Diagnostic and prognostic characteristics of phytotoxicity caused by fluoride on Spondias dulcis Forst. F. (Anacardiaceae) Anais da ABC (AABC)
Sant'Anna-Santos,Bruno F.; Azevedo,Aristéa A.; Silva,Luzimar C.; Oliva,Marco A..
The goal of this study was to determine the symptoms and microscopic damage caused by fluoride on Spondias dulcis, a fluoride-sensitive species. The plants were exposed to simulated fog with fluoride (0, 5, 10, 15 and 20 mg L-1) for 20 min daily during four consecutive days. Samples from leaflets without any apparent fluoride injury were collected to microscopic analysis. The percentage of necrosed leaf area was measured, and the level of pollutant in the dry matter from the basal and apical portions of the plant was determined. The necroses began 24 h after the first simulation mainly from the base of the leaflets. A higher level of necrosis was observed at the apical portion of the plants, a region of higher fluoride accumulation. The damage on the...

The use of papain gel cream and sunflower oil in promoting healing in a wound in dogs: three case reports Arq. Bras. Med. Vet. Zootec.
Porsani,M.Y.H.; Carvalho,L.A.R.; Pereira,C.S.; Paludetti,M.; Zangeronimo,M.G.; Pereira,L.J..
ABSTRACT Papain is a proteolytic enzyme removed from the leaves of green papaya and/or latex. This enzyme is widely known as a medicinal fruit used extensively in human medicine for the treatment of wounds of various etiologies. However, studies and reports in veterinary medicine are scarce. Another herbal drug widely used in wound healing is Sunflower oil (Helianthus annus). It has inflammatory and antimicrobial properties which stimulate the local neovascularization, promoting tissue granulation, cell migration, fibroblast proliferation, and differentiation. Three dogs were treated with infected necrosis wounds, considered large, extent, and severe, with varied etiology. All cases were treated with papain gel with the exception of one dog, which was...

Prolonged exposure of human embryonic stem cells to heat shock induces necrotic cell death Biocell
Heng,Boon Chin; Vinoth,Kumar Jayaseelan; Lu,Kai; Deng,Xuliang; Ge,Zigang; Bay,Boon Huat; Cao,Tong.
We investigated the effects of prolonged heat shock treatment on human embryonic stem cell (hESC) viability. The hESC viability steadily declined with longer exposure to heat shock treatment (43ºC). After 4 h of exposure to heat shock at 43ºC, only 56.2 ± 1.5% of cells were viable. Viability subsequently declined to 37.0 ± 3.3% and 3.5 ± 0.7% after 8 h and 16 h, respectively of heat shock treatment at 43ºC. Transmission electron micrographs showed that the morphology of the dead/dying cells after heat shock treatment was characteristic of cellular necrosis with an uncondensed chromatin and a non-intact plasma membrane. This was further confirmed by flow cytometry analysis which showed that the DNA of the dead/ dying cells was still mostly intact, unlike...

Ion movements in cell death: from protection to execution Biol. Res.
FELIPE BARROS,LUIS; CASTRO,JOEL; BITTNER,CARLA X..
Cell death is preceded by severe disruption of inorganic ion homeostasis. Seconds to minutes after an injury, calcium, protons, sodium, potassium and chloride are exchanged between the cell and its environment. Simultaneously, ions are shifted between membrane compartments inside the cell, whereby mitochondria and endoplasmic reticulum play a crucial role. Depending of the type and severity of injury, two mutually exclusive metastable states can be reached, which predict the final outcome. Cells characterized by large increases in cytosolic [Ca2+], [Na+]; and [Mg2+] swell and die by necrosis; alternatively, cells characterized by high [H+]and low [K+], with normal [Na+] and normal to moderate [Ca2+] increases die by apoptosis. The levels of these ions...

Silva,Ricardo Rocha Pavan da; Pires Junior,Osmindo Rodrigues; Grisolia,Cesar Koppe.
Studies of genotoxicity in fish caused by cyanobacterial microcystins can be useful both in determining the sensitivity of native species, as well as comparing exposure routes. The genotoxicity caused by the microcystins LR and LA from a bloom collected in a eutrophic lake, was revealed in the fish Astyanax bimaculatus, a native species from South America. LC50 (72 h) was determined as 242.81 µg L-1 and LD50 (72 h) as 49.19 µg kg-1 bw. There was a significant increase of DNA damage in peripheral erythrocytes, following intraperitoneal injection (ip) with tested concentrations of 24.58 µg kg-1 bw and 36.88 µg kg-1 bw, as well as through body exposure to a concentration of 103.72 µg L-1. Micronucleus (MN) induction was observed after ip injections of 24.58...

Histopathological lesions and DNA adducts in the liver of European flounder (Platichthys flesus) collected in the Seine estuary versus two reference estuarine systems on the French Atlantic coast ArchiMer
Cachot, Jerome; Cherel, Yan; Larcher, Thibaut; Pfohl-leszkowicz, Annie; Laroche, Jean; Quiniou, Louis; Morin, Jocelyne; Schmitz, Julien; Burgeot, Thierry; Pottier, Didier.
An epidemiological survey was conducted in the Seine estuary and in two smaller and relatively preserved estuaries on the French Atlantic coast in order to estimate the occurrence of liver lesions in European flounder, Platichthys flesus, and also to seek putative risk factors for the recorded pathologies. Four hundred and seventy-eight fish of both sexes and of different size ranges were sampled in the three studied areas, 338 of which in the Seine estuary. All fish were examined for histopathological liver lesions, while DNA adducts and otoliths were analyzed on a subsample. Five categories of hepatic lesions were recorded with the following prevalence for the Seine estuary: 36.7 % inflammations, 8 % parasites (mainly encysted nematodes), 6.5 % foci of...

Virus-like particles associated with brown muscle disease in Manila clam, Ruditapes philippinarum, in Arcachon Bay (France) ArchiMer
Dang, C.; Gonzalez, P.; Mesmer-dudons, N.; Bonami, J-r; Caill-milly, Nathalie; De Montaudouin, X..
Recently, Manila clam, Ruditapes philippinarum, populations have suffered mortalities in Arcachon Bay (SW France). Mortality was associated with extensive lesions of the posterior adductor muscle, which become progressively brown and calcified. Ultrastructural observations by transmission electron microscopy revealed tissue degradation with necrotized muscle fibres and granulocytomas. Unenveloped virus-like particles (VLPs) were detected in muscle, granulocytic, epithelial and rectal cells. VLPs were abundant in the extracellular space, in the cytoplasm (free or enclosed in vesicles) and in the nucleoplasm of granulocytes. Nuclei and mitochondria of granulocytes displayed changes which suggested reactive oxygen species production and apoptosis induction....

Early Administration of Peroxisomicine A1 (T-514 Extracted from K. parvifolia Seeds) Causes Necrosis of Implanted TC-1 Cells without Affecting Target Organs in a Murine Model International Journal of Morphology
Soto-Domínguez,Adolfo; Piñeyro-López,Alfredo; Saucedo-Cárdenas,Odila; Ramírez-Durónd,Rosalba; Waksman de Torres,Noemí; Sepúlveda-Saavedra,Julio.
Peroxisomicine A1 (PA1), one of the toxins isolated from seeds of plants of the Karwinskia genus, whose targets organs are the liver, kidney, and lungs. There is a selective toxicity in vitro to cancer-cell lines derived from the lungs, liver, and colon, compared to normal cell lines. PA1 caused apoptosis in several cancer-cell lines in culture. In toxic doses to rodents, it causes extensive apoptosis in the liver, kidney, and lungs. In our study we were interested in evaluating, for the first time, the morphological effects of administration of PA1 to implanted TC-1 cells and in the target organs in vivo. The TC-1 cells were cultured and injected into the hind limb of C57BL-6 mice. The animals were divided into 3 groups; those treated with four doses of 1...

Tissue necrosis after canine bothropic envenoming: a case report J. Venom. Anim. Toxins
FERREIRA JÚNIOR,R. S.; BARRAVIERA,B..
The authors report a case of bothropic envenoming in a male Cocker Spaniel. The animal was bitten in the ventral thoracic region, receiving treatment 4 hours later. Clinical examination revealed an extensive, painful and area of firm edema, absence of local or systemic hemorrhage, without evident neurological alterations. Clinical diagnosis was mild bothropic envenoming. Treatment consisted of 5 vials of polyvalent snake antivenom, two vials administered intravenously and three subcutaneously. Blood clotting time was always within normal values. Two days after envenoming, the animal showed hyperthermia and received enrofloxacin (5mg/kg/24h) for 10 days and ketoprofen (1mg/kg/24h) for 5 days. Seventy-two hours after envenoming, extensive subcutaneous,...

Cytotoxic effect of ICD-85 (venom-derived peptides) on MDA-MB-231 cell line J. Venom. Anim. Toxins incl. Trop. Dis.
Zare Mirakabadi,A; Mahdavi,S; Koohi,MK; Taghavian,M.
Since 1987, when chemopreventive testing programs began, more than 1,000 agents and agent combinations have been selected and evaluated in preclinical studies of chemopreventive activity against various types of cancers. In the present study we aimed to provide quantitative and qualitative characterization of biological and pharmacological activities of ICD-85 on MDA-MB-231 cell line (a highly invasive breast cancer cell line) in order to gain a better understanding of the cytotoxic and apoptotic effects of this compound. For this study, the MDA-MB-231 cell line was used and the effect of ICD-85 was assayed by measuring the activity of the cytosolic enzyme lactate dehydrogenase (LDH), released into the culture medium after membrane damage. Morphological...
