Asymbiotic culture of Cattleya intermedia Graham (Orchidaceae): the influence of macronutrient salts and sucrose concentrations on survival and development of plantlets 39

ABSTRACTCattleya intermediais an Atlantic Forest species endemic to Brazil that is classed as vulnerable on the list of threatened species. In this study, C. intermedia plantlets were micropropagated in an asymbiotic culture and the influence of different concentrations of sucrose (15, 30, 45 and 60 g L-1, plus a zero sucrose medium) and macronutrient salts (complete Murashige and Skoog (MS) medium and half MS medium (with half-strength macronutrients)) on survival and development of the plantlets was evaluated. Ultra-low temperature conservation of Brazilian Pine embryogenic cultures 42

This study aimed to establish a cryopreservation protocol for embryogenic cultures of A. angustifolia, enabling the ex situ conservation of the species. Embryogenic cultures were established from immature seeds and treated with variations of the cryoprotectant solutions SuDG, SoD and PVS2 prior to immersion in liquid nitrogen. Cell viability was evaluated after 30, 60 and 90 days of re-growth. The highest re-growth without morphological alterations and with normal biochemical composition was obtained with the PVS2 solution with 40 min immersion in ethanol (-20 °C). This procedure opens new horizons for the ex situ conservation of the species genetic.

Ovaries were collected from adult cows and fragments of ovarian cortex were immediately fixed (non-cultured control) or cultured in vitro in α-MEM+ alone or containing 10, 50, 100 or 1,000ng/mL rbST. Optimization of a protocol for direct organogenesis of red clover (Trifolium pratense L.) meristems for breeding purposes 48

A series of experiments were carried out in order to optimize a protocol for the direct organogenesis of Chilean red clover germplasm. A range of cultivars were used to analyze the effect of explant source (crown or stem meristems of vegetative plants), culture media and plant growth regulators. Our findings showed that stem meristems were easier to obtain, presented lower levels of contamination and a better development than crown meristems. The L2 medium showed better results than B5 and MS media for the cultivars and experimental lines studied. Micropropagation studies were carried out using the seeds for establishing an in vitro culture of Vriesea gigantea and Vriesea philippocoburgii. Germination rate of V. gigantea was higher than of V. philippocoburgii. Plantlets of V. philippocoburgii gave rise to many adventitious shoots when cultivated in Knudson basal medium. In contrast, for V. gigantea, a higher salts-concentration was needed, so that the number of shoots was increased by Murashige and Skoog medium. This study aimed at applying the generalized linear models (GLM) for the analysis of a germination experiment of Cattleya bicolor in which the response variable was binary. The purpose of this experiment was to assess the effects of the storage temperatures and culture mediums on the seed viability. The analyses of variance was also carried out either with or without the data transformation. All the statistical approaches indicated the importance of the storage temperature on the seed viability. The aim of this study was to establish an efficient protocol for the in vitro propagation of the endangered, endemic and decorative species Dianthus pinifolius Sibth. et Sm. The effects of different concentrations of 6-benzylaminopurine (BAP) and naphtalenacetic acid (NAA), and different explant types (single-node cuttings, terminal buds and shoot cuttings) on shoot multiplication were examined on Murashige and Skoog (MS) and half-strength MS media. A Brazilian isolate of Anaplasma marginale with appendage was successfully established and maintained in vitro in a tick cell line (IDE8). Infection was confirmed by optical and transmission electron microscopy. In addition, primers MSP1aNF2 and MSP1aNR2 amplified products from DNA extracted from infected IDE8 cells. Comparisons with partial sequences of the msp1α gene and the complete genome of A. marginale confirmed that the sequences of amplified fragments were from the A. marginale genome. This is the first establishment of a Brazilian A. marginale isolate in tick cells, representing a new system for biological and molecular studies and also a new source of material for diagnosis and development of vaccines.
