ABSTRACT This study describes the in vitro seed germination and micropropagation of Plukenetia volubilis (sacha inchi), an oilseed crop rich in omega-3 fatty acids, with health benefits and several industrial applications. Seed germination was evaluated in different culture media (MS and 1/2 MS), seed coat presence/absence and culture temperature (18 °C and 28 °C). Micropropagation was performed using axillary bud development (ABD) on nodal segments from in vitro seedlings. KIN, BAP and 2-ip were evaluated for ABD, and the effect of modified MS in 453 mg L-1 CaCl2 and 351.62 mg L-1 MgSO4 on ABD and shoot survival was assessed to improve the process. Survival and development of reintroduced Cattleya intermedia plants related to abiotic factors and herbivory at the edge and in the interior of a forest fragment in South Brazil Acta Botanica

ABSTRACT Biotic and abiotic factors, such as luminosity, temperature, air humidity, and herbivory, can affect the establishment of reintroduced plants in natural habitats. This study evaluated the effects of these factors on the survival and growth of Cattleya intermedia plants reintroduced into a forest fragment in South Brazil. Plants of C. intermedia were obtained from in vitro seed germination in asymbiotic culture. Eighty-eight plants were reintroduced at both the forest edge and forest interior. Plants with greater shoot heights and number of leaves and pseudobulbs suffered more damage from herbivores at the edge. This study aimed to establish and propagate in vitro plants of Bouchea fluminensis, a medicinal species known in Brazil as gervão-falso ("false verbena"), evaluating the influences of different growth regulators on in vitro multiplication and rooting stages, as well as examining ex vitro acclimatization of rooted plants. Explants were established on Murashige and Skoog medium at half strength of salts and vitamins without growth regulators. The internal morphology of embryos from immature and mature fruits of Heliconia bihai (L.) L. cv. Lobster Claw Two was examined. Embryos were inoculated into MS media (full MS and ½ MS) and GA3 (0, 2.5 and 5 mg L-1) with either sucrose or glucose. These plantlets were then replicated and transferred to MS medium (full MS or ½ MS) with 0 or 2.5 mg L-1 BAP and their multiplication was evaluated 30 and 45 days after inoculation. No previous studies have investigated the use of graphite instead of activated charcoal in orchids. In this work, different concentrations of activated charcoal or graphite were added to KC medium to darken the culture medium and stimulate the in vitro propagation of Cattleya bicolor and of a double hybrid orchid (‘BLC Pastoral Innocence’). Biochemical characterization of systemic bacteria in bananas, sensitivity to antibiotics and plant phytotoxicity during shoot proliferation Agronomy

The objective of this work was to characterize the biochemically systemic bacterial isolated from banana plants, to evaluate the bacterial sensitivity to antibiotics, and to determine the phytotoxicity of banana shoots during in vitro proliferation. Systemic bacteria belonging to the Klebsiella and Aeromonas genera were isolated from the “Maravilha” (FHIA 01 AAAB), “Preciosa” (PV 4285 AAAB) and “Thap Maeo” (AAB) varieties and were then characterized. Dynamics of proteins, carbohydrates and global DNA methylation patterns during induction of nodular cluster cultures from seeds of Vriesea reitzii Agronomy

Tissue culture techniques have been employed for bromeliad mass propagation by means of the morphogenetic route of nodular cluster cultures (NCs). This study aimed to assess protein, carbohydrate and global DNA methylation (GDM) level dynamics during NCs induction from Vriesea reitzii seeds. Seeds were inoculated into Murashige and Skoog (MS) liquid medium supplemented with 4 µM α-naphthaleneacetic acid (NAA) to induce NCs and in culture medium without plant growth regulators to form normal seedlings. In Iran, a large number of Rosa species have been exposed to extinction and therefore preservation techniques are necessary to safeguard their future. In the present investigation, the objectives were to optimize the micropropagation and medium-term conservation of one of these wild species, Rosa pulverulenta. The objective of this study was to morpho-anatomically characterize nodular embryogenic calluses from zygotic embryos of peach palm during the induction of somatic embryogenesis. Immature zygotic embryos were pre-treated in MS medium added to Picloram and 2,4-D (25 µM) and BAP (0, 5, 10 µM). After three months, primary calluses were transferred to MS induction medium added to Picloram and 2,4-D (450 µM). After six months, the embryogenic calluses were then histologically analyzed and cultivated in the maturation medium. Effects of glutathione supplementation and carbon source during somatic embryogenesis of Acca sellowiana (O.Berg) Burret (Myrtaceae) Biological Sciences

 The present study intended to investigate the effects of different glutathione (GSH) levels (0, 0.1, 0.5 and 1 mM) on the somatic embryogenesis (SE) induction of Acca sellowiana. Besides, we evaluated the effect of different carbon sources (sucrose and maltose) on the somatic embryos conversion. GSH-supplemented treatments resulted in improved SE induction rates (~70%) as compared to the control GSH-free (~35%) after 50 days of culture. The total number of somatic embryos obtained did not differ between treatments, but significant differences were observed for the embryonic stages after 80 days of culture. Micropropagation of banana plants has been used successfully in commerce. Banana plantation has a great interest in Egypt due to suitable climate. However, some problems might thwart the plantation progress like the root-knot nematode; Meloidogyne incognita infection which can affect acclimatization and plantation of in vitro plants. Thus, our investigation aimed to produce nematode resistant in vitro plants using gamma radiation (γ). Banana in vitro plantlets were exposed to gamma radiation at several doses then replanted in vitro. Sensitivity of banana plants to irradiation was determined. Abtract Apuleia leiocarpa is an endangered tree species with high quality timber. Propagation by seed is difficult due to irregular fruiting and water-impermeable tegument, so asexual propagation is a promising alternative. This study aimed to evaluate in vitro and ex vitro rooting of A. leiocarpa and acclimatization of micropropagated plantlets. The hypothesis was that type of explant, indolebutyric acid (IBA) concentration and substrate influence the induction of adventitious roots in apuleia.
