Assessment of lipolytic activity of isolated microorganisms from the savannah of the Tocantins Biological Sciences

Current study assesses the biodiversity and selects lipase-producer microorganisms with industrial interest, from the savannah of the state of Tocantins, Brazil. Seventeen pequi microorganisms (Caryocar brasiliense) were isolated in the decomposition stage and 35 microorganisms were retrieved from the soil fraction under the collected pequi. Yarrowia lypolitica strain was used as positive control in all assays. The 52 strains were subjected to tests in a solid medium with Tween 20 for checking halos formed by crystals, indicating lipase production by inoculated strains. Analisou-se a atividade enzimática in vitro de uma lipase em diferentes condições de temperatura (simulando o processo de peletização) e pH (acidez gástrica) e na presença de íons minerais e metálicos. Para avaliar a estabilidade térmica, a enzima foi incubada a 80(0)C por quatro tempos (0, 5, 10 e 15 minutos). O efeito do pH foi estudado submetendo-se a enzima a quatro valores de pH (7,3, 2,0, 3,2 e 5,0). Para avaliar o efeito da lipase na presença de íons minerais e metálicos, incubou-se a enzima em diferentes soluções contendo cálcio, cobre, cobalto, manganês e ferro. Os objetivos deste trabalho foram determinar o efeito de três períodos de jejum (zero, 24 e 48 horas) entre o nascimento e o alojamento de pintos e da adição de óleo à ração sobre o crescimento alométrico dos órgãos digestivos e estudar a atividade da lipase no pâncreas e intestino delgado. Foram utilizados 720 pintos de corte machos, criados de um a 15 dias de alojamento. As avaliações do crescimento dos órgãos digestivos foram feitas nos dias zero, três, seis, nove, 12 e 15 de idade. Estimou-se a origem da atividade lipolítica do leite com contagens de células somáticas (CCS) ajustadas para diferentes níveis. A partir de lotes de leite cru com baixa (100.000 cel/ml) e alta (1.000.000 cel/ml) CCS, formaram-se três tratamentos: A) leite com baixa CCS; B) leite com alta CCS; C) leite originalmente com baixa CCS e com adição de células somáticas. Endophytic fungi found in association with Bacopa monnieri as potential producers of industrial enzymes and antimicrobial bioactive compounds BABT

This study aimed to screen the endophytic fungal species of ethano-medicinal plant Bacopa monnieri (L.) Pennell for their ability to produce antimicrobial substances against Bacillus subtilis, Pseudomonas aeroginosa, Salmonella typhimurium, Escherichia coli, Klebsiella pneumonia, Staphylococcus aureus, and Candida albicans. Endophytes were also screened for their ability to produce amylase, cellulase, protease and lipase to evaluate their ecological role within the host plant. Immobilization of Beauveria bassiana Lipase on Silica Gel by Physical Adsorption BABT

Extracellular lipase from Beauveria bassianastrain CG481 was immobilized by using thirteen different immobilization protocols. Silica gel was chosen as the most suitable adsorbent with 94.8% of activity yield. The adsorption on silica gel did not change the optimum pH (8.5) and temperature (45ºC) values of the free lipase (FL) for lipolytic activity, and it showed higher activities in extreme conditions (pH 9.0 to 10.5, 60ºC). The lipase immobilized on silica gel (ILS) showed enhanced stability at pH 7.0 after 120 h incubation (69.0%) when compared to FL (33.3%). Extraction of lipase from Burkholderia cepacia by PEG/Phosphate ATPS and its biochemical characterization BABT

This work aimed to study the partitioning of a lipase produced by Burkholderia cepacia in PEG/Phosphate aqueous two phase system (ATPS) and its characterization. Lipase was produced by B. cepacia strains in a fermenter. Enzyme partitioning occurred at pH 6.0 and 8.0, using PEG 1500 and 6000 on two tie lines. Metal ions, pH and temperature effects on enzyme activity were evaluated. Five milliliter of 7.5% olive oil emulsion with 2.5% gumarabic in 0.1M sodium phosphate buffer at pH 8.0 and 37ºC were used for the activity determinations. Statistical optimization of culture conditions by response surface methodology for synthesis of lipase with Enterobacter aerogenes BABT

Optimization of lipase production by Enterobacter aerogenes was carried out using response surface methodology (RSM) where the statistical model was obtained by fractional factorial central composite design. The influence of various physico-chemical parameters, viz. temperature, oil concentration, inoculum volume, pH and incubation period on lipase production was examined. Optimization of physico-chemical parameters resulted 1.4- fold increase in lipase activity. The optimum levels of parameters were 34°C, oil concentration 3%, inoculum volume 7%, pH 7 and incubation time 60 h for obtaining a maximum lipase activity of 27.25 U/ml.

Immobilization of Lipases Produced by the Endophytic Fungus Cercospora kikuchii on Chitosan Microparticles BABT

This work studied the immobilization of Cercospora kikuchii lipases on chitosan microparticles by chemical attachment on chitosan acetate microparticles activated by glutaraldehyde (CAM) added before or after the enzyme and physical adsorption on highly deacetylated chitosan hydrochloride microparticles (CHM). Lipases covalently immobilized on pre-activated CAM showed better performance retaining 88.4% of the enzymatic activity, with 68.2% of immobilization efficiency (IE). The immobilized enzyme retained an activity of about 53.5 % after five reuses, using p-NPP as substrate. Lipase production by solid-state fermentation in fixed-bed bioreactors BABT

In the present work, packed bed bioreactors were employed with the aim of increasing productivity and scaling up of lipase production using Penicillium simplicissimum in solid-state fermentation. The influence of temperature and air flow rate on enzyme production was evaluated employing statistical experimental design, and an empirical model was adjusted to the experimental data. It was shown that higher lipase activities could be achieved at lower temperatures and higher air flow rates. The maximum lipase activity (26.4 U/g) was obtained at the temperature of 27°C and air flow rate of 0.8 L/min.

Mutation breeding of lipase-producing strain Flavobacterium sp. by supercritical CO2 with hydrazine hydrate BABT

This work aimed to obtain an ideal mutant strain with higher lipase yield using hydrazine hydrate (HZH) as a novel additive to treat Flavobacterium sp. strain YY25 by supercritical CO2. The survival rate and the positive mutation rate of the tested strain were strongly dependent on the dose of HZH. The treatment by 0.5% HZH in supercritical CO2 (8 MPa, 35°C) for 30 min provided 58.3% of positive mutation rate and an expected mutant strain with about 76.7% increase in lipase yield compared with the wild strain. Abstract The search for new biocatalysts has aroused great interest due to the variety of micro-organisms and their role as enzyme producers. Native lipases from Aspergillus niger and Rhizopus javanicus were used to enrich the n-3 long-chain polyunsaturated fatty acids content in the triacylglycerols of soybean oil by acidolysis with free fatty acids from sardine oil in solvent-free media. This work studied amylase, lipase, polygalacturonase and protease production by fifty-six isolates of Mucor belonging to 11 different taxa, selected from herbivores dung using solid media. The results showed that the majority of the isolates presented several enzymatic activities with predominance of polygalacturonase (96%), followed by amylase (84%), protease (82%) and lipase (66%).
