The goal of this research was to find the frequency and spatial distribution of infection caused by Leptospira interrogans in equidae in Minas Gerais State from September 2003 to March 2004. Samples of blood serum (6,475) were analyzed by microscopic agglutination test. From the total, 381 samples were positive (5.9 %), with title equal or superior to 1:200 for one or more serovars of leptospira. The most frequent serovars were Hardjo (Norma), Pomona, Bratislava, and Batavie. The higher frequency of equidae reagents were recorded at the North and Northeast region of Minas Gerais, followed by Triângulo Mineiro and Alto Paranaíba, then Central, West, Metropolitan area of Belo Horizonte, and South/Southwest. |

ABSTRACT Bovine digital dermatitis (BDD) is an infectious and contagious disease characterized by ulcerative and proliferative lesions affecting the skin on the bulbs of the heel or the interdigital cleft in dairy cattle, often associated with lameness. Evidences on the etiology of BDD indicate that it is multifactorial, involving environmental factors and multiple bacterial colonization. Leptospirosis caused by Leptospira interrogans is the most widespread zoonosis and a major public health problem worldwide. Based on light-scattering and absorption, quantification of leptospires using UV-VIS spectroscopy was used as an indirect counting technique by measuring the optical density and comparing this to automated direct counting using a counting chamber in combination with imaging and analyzing software. Two serovars, Bangkok and Copenhagenii, from log-phase growth were used for the establishment of standard curves. They were found to be linear and slightly different in gradient for each serovar. Baypamun® is an immunity modulator recommended as prophylactic and therapeutic use. In the present study, this product was used in the hamsters experimentally infected with Leptospira interrogans serogroup canicola. The animals were divided in 5 groups, with 20 animals each. This product was used as a therapeutic, prophylactic and as adjuvant. As a therapeutic use all the animals died. Despite of eight (40.0%) survivors, the prophylactic use wasn't also totally favorable due to leptospire recovering from kidneys of those animals. Based on the analysis of antibodies levels among animals that received vaccine and vaccine with Baypamun® as adjuvante, this product didn't conferred superior humoral answer. Genes encoding lipoproteins LipL32, LipL41 and the outer-membrane protein OmpL1 of leptospira were recombined and cloned into a pVAX1 plasmid. BALB/c mice were immunized with LipL32 and recombined LipL32-41-OmpL1 using DNA-DNA, DNA-protein and protein-protein strategies, respectively. Two Leptospira sp. isolates were obtained by the first time from goats in Brazil and characterized by sequencing rrs, rpoB and secY genes, PFGE and typing with monoclonal antibodies. Both isolates are identical and belong to Leptospira santarosai. Analysis of the rrs and the rpoB genes sequences revealed 100% identity between the goat isolates and the Bananal reference strain. When secY sequences of the two isolates were compared to each other, it was observed that they had identical sequences. Swine leptospirosis is recognized world wide as an important cause of reproductive failure. Serum and histo-pathological examinations have been constantly used to diagnose this disease. This study was carried out on slaughtered sows from Northern Paraná State to compare serum findings with kidney, liver, ovary and uterus histo-pathological examinations. The microscopic serum-agglutination carried out on 36 animals detected 24 with titles > 100 and 12 negative (< 100). Pathogenic Leptospira spp. are the etiological agents of leptospirosis, an important disease of both humans and animals. In urban settings, L. interrogans serovars are the predominant cause of disease in humans. The purpose of this study was to characterize a novel Leptospira isolate recovered from an abandoned swimming pool. Leptospires are usually classified by methods based on DNA-DNA hybridization and the conventional cross-agglutination absorption test, which uses polyclonal antibodies against lipopolysaccharides. In this study, the amplification of the rpoB gene, which encodes the beta-subunit of RNA polymerase, was used as an alternative tool to identify Leptospira. In leptospirosis, the Microscopic Agglutination Test (MAT) is the basis of serological diagnosis and the most widely used. In our study, the MAT was applied to evaluate the influence of temperature, incubation time, antigen density and age of the culture in the agglutinability of Leptospires and the interference of these factors in diagnosis. Three serum samples with titers of 100, 800 and 25.600 to the serovar tande (local isolate) of canicola serogroup were used with different combinations among the four factors. Foram estudadas aglutininas antileptospíricas em 403 amostras de soro de búfalos, provenientes de sete Municípios do Vale do Ribeira, Estado de São Paulo, coletadas no período de janeiro de 1992 a junho de 1993. Utilizou-se o teste de soroaglutinação microscópica, considerando-se positivas as amostras cujo título fosse igual ou superior a 100. O maior título encontrado foi 1600 para o sorovar bratislava (1 amostra), seguido de 800 para wolffi (4 amostras). Equine leptospirosis can present a non-symptomatic form, an acute clinical form, or even develop chronically, causing reproductive alterations, such as abortion and recurrent uveitis. Since the prevalence of leptospirosis in several countries and regions is widely reported, the objective of this study was to verify the prevailing equine leptospirosis in different regions of Brazil.
