El objetivo del presente estudio fue obtener un cultivo puro de la bacteria ruminal Streptococcus bovis a partir de líquido ruminal de ovinos alimentados con una dieta alta en sorgo (70 % en base seca). Además, se analizó su actividad fermentativa (producción de AGV y lactato) y el proceso de liofilizado como método de conservación, y se realizaron ensayos para expresar su bacteriófago. Para el aislamiento se usó un medio comercial para Streptococcus spp. (Merck) preparado bajo anaerobiosis. Para la identificación se usó el sistema API 20 Strep, reacción catalasa, morfología celular y tinción de Gram. The aims were to explore an appropriate isolating medium for obtaining purple nonsulfur bacteria (PNSB) for use as biofertilizers in saline paddy fields and to obtain pure cultures. We therefore chose a defined isolating medium containing 0.25% NaCl, (Glutamate-Acetate broth, GA) and a rice straw broth to compare them for numbers of PNSB obtained, time to obtain pure cultures, diversity and costs. Endophytic bacteria from three arboreal species native to the Amazon (Carapa guianenses, Ceiba pentandra, and Swietenia macrophylla), were isolated and identified, through partial sequencing of the 16S rRNA encoding gene. From these, 16 isolates were obtained, although, when compared to sequences deposited in GenBank, only seven had produced identifiable fragments. Bacillus, Pantoea and two non-culturable samples were identified. Results obtained through sequence analysis revealed low genetic diversity across the isolates, even when analyzing different species and plant structures. This is the first report concerning the isolation and identification of endophytic bacteria in these plant species. |

Since the invention of the Petri dish, there have been continuous efforts to improve efficiency in microbial cultivation. These efforts were devoted to the attainment for diverse growth conditions, simulation of in situ conditions and achievement of high-throughput rates. As a result, prokaryotes catalysing novel redox reactions as well as representatives of abundant, but not-yet cultured taxa, were isolated. This work reports the first isolation and characterization of an alkaline phosphatase (AP) from a hyperthermophilic archaeon. An AP gene from Pyrococcus abyssi, a euryarchaeon isolated from a deep-sea hydrothermal vent, was cloned and the enzyme expressed in Escherichia coli. Analysis of the sequence showed conservation of the active site and structural elements of the E. coli AP. The recombinant AP was purified and characterized. Monomeric and homodimeric active forms were detected, with a monomer molecular mass of 54 kDa. Apparent optimum pH and temperature were estimated at 11.0 and 70 degreesC, respectively. Two banana supply chains (BSC), one in Indonesia (BSC1) and one in Australia (BSC2), are described based on case study research conducted in 2002. BSC1 sources bananas from small farmers for distribution through traditional markets in Jakarta. BSC2 includes farmers supplying to both major and independent retailers in Sydney through a wholesale agency with some farmer ownership. Reservoir competence for the Newcastle Disease virus (NDV) was evaluated in sparrows (Passer domesticus, Linnaeus 1758) captured on a commercial poultry farm and a chicken hatchery in the State of Pernambuco, Northeastern Brazil. A total number of 103 birds collected from a poultry farm (24/103) and a chicken hatchery (79/103) were examined. Hemagglutination inhibition tests, isolation, and viral characterization were performed in all samples collected from each bird. Titers ranging from 1:2 to 1:64 were detectable in 10.68% of sparrows, but positive serology and viral isolation were obtained only from sparrows captured at the hatchery. The hygienic status of raw chicken meat marketed at Ibague, Tolima, Colombia, is currently unknown. To address this issue, a cross-sectional study was conducted to estimate the prevalence of Salmonella spp., in raw chicken marketed at different outlets in this city.
