminals of the posterior hypophysis into the circulation where it stimulates uterine contractions during parturition, and milk ejection during lactation. However, equivalent concentrations of OT were found in the male hypophysis, and similar stimuli of OT release were determined for both sexes, suggesting other physiological functions.
