ABSTRACT Salinity is one of the major environmental stress factors that affect crop productivity, as well as interfering with plant growth and development, resulting in reduced production quality. Given this, we highlight the importance of research in response to plants subjected to salt stress in order to assess the physiological and biochemical behavior of genotypes, with the objective of selecting the more tolerant. One way to ensure the uniformity of the response of the plants is through in vitro cultivation, which allows control of the cultivation conditions. Therefore, the objective of this work was to evaluate the response of two commercial varieties of sugar cane exposed to saline stress in different conditions (gradual and abrupt). Two varieties... |

ABSTRACTCattleya intermediais an Atlantic Forest species endemic to Brazil that is classed as vulnerable on the list of threatened species. In this study, C. intermedia plantlets were micropropagated in an asymbiotic culture and the influence of different concentrations of sucrose (15, 30, 45 and 60 g L-1, plus a zero sucrose medium) and macronutrient salts (complete Murashige and Skoog (MS) medium and half MS medium (with half-strength macronutrients)) on survival and development of the plantlets was evaluated. In all treatments 100% plantlet survival was achieved. The integrated analysis of height of aerial part, number of leaves per plantlet, fresh mass, number of roots per plantlet and length of the longest root showed that the plantlets exhibited... |

Monoxenic cultures of arbuscular mycorrhizal fungi (AMF) in transformed roots have been used to better understand the symbiosis with these fungi, but few species have been successfully established in vitro. The object was to establish monoxenic cultures of Gigaspora decipiens and Glomus clarum and to verify the effects of temperature and pH on spore formation. Cultures were established from superficially disinfested spores that were germinated on an agar and water. After germination, the spores were transferred to Petri dishes containing transformed carrot roots. After 4-6 days formed newly formed spores and structures typical. The effects of temperature (22, 25, 28 or 32°C) and pH (4.0, 4.5, 5.5 or 6.5) on the production of spores were assessed over three... |

Research on anatomical modifications under in vitro culture is essential to the definition and understanding of the development of micropropagated plants. Likewise, such research is essential to improve the steps of the acclimatization process. Accordingly, the objective of the present study is to verify the differences in the leaves anatomical traits of micropropagated orchids under calcium silicate concentrations. Seedlings of in vitro-germinated seeds, measuring 0.5 cm in length, were inoculated in 250 cm3 pots with 60 mL MS culture medium and different silicon concentrations (0, 0.5, 1.0 and 2.0 mg L-1) in different culture environments (natural environment, in a greenhouse and an artificial environment in a growth chamber) and in all statistical... |

The tree fern Cyathea corcovadensis (Raddi) Domin is an endangered species in the state of Rio Grande do Sul, Brazil. It currently occurs only in the northern segment of the coastal region. Spore storage would help in conservation programs since it maintains genetic variability and provides material for in vitro cultures. Current study evaluates the effect of low temperatures combined to different spore storage times on the germination and initial gametophyte development of C. corcovadensis. Spores were divided into two groups: spores of the first group were sowed immediately in Meyer culture medium with nystatin, at pH 4.0, while spores of the second group were stored at 7, -20 and -196°C during 60, 120, 180, and 365 days and then sowed in the same... |

ABSTRACT An efficient regeneration system is a pre-requisite for the application of genetic transformation and functional genomics study of important plants. In this study, the effect of different factors (plant growth regulators, casein hydrolysate, aspartic acid and ascorbic acid) on in vitro embryogenesis and regeneration of Arta, Bahar and Zagros cultivars from mature and immature explants were investigated. Immature and mature embryos were dissected from disinfected seeds 20-25 days after pollination and imbedded mature seeds, respectively, and cultured on MS (Murashige and Skoog) medium supplemented with different compounds. The results showed that immature embryos expose high capacity of embryogenesis and regeneration in comparison with mature... |

The aim of this study was to establish an efficient protocol for the in vitro propagation of the endangered, endemic and decorative species Dianthus pinifolius Sibth. et Sm. The effects of different concentrations of 6-benzylaminopurine (BAP) and naphtalenacetic acid (NAA), and different explant types (single-node cuttings, terminal buds and shoot cuttings) on shoot multiplication were examined on Murashige and Skoog (MS) and half-strength MS media. The best results were obtained for shoot cuttings on the MS medium supplemented with 0.5 mg/L BAP and 0.1 mg/L NAA, achieving a maximum rate of regeneration (100%) and a total of 15.4 newly-developed shoots per explant. The highest rooting rate (96.7%) was obtained on MS medium containing 1 mg/L NAA, while the... |

ABSTRACT The in vitro seed germination is an effective alternative for quickly obtaining explants with sanitary quality. However, jenipapo seeds present slow and uneven germination. Therefore, internal and external factors to seed which directly interfere in the process, they must be identified, in order to adapt better techniques to obtain seedlings. In this sense, this work aimed to optimize the in vitro germination of Genipa americana L. seeds by evaluating different factors (light quality, GA3 treatment, pre-soaking in distilled water, growing media and stratification in the dark). It was found that the seed germination of G. americana was indifferent to light, however, the best results were obtained under conditions of continuous darkness; There was... |

A. Bastías, R. Almada, P. Rojas, J.M. Donoso, P. Hinrichsen, and B. Sagredo. 2016. Aging gene pathway of microRNAs 156/157 and 172 is altered in juvenile and adult plants from in vitro propagated Prunus sp. Cien. Inv. Agr. 43(3):429-441. In vitro culture is a very popular technique to mass propagate valuable plant genotypes, including Prunus sp. cultivars. Plants that undergo tissue culture processes often change their morphology and behavior due to the "rejuvenation" caused by the plant growth regulators included in the medium. To evaluate the effects of rejuvenation by tissue culture in Prunus sp., the expression patterns of the aging gene pathway described in plant models, which include the highly conserved microRNA (miRNA or miR) 156/157 and 172... |
