CELL CULTURE ISOLATION OF HEPATITIS E VIRUS GENOTYPE 3 STRAIN OBTAINED FROM HUMAN FECES Acta biol.Colomb.
QUINTERO-GIL,Carolina; MIRAZO,Santiago; PARRA-SUESCÚN,Jaime; LÓPEZ-HERRERA,Albeiro; MAINARDI,Victoria; ARBIZA,Juan; ORDÚZ,Sergio.
ABSTRACT Hepatitis E virus (HEV) is considered one of the leading causes of acute viral hepatitis worldwide, and about 20 million infections and approximately 57 000 deaths occurred every year. However, little is known about the replicative virus cycle due to the absence of a consensus cell culture model. A549 cell line is considered susceptible to HEV genotype 3, however, both viral strain and cell culture conditions could affect the viral isolation in vitro. The objective of this work was to isolate in vitro an HEV-3 strain obtained from human feces. To this, a genotype 3 HEV strain previously identified by genetic characterization was inoculated in A549 monolayers, and incubated for two hours at 37 °C. Five days post-infection, cells were passaged...

ISOLATION OF THE HUMAN CYTOMEGALOVIRUS FROM BODILY FLUIDS Acta biol.Colomb.
CAMACHO ORTEGA,Sigrid Johanna; BOHORQUEZ ÁVILA,Sonia del Pilar; VELANDIA ROMERO,Myriam Lucia; CASTELLANOS PARRA,Jaime Eduardo.
ABSTRACT In vitro studies on the pathogenesis of the human cytomegalovirus (HCMV) are conducted regularly using laboratory adapted strains that lose some characteristics during the adaptation process. Since HCMV is excreted from bodily fluids during infection or reactivation, this work aimed to isolate and culture HCMV from the MRC-5 human cells found in the urine, bronchoalveolar lavage, saliva, and plasma samples of pediatric patients with probable or confirmed infection. The samples were inoculated on cell cultures either for 14 days or until a cytopathic effect (CPE) of 80 % was observed. The cell lysates and supernatants were used to perform successive viral passages. Besides HCMV, the herpes simplex virus was detected from all the saliva samples....

Domingues,S.F.S.; Ferreira,H.S.; Muniz,J.A.P.C.; Lima,A.K.F.; Ohashi,O.M.; Figueiredo,J.R.; Silva,L.D.M..
The aim of this study was to adapt a mechanical procedure for the isolation of intact preantral follicles from Cebus apella ovaries. The interval effect of serial sections of the tissue chopper was tested on a number of preantral follicles isolated from ovaries (n=6) of three C. apella females, two prepubertal and one adult. Ovaries were divided into four equal parts and fragmented with a tissue chopper, adjusted for serial sections at intervals of 250, 500, 750 and 1,000µm, respectively. Isolated follicles were counted in a Neubauer's chamber and classified as primordial, primary or secondary. The number (mean±SE) of preantral follicles isolated from 1/4 ovary varied from 68,330+17,590 (at the 1,000µm cut interval) to 300,830+111,460 (at the 500µm cut...

Isolation, Culturing, Characterization and Aging of Adipose Tissue-derived Mesenchymal Stem Cells: A Brief Overview BABT
Fathi,Ezzatollah; Farahzadi,Raheleh.
ABSTRACT The aim of this review was to describe the current state-of-the-art regarding isolation, characterization and aging of adipose tissue-derived mesenchymal stem cells (ADSCs). Mesenchymal stem cells (MSCs) have recently received widespread attention because of their potential use in tissue-engineering applications. Various studies have indicated that MSCs with a fibroblast-like morphology migrate to the sites of injury and help to regenerate damaged tissue. Over the past few years, it has been recognized that fat is not only an energy supply, but also a rich source of multipotent stem cells that can be easily harvested, isolated and selected as compared with other tissues. ADSCs are particularly interesting because of their rapid proliferation and...

Primary cultures of mouse small intestinal epithelial cells using the dissociating enzyme type I collagenase and hyaluronidase BJMBR
Ren,H.J.; Zhang,C.L.; Liu,R.D.; Li,N.; Li,X.G.; Xue,H.K.; Guo,Y.; Wang,Z.Q.; Cui,J.; Ming,L..
The epithelium is a highly dynamic system, which plays a crucial role in the homeostasis of the intestinal tract. However, studies on the physiological and pathophysiological functions of intestinal epithelial cells (IECs) have been hampered due to lack of normal epithelial cell models. In the present study, we established a reproducible method for primary culture of mouse IECs, which were isolated from the viable small intestinal crypts of murine fetuses (on embryonic day 19), using type I collagenase and hyaluronidase in a short span of time (≤20 min). With this method, continuously growing mouse IECs, which can be subcultured over a number of passages, were obtained. The obtained cell lines formed a tight cobblestone-like arrangement, displayed long and...

Social condition affects hormone secretion and exploratory behavior in rats BJMBR
Genaro,G.; Schmidek,W.R.; Franci,C.R..
Studies of behavior, endocrinology and physiology have described experiments in which animals housed in groups or in isolation were normally tested individually. The isolation of the animal from its group for testing is perhaps the most common situation used today in experimental procedures, i.e., there is no consideration of the acute stress which occurs when the animal is submitted to a situation different from that it is normally accustomed to, i.e., group living. In the present study, we used 90 male 120-day-old rats (Rattus norvegicus) divided into 5 groups of 18 animals, which were housed 3 per cage, in a total of 6 cages. The animals were tested individually or with their groups for exploratory behavior. Hormones were determined by radioimmunoassay...

Isolation, properties and kinetics of growth of a thermophilic Bacillus BJM
Souza,Adriane Nunes de; Martins,Meire Lelis Leal.
A strictly aerobic, thermophilic, spore-forming bacteria was isolated from a soil sample collected in Campos dos Goytacazes City, Rio de Janeiro, Brazil. The cells of this organism were Gram-positive, catalase positive, actively motile, 1.2 µm wide and 5.3 µm long. Growth occurred at pH values ranging from 6.5 to 9.0, and optimum growth occurred at about pH 7.0. The optimum temperature for growth was around 55ºC, and the upper temperature limit for growth was around 70ºC. Yeast extract stimulated growth on glucose and was obligately required at supraoptimal temperatures (65ºC). The results of 16S rRNA sequence comparisons indicated that the isolate was closely related to Bacillus caldoxylolyticus and Bacillus sp strain AK1, and these three organisms...

Antimicrobial resistance and virulence gene profiles in P. multocida strains isolated from cats BJM
Ferreira,Thais Sebastiana Porfida; Felizardo,Maria Roberta; Gobbi,Debora Dirani Sena de; Moreno,Marina; Moreno,Andrea Micke.
Cats are often described as carriers of Pasteurella multocida in their oral microbiota. This agent is thought to cause pneumonia, conjunctivitis, rhinitis, gingivostomatitis, abscess and osteonecrosis in cats. Human infection with P. multocida has been described in several cases affecting cat owners or after cat bites. In Brazil, the cat population is approximately 21 million animals and is increasing, but there are no studies of the presence of P. multocida in the feline population or of human cases of infection associated with cats. In this study, one hundred and ninety-one healthy cats from owners and shelters in São Paulo State, Brazil, were evaluated for the presence of P. multocida in their oral cavities. Twenty animals were positive for P....

Curtobacterium flaccumfaciens pv. flaccumfaciens detection in bean seeds using a semi-selective culture medium BJM
Maringoni,Antonio Carlos; Camara,Renata de Cassia.
The bacterial wilt caused by Curtobacterium flaccumfaciens pv. flaccumfaciens is currently considered one of the most important bacterial bean disease in Brazil. One of the most effective control methods against this disease is the use of healthy seeds. However, no methods are known that could be routinely used to detect this bacterium in bean seeds under Brazilian condition. The aim of this work was to evaluate qualitative and quantitative detection methods for Curtobacterium flaccumfaciens pv. flaccumfaciens in naturally-infected bean seeds, and the detection of this pathogen in thirty bean seed samples, by sowing onto a semi-selective culture medium the leachate obtained from soaked bean seeds. Both the qualitative and quantitative methods were...

Arcobacter butzleri: first isolation report from chicken carcasses in costa rica BJM
Arias,Maria Laura; Cid,Adriana; Fernandéz,Heriberto.
Arcobacter butzleri isolation from chicken carcasses in Costa Rica is reported for the first time. The isolated strains (P and R) were presumptively identified by their phenotypic characteristics. Definitive identification was made using a multiplex PCR assay for the simultaneous detection and identification of Arcobacter butzleri, Arcobacter cryaerophilus and Arcobacter skirrowii. These first isolations indicate the necessity of further investigation about the prevalence, distribution, ecology and interactions with human beings of this and other Arcobacter species.

Puentes,R; Eliopulos,N; Pérez,R; Franco,G; Sosa,K; Bianchi,P; Furtado,A; Hübner,S.O.; Esteves,P.A..
Canine parvovirus type 2 (CPV-2) is a leading cause of diarrhea in puppies in several parts of the world. In this study CPV-2 was detected and recovered from puppies showing clinical disease from Montevideo, Uruguay. Samples were processed and used to infect CRFK and MDCK cells in order to isolate the virus. Out of twelve, two samples were positive for CPV-2. A genomic region of 583 bp was amplified and the molecular characterization was performed by sequencing, phylogenetic analysis and Restriction Fragment Length Polymorphism (RFLP). Two isolated viruses (UY1 and UY2) were CPV-2c-like viruses. The comparison between the cytophatic effect (CPE) of CPV-2 (vaccinal virus) and CPV-2c (isolated virus) on primary canine cells cultures and on CRFK line cells,...

Isolation of bovine coronavirus (BCoV) in monolayers of HmLu-1 cells BJM
Jerez,José Antonio; Gregori,Fábio; Brandão,Paulo Eduardo; Rodriguez,Cesar Alejandro Rosales; Ito,Fumio Honma; Buzinaro,Maria da Glória; Sakai,Takeo.
Isolation of BCoV was performed in monolayers of HmLu-1 cells, using 20 fecal samples from clinical cases of diarrhea in calves. Samples were positive for BCoV by means of hemagglutination / hemagglutination inhibition (HA/HI). Up to the fifth serial passage, 13 of these isolates presented syncytial cytopathic effect, similar to Kakegawa standard strain. When isolates were submitted to seroneutralization with gammaglobulin anti-bovine coronavirus, 8 of them were considered to be positive, once cytopathic effect was neutralized. After titration and seroneutralization in microplates, only three of them were confirmed as positive. The lineage HmLu-1 showed higher permissivity to BCoV isolation, but the low intensity of viral replication demonstrated that new...

Comparison of methods for mycobacteria isolation from swine feces BJM
Oliveira,Eugenia Márcia de Deus; Rodriguez,César Alejandro Rosales; Rocha,Vivianne Cambuí Mesquita; Ambrosio,Simone Rodriguez; Ohara,Patrícia Miyuki; Amaku,Marcos; Ferreira,Fernando; Dias,Ricardo Augusto; Leão,Sylvia Cardoso; Ferreira Neto,José Soares.
Swine mycobacteriosis is an important cause of carcass condemnation at abattoirs. One of the best ways to recognize the etiologic agent involved, in live animals, is the fecal isolation, as 94% of the lesions are located in the digestive tract. Therefore, the goal of the present study was to compare the performance of four decontamination methods followed by inoculation in three different culture media, totalizing twelve procedures of mycobacteria search from swine fecal samples experimentally contaminated. The swine feces were artificially contaminated with 0.02 g of Mycobacterium avium, PIG-B strain, and subjected to mycobacteria isolation trial. The protocols used were: 1) modified Petroff or basic method; 2) modified Lowenstein-Jensen or acidic method;...
