Effect of raffinose and ultrasound pulses on invertase release by free and immobilized Saccharomyces cerevisiae in loofa (Luffa cylindrica) sponge BABT
Marques,Leila Larisa Medeiros; Buzato,João Batista; Celligoi,Maria Antonia Pedrine Colabone.
This study investigated the effect of raffinose and ultrasound pulses on invertase release from free S. cerevisiae and S. cerevisiae immobilized in Luffa cylindrica. The free cell culture was submitted to 2% raffinose pulse and irradiated for 2 minutes at 0.12 and 0.46 h-1 dilution rates. The immobilized cell culture was submitted to raffinose pulse and irradiated for 1, 2 and 4 minutes, at 0.10 h-1 dilution rate. In immobilized cells, the raffinose pulse increased the invertase activity from 5.38 to 7.27 U/mg. Ultrasound application in free cell culture at the 0.12 h-1 dilution rate gave the best results. The activity varied from 25.08 to 29.38 U/mg while the increase in immobilized cells was from 5.22 to 9.70 U/mg when sonicated for two minutes. These...

Production of thermostable invertases by Aspergillus caespitosus under submerged or solid state fermentation using agroindustrial residues as carbon source BJM
Alegre,Ana Cláudia Paiva; Polizeli,Maria de Lourdes Teixeira de Moraes; Terenzi,Héctor Francisco; Jorge,João Atílio; Guimarães,Luis Henrique Souza.
The filamentous fungus Aspergillus caespitosus was a good producer of intracellular and extracellular invertases under submerged (SbmF) or solid-state fermentation (SSF), using agroindustrial residues, such as wheat bran, as carbon source. The production of extracellular enzyme under SSF at 30ºC, for 72h, was enhanced using SR salt solution (1:1, w/v) to humidify the substrate. The extracellular activity under SSF using wheat bran was around 5.5-fold higher than that obtained in SbmF (Khanna medium) with the same carbon source. However, the production of enzyme with wheat bran plus oat meal was 2.2-fold higher than wheat bran isolated. The enzymatic production was affected by supplementation with nitrogen and phosphate sources. The addition of glucose in...

Synthesis of sorbitol by Zymomonas mobilis under high osmotic pressure BJM
Barros,Márcio de; Celligoi,Maria Antonia Pedrine Colabone.
The bacterium Zymomonas mobilis presents potential for sorbitol production when grown in culture medium with high sugar concentration. Sorbitol is produced and accumulated in the periplasma of the bacterium to protect the cells from the harmful effects of high osmotic pressure that results from the action of invertase on sucrose. The conversion of sucrose into glucose and fructose increases the osmolarity of the medium. However, an excessive increase in the osmotic pressure may decrease the sorbitol production. In this work Saccharomyces cerevisiae invertase was added two media containing sucrose 200 and 300 g.L-1. Sorbitol production in sucrose at 200 g.L-1 was 42.35 and 38.42 g.L-1, with and without the invertase treatment, respectively. In the culture...

Trehalose accumulation, invertase activity and physiological characteristics of yeasts isolated from 24 h fermentative cycles during the production of artisanal Brazilian cachaça BJM
Pataro,Carla; Guerra,Juliana B.; Gomes,Fátima C.O.; Neves,Maria J.; Pimentel,Patrícia F.; Rosa,Carlos A..
Trehalose accumulation, invertase activity and physiological characteristics of 86 yeast isolates from short fermentative cycles during the production of cachaça in three artisanal distilleries of the State of Minas Gerais were studied. Among these isolates, 70% were able to grow at temperatures between 40 and 42ºC. Only Saccharomyces cerevisiae isolates were able to grow over 40ºC. Lower temperatures (<40ºC) favoured the growth of yeasts such as Candida parapsilosis-like, C. maltosa-like, Kloeckera japonica, S. exiguus and C. bombicola-like. The isolates from all three distilleries were ethanol tolerant, produced invertase, and accumulate trehalose in the presence of glucose. The strains isolated from distillery A presented more resistance to ethanol...

Sucrose hydrolysis by invertase using a membrane reactor: effect of membrane cut-off on enzyme performance BJPS
Addezio,Francesco Di; Yoriyaz,Ester Junko; Cantarella,Maria; Vitolo,Michele.
Sucrose hydrolysis by invertase [EC.3.2.1.26] produces inverted sugar syrup, an ingredient mainly used in the food industry. To properly catalyze hydrolysis, the enzyme should be reused after this reaction. It is advisable to maintain constant activity over a considerable period. Thus, sucrose hydrolysis was performed in a membrane bioreactor - a continuously stirred tank reactor coupled with an ultrafiltration membrane (UFM) which provides good diffusion and high activity per unit volume. Molecular weight cut-off for soluble invertase UFMs was up to 100kDa. This study focused on the role of UFM invertase cut-off as it is the main element in the process. We demonstrated that both the cut-off and chemical nature of the UFM affected specific invertase...

Invertase, glucose oxidase and catalase for converting sucrose to fructose and gluconic acid through batch and membrane-continuous reactors BJPS
Silva,Aline Ramos da; Tomotani,Ester Junko; Vitolo,Michele.
Conversion of sucrose into fructose and gluconic acid using invertase, glucose oxidase and catalase was studied by discontinuous (sequential or simultaneous addition of the enzymes) and continuous (simultaneous addition of the enzymes in a 100 kDa-ultrafiltration membrane reactor) processes. The following parameters were varied: concentration of enzymes, initial concentration of substrates (sucrose and glucose), pH, temperature and feeding rate (for continuous process). The highest yield of conversion (100%) was attained through the discontinuous (batch) process carried out at pH 4.5 and 37 ºC by the sequential addition of invertase (14.3 U), glucose oxidase (10,000 U) and catalase (59,000 U).

Anatomical characteristics and enzymes of the sucrose metabolism and their relationship with latex yield in the rubber tree (Hevea brasiliensis Muell. Arg.) Braz. J. Plant Physiol.
Mesquita,Alessandro Carlos; Oliveira,Luiz Edson Mota de; Mazzafera,Paulo; Delú-Filho,Nelson.
Rubber production is dependent on the availability of carbohydrates in the tissues engaged in latex synthesis. In this study three rubber tree clones were used to study the protein profile, the activities of invertase and sucrose synthase in the latex and anatomical characteristics of the bark, as well as to establish a possible relationship with latex production. The clones used in the study, RRIM 600, GT 1 and Fx2261, differed in latex productivity, from high to low in this order. Although present, few differences were observed in the latex protein profile obtained by denaturing electrophoresis. The activity of neutral invertase was higher than the acidic isoform, and both activities were higher in the most productive clones RRIM 600 and GT 1. The same...

Sucrose hydrolysis catalyzed by auto-immobilized invertase into intact cells of Cladosporium cladosporioides Electron. J. Biotechnol.
Santana de Almeida,Ana Cláudia; Costa de Araújo,Luciares; Mendes Costa,Andressa; Moraes de Abreu,César Augusto; Gomes de Andrade Lima,Maria Alice; Perez Fernandez Palha,Maria de Los Angeles.
The enzyme known as invertase (E.C. 3.2.1.26 - β-D-fructofuranosidase) catalyzes the sucrose hydrolysis producing an equimolar mixture of glucose and fructose named inverted sugar. The fungus Cladosporium cladosporioides has invertase as its constituent. Hence, its use as a natural immobilized support for the invertase produces interesting results for the enzyme. The present work has the objective of determining the optimum operational conditions of auto-immobilized invertase, as well as its kinetic parameters (K M and Vmax). A complete 2³ factorial planning was done for the evaluation of such parameters. Temperature, pH and agitation level were the studied variables. The hydrolysis percentage was the monitored result. Batch tests in optimum...

Characterization of an Alternative Baking Strain of Saccharomyces cerevisiae Isolated from Fermented Cherry Fruits by the Analysis of SUC2 Gene OAK
ODA, Yuji; MIKUMO, Dai; TAJIMA, Kanako; YAMAUCHI, Hiroaki; 小田, 有二.
The yeast strain AK 46 was isolated from dough containing fermented cherry fruits and identified as Saccharomyces cerevisiae. The nucleotide sequence of the rDNA spacer region from this strain coincided with those from the yeasts used for the production of whisky, wine, and bread. In a comparison with six baking strains, strain AK 46 possessed less leavening ability in dough with and without addition of 5% sucrose (based on the weight of flour) but a high ability in sweet dough containing 30% sucrose. The activities of the two enzymes α-glucosidase and invertase, which are related to dough fermentation, were lower than those in the other strains. A baking test showed that strain AK 46 was applicable for breadmaking using the straight dough and sponge dough...
