This work studied the effect of the lectin phytohaemagglutinin (PHA) and bacterial lipopolysaccharide (LPS) on the activity of nonspecific cytotoxic cells (NCCs) and macrophages of peripheral blood in the rainbow trout. NCCs are cells of the innate immunity, homologues to Natural Killer (NK) cells of mammals, capable of eliminating cells infected with intracellular pathogens and also tumoral cells. Their lytic activity is mediated by the perforin/granzyme system and the Fas receptor-Fas ligand (FasR-FasL) system. Activity of NCCs was evaluated by their capacity to lyse HL-60 tumor cells. On the other hand, macrophages eliminate microorganisms by several mechanisms, including the production of reactive oxygen species (ROS) and nitric oxide (NO). Results... |

The defence against infections is the essential function of the immune system of all Vertebrates. Fish present an efficient and developed immunologic response and in the case of Teleosts, with some similarities to the higher vertebrates. It can be divided into two types: innate or non specific and adquired or specific, both with a humoral and cellular compartments. Their relative importance can change according to the age of fish and is always under the influence of different factors. The most relevant components are mentioned here. There are two factors in fishes which strongly influence the immune response and antibody production, seasonal changes and temperature. The main lymphoid organs are described here: the thymus, the kidney and the spleen, as well... |

Arnoglossus laterna is only infested by intestinal parasites. Each parasite distribution is independent of the distribution of other parasites and also independent of the sex of host. When a fish is infested by a great number of the two helminths Acanthocephaloides and Bothriocephalus clavibothrium , the proportion of lymphocytes tends to decrease and the proportion of neutrophils tends to increase (inflammatory response). Important infestation by Scolex pleuronectis induces an increase of the thrombocyte count for 100 counted leucocytes. After infestation by the nematode Thynnascaris sp. alone or with another parasite, even by small number, the proportion of lymphocytes decreases and the proportion of neutrophils increases. |

Leukocyte-endothelial cell interactions have a pivotal role in immune responses. In our study Ten adult wistar albino rats weighing 180-200 g were used to obtain tissue samples. The animals were sacrificed by decapitation under ether anesthesia. The lymph nodes of the animals were then quickly removed. Specimens were immersed in the zinc iodide-osmium tetroxide (ZIO) solutation by Niebauer et al. (1969) and kept in the dark for 2 hours in this solution from temperature for fixation/staining. Samples were processed according to routine plastic embedding tecnique. Semithin sections of 1µm thick were cut and stained with methilene blue-azure II for light microscopic examination. We used zinc iodide-osmium tetroxide staining tecnique to distinguish endothelial... |

Purification of a lectin from Bothrops jararacussu venom (BjcuL) was carried out using agarose-D-galactose affinity gel. MALDI-TOF gave a major signal at m/z 32028, suggesting the presence of a dimmer composed of two identical subunits. Divalent cations were required for the lectin activity, as complete absence of such ions reduced hemagglutination. BjcuL was more effective at neutral pH and showed total loss of activity at pH values below 4.0 and above 9.0. Its agglutinating activity remained stable at 25°C until 60min, but increased when at 35°C for at least 15min. Adhesion assays to extracellular matrix (ECM) glycoproteins showed that the biotinylated lectin (0.039-5.0µg/100µl) was capable of binding to fibronectin and vitronectin in a dose-dependent... |

Farming of native carnivore fish species has drawn attention due to their promising use in aquaculture. Among these species, tucunaré of the genus Cichla stand out, them being of high economical interest for sport fishing and Amazon's industry of ornamental fish. The present study describes the weight-length relationship (WLR), relative condition factor (Kn), red blood cell parameters, thrombocytes and leukocytes count of Cichla temensis Humboldt, 1821, farmed in central Amazon. Fish that underwent food training during fingerling culture received extruded ration containing 45% of crude protein during fattening, and had Kn with values from 0.925-1.199, which indicated good health condition during the culture. The equation obtained from the WLR was W =... |

The objective of this work was to characterize the hematological profiles of steers and bulls, according to residual feed intake (RFI), after feed deprivation. Twenty‑month‑old Nellore steers and bulls were fed feedlot diets for 70 and 56 days, respectively. RFI was calculated as the difference between actual feed intake and expected feed intake. More and less efficient steers and bulls, according to RFI, were subjected to 24 hours of food deprivation. Blood was sampled prior to and following the withdrawal period. Hematological analyses included total and differential white blood cell count, red blood cell count and morphology, and plasma glucose concentration. Variation in RFI did not influence plasma glucose concentration or blood cell profiles. Glucose... |

ABSTRACT In this study, we evaluated the effects of substitution of partial and total spray-dried blood plasma (BP) with soy protein concentrate (SPC) in the diet of post-weaning piglets on their performance, intestinal histomorphometry, and blood parameters. Fifty-four castrated male piglets, weaned at an average age of 21 days and an initial average weight of 7.01±0.294 kg, were distributed in a completely randomized block design, with three diets (0%SPC-5%BP, 2.5%SPC-2.5%BP, and 5%SPC-0%BP), six replicates, and three piglets per experimental unit. The experimental feeds were formulated to meet the requirements of the piglets at 21-32 and 33-42 days of age (pre-starter diet) and 43-66 days of age (starter diet). From 21 to 32 and 21 to 42 days of age,... |

The aim of this paper is to compare three different methods for counting white blood cells [WBC] (Natt and Herrick method, estimation with 1,000 and 2,000 erythrocytes) and three methods for counting total thrombocytes [TT] (Wojtaszek method, estimation with 1,000 and 2,000 erythrocytes) in a South American freshwater turtle species, Podocnemis expansa, Schweigger 1812 (Reptilia, Pelomedusidae). Direct WBC counts using the Natt and Herrick method showed limitations, which are discussed here. The WBC and TT counts using 1,000 erythrocytes from blood smears are not recommended for Amazon turtles nor other reptilian species, since wide variation in counts can be observed. Estimation methods for determining WBC and TT based on 2,000 erythrocytes of blood... |

ABSTRACT The hematological and biochemical responses of pirarucu fingerlings (Arapaima gigas) fed with diets containing different concentrations of a glucomannan product derived from yeast and algae were evaluated in order to ascertain the effect of these diets on fish physiology. Four treatments were conducted, with three replications, with 12 fish in each tank. The product evaluated (MycosorbA+(r)) was incorporated into the commercial diet, at four concentrations: 0, 1, 2 and 4 g.kg-1, called M0%, M0.1%, M0.2% and M0.4%, respectively. After 45 days of feeding, blood samples from six fish in each replicate were collected to perform the analyses. Their weight and length were determined to calculate the condition factor and weight gain, but no differences... |

The aim of this study was not only to determine the red blood cells parameters, thrombocyte and leukocyte counts in farmed Brycon amazonicus (matrinxã), to compare these parameters among Bryconinae species from literature, and also to investigate the presence of special granulocytic cells in these fish. The results of the blood cells parameters here established for farmed B. amazonicus, a species of great economic importance in Brazilian aquaculture, could help a better understanding of the blood features in natural populations of this Amazon species. Blood parameters varied between Bryconinae species investigated, mainly the red blood cell counts, hemoglobin, hematocrit and mean corpuscular volume (MCV). The presence of the blood granulocytes, neutrophils... |

Natural insecticides derived from plant extracts have been used as an alternative to synthetic products in order to reduce environmental contamination. The present study aimed to examine the effects of Fumydro®, a natural insecticide based in the tobacco plant Nicotiana tabacum, on the Nile tilapia (Oreochromis niloticus) by determining the 48-h LC50 and evaluating their effects on hematological variables. Adult specimens of O. niloticus were exposed to four Fumydro® concentrations (200, 300, 400 and 500 μL L-1). The 48-h LC50 of Fumydro® was determined as 370 ± 50 μL L-1. Surviving fish showed increasing in the red blood cells, hemoglobin concentration, mean corpuscular hemoglobin and mean corpuscular hemoglobin concentration. The thrombocytes did not... |

ABSTRACT Euphorbia tirucalli (L.), commonly known as aveloz, has been indiscriminately used in popular medicine to treat various illnesses. However, some components can have devastating consequences. Injury to a cell's genetic material can cause mutations, cancer, and cell death. Our main goal in this work was to evaluate the genotoxic and cytotoxic effects of E. tirucalli extract on human leukocytes. For this purpose, we performed a phytochemical analysis to evaluate the plant's components. In the second step, we treated cultured human leukocytes with different concentrations of the dry extract of the plant and then evaluated the oxidative and genotoxic profiles of these leukocytes. We found that at 1% and 10% concentrations, the aveloz extract acted as a... |

Abstract: The systemic effect of chronic periodontitis (CP) has been suggested by several studies as an etiologic factor and modulator of diseases based on the changes in the inflammatory marker levels. This study aimed to investigate the relationship between the changes in clinical periodontal outcomes and serum biomarkers (CRP, IL-6, albumin and percentage of leukocytes) after non-surgical periodontal therapy in systemically healthy adults. An interventional study was conducted with a sample of 29 individuals without CP (control group) and 33 with CP (CP group). Periodontal clinical variables were recorded, and the serum levels of inflammatory markers were measured. Statistical analysis included the chi-square and Student’s t-tests and Pearson’s... |

DNA damage repair was assessed in quiescent (G0) leukocytes and in hepatocytes of mice, after 1 and 2 hours recovery from a single whole body y-irradiation with 0.5, 1 or 2 Gy. Evaluation of single-strand breaks (SSB) and alkali-labile sites together were carried out by a single-cell electrophoresis at pH>13.0 (alkaline comet assay). In non-irradiated (control) mice, the constitutive, endogenous DNA damage (basal) was around 1.5 times higher in leukocytes than in hepatocytes. Irradiation immediately increased SSB frequency in both cell types, in a dose-dependent manner. Two sequential phases took place during the in vivo repair of the radio-induced DNA lesions. The earliest one, present in both hepatocytes and leukocytes, further increased the SSB... |
