Audemard, Corinne; Le Roux, Frédérique; Sauriau, Pierre-guy; Sautour, Benoît; Combes, C.; Coustau, C.; Blachier, Philippe; Berthe, Franck.
Over the last thirty years, the French production of flat oyster Ostrea edulis dropped from15,000 tons/year to 1,500 tons. Marteilia refringens, a parasite of the flat oyster's digestive gland, is partly responsible for this drop. The fight against this pathogen agent requires a better knowledge of its parasitic cycle. The main hypothesis is the existence of a heteroxenous cycle, that is to say a cycle involving more than one host. However, for thirty years this or these hosts have not been found, and the cycle of Marteilia refringens remains unknown.

Tige, Gilbert; De Kergariou, Gabriel; Cochennec, Nathalie; Rabouin, Marie-agnès.
A systematic surveillance progam conducted since 1980 along the coasts of Britanny has followed the evolution and epidemiological state of the natural O. eulis oyster beds and seed areas in the intertidal zones and deep water. Presently, Marteilia refringens exists endemically in Brittany estuaries ; the rate and degree of infestation varies from year to year depending on the hydrological conditions and the geographical region. In no case, however, is the return of flat oyster culture envisoned. Bonamia ostreae is present not only in the estuaries but also in the deep-water rearing areas (Baie de Quiberon, Cancale, Baie de St Brieuc). The infestation increases with age, but also depends on the quantity of the stock and the culture practises. In particular,...

In this study, the life cycle of Paracartia grani, a calalloid copepod belonging to the Acartiidae family was determined in Thau lagoon. Indeed, the copepod involvement in the life cycle of the parasite Marteilia refringens affecting the bivalve productions is suspected. Mesozooplanktonic community .was monitored twice a month. over two years at a fixed station in the lagoon. Sampling has identified P. grani as the acartiid dominant species in summer. From April to January, the copepod is found in the water column while from February to early April it remains in the sediment as diapausing eggs. The analysis of the population structure (size spectmm, contribution of developmental stages and sex ratio) has revealed that there are in between 6 and 9...

Robert, Rene; Pichot, Yves; Comps, Michel.
From 1988 to 1989, a study has been conducted pertaining to the growth of the flat oyster Ostrea edulis, as well as Marteilia refringens and Bonamia ostreae outbreaks. Commercial mono specific farming (flat oysters), with low density or mixed (flat and hollow oysters) along with higher density, has been taking place in the Arcachon basin oceanographic sectors, where main hydrobiological parameters were acquired simultaneously. The excellent growth of the flat oyster was witnessed in mono specific farming. However this growth was average in mixed farming, probably due to higher total oyster density and interspecies competition. Regardless of the type of farming, Marteilia refringens infection was very quick and may have been related to climatic bay water...

Testing of the Argentina oyster, Ostrea puelchana, in several French oyster farming sites ArchiMer
Pascual, Marcela; Martin, Anne-genevieve; Zampatti, Eduardo; Coatanea, Denis; Defossez, Jérôme; Robert, Rene.
Because of diseases affecting fIat oyster Ostrea edulis, acclimatization of the Argentina oyster Ostrea puelchana has been tested in 1989-90 in five French oyster farming sites. Following ICES recommendations , hatchery produced juveniles, from parents imported from Argentina, were transferred to experimental si tes in summer 1989. Very high mortalities occurred, reaching 46 to 98% in March-Apri1 1990. They continued later on, resulting in the end of experiments in March 1991. The growth of surviving oysters was relatively poor in comparison of the local species. Parasites Marteilia refringens and Bonamia ostreae have been detected but at rates inadequate ta explain the high mortalities. AlI these results lead not to retain this species for breeding on...

Characterization of the protozoan parasite Marteilia refringens infecting the dwarf oyster Ostrea stentina in Tunisia ArchiMer
Elgharsalli, Refka; Aloui-bejaoui, Nejla; Sala, Hedi; Chollet, Bruno; Joly, Jean-pierre; Robert, Maeva; Couraleau, Yann; Arzul, Isabelle.
Marteilia refringens is a protozoan parasite recognized as a significant pathogen of the European flat oyster Ostrea edulis. The life cycle of this species is still poorly known, although there is evidence of the need for intermediate host(s). In the present study, we have used molecular approaches to identify this parasite in samples of the dwarf oyster Ostrea stentina after reports of massive mortality along the Tunisian coasts. In 2009 we evaluated the status of O. stentina from Monastir and checked if there was an infection with M. refringens, using polymerase chain reaction assays. Of the 103 tested O. stentina, 85 were PCRpositive using a Marteilia genus-specific assay. Additional assays were subsequently carried out on some samples collected in 2010...

Grizel, Henri; Tige, Gilbert.
Observations on the cycle of Marteilia refringens in light and electronic microscopy led us to realise in situ experiences of experimental infestation. According to the rivers the period of infestation varied between may and auguste. During the winter the parasite seems to be absent from the tissues of the oyster.

Report about mollusc diseaes ArchiMer
Berthe, Franck.
Of the 75 laboratories contacted through the survey, 54 answered, 14 of which are partially or totally devoted to mollusc diseases. In fact, only 4 laboratories are devoted full-time to this work. Of the 14 laboratories that declared to work on the diagnosis of mollusc diseases, 12 stated that they worked with mussels, 12 with oysters, 9 with clams and 8 with other mollusc species. This is coherent with available production information. According to production data in 2001 (FAO source), the main mollusc species in the region are the blue mussel, Mytilus edulis (297,485 tonnes), the Mediterranean mussel, Mytilus galloprovincialis (131,014 tonnes), the Pacific cupped oyster, Crassostrea gigas (127,323 tonnes, an important part of which is produced on the...

Contribution to the understanding of the cycle of the protozoan parasite Marteilia refringens ArchiMer
Arzul, Isabelle; Chollet, Bruno; Boyer, S.; Bonnet, D.; Gaillard, Juliette; Baldi, Yoann; Robert, Maeva; Joly, Jean-pierre; Garcia, Celine; Bouchoucha, Marc.
The paramyxean parasite Marteilia refringens infects several bivalve species including European flat oysters Ostrea edulis and Mediterranean mussels Mytilus galloprovincialis. Sequence polymorphism allowed definition of three parasite types ‘M’, ‘O’ and ‘C’ preferably detected in oysters, mussels and cockles respectively.Transmission of the infection from infected bivalves to copepods Paracartia grani could be experimentally achieved but assays fromcopepods to bivalves failed. In order to contribute to the elucidation of the M. refringens life cycle, the dynamics of the infection was investigated in O. edulis, M. galloprovincialis and zooplankton over one year in Diana lagoon, Corsica (France). Flat oysters appeared non-infected while mussels were infected...

Responsible for massive epizootics, the Marteilia type parasites are very dangerous infectious agents for oysters. As a matter of fact, in France the spreading out of the parasitosis caused by Marteilia refringens disturbed dangerously the flat oyster, Ostrea edulis, production; destroying a large percentage of the farms on the Atlantic coast. The fight against marteiliose implies a good management of the contamination risk, that is to say the identification of the parasite(s), the characterization of the host(s), vector(s) or reservoir(s) and the understanding of the parasite's transmission mode.

Validation of in situ hybridisation and histology assays for the detection of the oyster parasite Marteilia refringens ArchiMer
Thebault, Anne; Bergman, S; Pouillot, R; Le Roux, Frédérique; Berthe, Franck.
An in situ hybridisation technique has been developed for the detection of infection in oysters with Marteilia refringens with particular emphasis on light infections or confirmation of suspected cases by means of histology. Although validation of new diagnostic methods is usually achieved by comparison with standard techniques, in our case the sensitivity and specificity of the standard (histology) had not previously been established. Another point to consider is that surveillance and monitoring frequently target populations displaying different levels of prevalence under different field conditions. The objective of our study was to evaluate the sensitivity and specificity values of in situ hybridisation and histology for the detection of M refringens,...

Infection dynamics of Marteilia refringens in flat oyster Ostrea edulis and copepod Paracartia grani in a claire pond of Marennes-Oleron Bay ArchiMer
Audemard, Corinne; Sajus, Marie-céline; Barnaud, Antoine; Sautour, Benoît; Sauriau, Pierre-guy; Berthe, Franck.
The protozoan parasite Marteilia refringens has been partly responsible for the severe decrease in the production of the European flat oyster Ostrea edulis Linnaeus in France since the 1970s. The calanoid copepod Paracartia grani Sars was recently found to be a host for M refringens in French shallow-water oyster ponds ('claires'). This study reconsidered M refringens transmission dynamics in the light of this finding, taking into account not only oyster infection dynamics and environmental factors but also data concerning the copepod host. P. grani population dynamics in the claire under study revealed that this species is the dominant planktonic copepod in this confined ecosystem. During winter, M refringens overwintered in O. edulis, with P. grani...
