Responses of plankton and fish from temperate zones to UVR and temperature in a context of global change Ecología austral
Gonçalves,Rodrigo J; Souza,María Sol; Aigo,Juana; Modenutti,Beatriz; Balseiro,Esteban; Villafañe,Virginia E; Cussac,Víctor; Helbling,E Walter.
In the last decades, both temperature and UVB (280-315 nm) radiation on the surface of the Earth increased at growing rates as a result of human activities. Many studies had evaluated the effects of temperature on aquatic ecosystems, but now the field broadens as the combination and variations of temperature and radiation gains especial importance. In this work we attempt to revisit some of our knowledge about the effects of UVR and temperature on marine and freshwater plankton and fish from temperate regions (defined here as latitudes between 30-60º), especially from the Patagonia area, due to the special characteristics found in these sites. UVR affects (often negatively) almost all processes, from carbon fixation to behavior; and certainly all trophic...

Effect of the Puyehue-Cordon Caulle volcanic complex eruption on crustacean zooplankton of Andean lakes Ecología austral
Balseiro,Esteban; Souza,María S.; Serra Olabuenaga,Ignacio; Wolinski,Laura; Bastidas Navarro,Marcela; Laspoumaderes,Cecilia; Modenutti,Beatriz.
We studied the effect of volcanic ash on the crustacean zooplankton (mainly cladocerans) in the Andean North-Patagonian lakes that were affected by the eruption of the Puyehue-Cordón Caulle complex. The study was carried out at three different scales: sub cellular, population and ecosystemic. Cladocerans are filter feeding organisms that cannot distinguish between food (algae less than 30 µm) and suspended ash, therefore when feeding they ingest this material. The ash affected negatively the individuals at a biochemical level, increasing the oxidative stress reacting enzymes like Glutation-S-transferase and the biomarker Acetyl cholinesterase. At the population level, we recorded a decrease in survival and fecundity, in particular, when ash concentration...

Discrimination of population of recombinant inbred lines of rye (Secale cereale L.) for different responses to nitrogen-potassium stress assessed at the seedling stage under in vitro conditions Electron. J. Biotechnol.
Smolik,Miłosz.
Background: Plants differ in the methods used to acquire nutrients from environments with low nutrient availability, and may change the morphology of their ‘root architecture’ to be able to take up nutrients. Results: In the present study rye response to stress caused by high and low nitrogen-potassium treatments in mature embryos cultures was described within a population consisting of one hundred and thirty eight recombinant inbred lines of rye. Characterization of the response of recombinant inbred lines (RILs) to nutrient stress was presented as the results of analyses of morphological traits, and physiological and biochemical parameters of the seedlings grown in both treatments. A wide range of variability of individual RILs to induced stress was...

Oxidative stress assessment by glutathione peroxidase activity and glutathione levels in response to selenium supplementation in patients with Mucopolysaccharidosis I, II and VI Genet. Mol. Biol.
Oliveira-Silva,José Araújo de; Yamamoto,Joyce Umbelino Pinto; Oliveira,Renata Bernardes de; Monteiro,Vaneisse Cristina Lima; Frangipani,Beatriz Jurkiewcz; Kyosen,Sandra Obikawa; Martins,Ana Maria; D’Almeida,Vânia.
Abstract We assessed levels of plasma selenium (Se), selenoproteins and their change after Se supplementation in patients with mucopolysaccharidosis (MPS) types I, II and VI. This was done in a retrospective study of the medical records of 30 patients with MPS I (n=13), MPS II (n=9) and MPS VI (n=8) who were being treated with enzyme replacement therapy. As part of routine nutritional monitoring, Se levels were measured, revealing that 28 patients (93.3%) had values below the normal range. Therefore, they received supplementation for 12 months, and Se was measured after 6 and 12 months. Glutathione peroxidase (GPx) activity, total glutathione (GSHt), oxidized glutathione (GSSG) and reduced glutathione (GSH) were measured at baseline and 6 months after Se...

DNA repair diseases: what do they tell us about cancer and aging? Genet. Mol. Biol.
Menck,Carlos FM; Munford,Veridiana.
The discovery of DNA repair defects in human syndromes, initially in xeroderma pigmentosum (XP) but later in many others, led to striking observations on the association of molecular defects and patients' clinical phenotypes. For example, patients with syndromes resulting from defective nucleotide excision repair (NER) or translesion synthesis (TLS) present high levels of skin cancer in areas exposed to sunlight. However, some defects in NER also lead to more severe symptoms, such as developmental and neurological impairment and signs of premature aging. Skin cancer in XP patients is clearly associated with increased mutagenesis and genomic instability, reflecting the defective repair of DNA lesions. By analogy, more severe symptoms observed in...

Abstract Human longevity is a polygenic and multifactorial trait. Pathways related to lifespan are complex and involve molecular, cellular, and environmental processes. In this analytical observational study, we evaluated the relationship between environment factors, oxidative stress status, DNA integrity level, and the association of FOXO3 (rs2802292), SOD2 (rs4880), APOE (rs429358 and rs7412), and SIRT1 (rs2273773) polymorphisms with longevity in oldest-old individuals from southeastern Brazil. We found an association between the FOXO3 GG genotype and gender. While lifestyle, anthropometric, and biochemical characteristics showed significant results, DNA damage and oxidative stress were not related to lifespan. We found that long-lived individuals with...

Adaptive response: some underlying mechanisms and open questions Genet. Mol. Biol.
Dimova,Evgeniya G.; Bryant,Peter E.; Chankova,Stephka G..
Organisms are affected by different DNA damaging agents naturally present in the environment or released as a result of human activity. Many defense mechanisms have evolved in organisms to minimize genotoxic damage. One of them is induced radioresistance or adaptive response. The adaptive response could be considered as a nonspecific phenomenon in which exposure to minimal stress could result in increased resistance to higher levels of the same or to other types of stress some hours later. A better understanding of the molecular mechanism underlying the adaptive response may lead to an improvement of cancer treatment, risk assessment and risk management strategies, radiation protection, e.g. of astronauts during long-term space flights. In this mini-review...

Combined GSTM1 and GSTT1 null genotypes are strong risk factors for atherogenesis in a Serbian population Genet. Mol. Biol.
Grubisa,Ivana; Otasevic,Petar; Vucinic,Nada; Milicic,Biljana; Jozic,Tanja; Krstic,Slobodan; Milasin,Jelena.
Abstract Oxidative stress (OS) plays an important role in atherogenesis and since glutathione S-transferases (GSTs) provide protection against OS, we have tested the hypothesis that deletion polymorphisms in two GSTs (GSTM1 and GSTT1) may affect the risk of developing atherosclerosis. A total of 382 individuals (200 patients with atherosclerosis and 182 healthy controls) were included in this association study. Genomic DNA was isolated from peripheral blood cells or from buccal epithelial cells and genotyping was performed using multiplex-PCR or real-time PCR methods. GSTM1 null genotype was significantly more frequent in atherosclerotic patients than in controls (52.0% vs 34.1%) and individuals with the GSTM1 null genotype had an approximately 2-fold...

Differential gene expression between wild-type and Gulo-deficient mice supplied with vitamin C Genet. Mol. Biol.
Jiao,Yan; Zhang,Jifei; Yan,Jian; Stuart,John; Gibson,Griffin; Lu,Lu; Willaims,Robert; Wang,Yong Jun; Gu,Weikuan.
The aim of this study was to test the hypothesis that hepatic vitamin C (VC) levels in VC deficient mice rescued with high doses of VC supplements still do not reach the optimal levels present in wild-type mice. For this, we used a mouse scurvy model (sfx) in which the L-gulonolactone oxidase gene (Gulo) is deleted. Six age- (6 weeks old) and gender- (female) matched wild-type (WT) and sfx mice (rescued by administering 500 mg of VC/L) were used as the control (WT) and treatment (MT) groups (n = 3 for each group), respectively. Total hepatic RNA was used in triplicate microarray assays for each group. EDGE software was used to identify differentially expressed genes and transcriptomic analysis was used to assess the potential genetic regulation of Gulo...

Evaluation of oxidative stress markers and cardiovascular risk factors in Fabry Disease patients Genet. Mol. Biol.
Müller,Karen B.; Galdieri,Luciano C.; Pereira,Vanessa G.; Martins,Ana M.; D'Almeida,Vânia.
Fabry Disease, an X-linked inborn error of metabolism, is characterized by progressive renal insufficiency, with cardio and cerebrovascular involvement. Homocysteine (Hcy) is considered a risk factor for vascular diseases, but the mechanisms by which it produces cardiovascular damage are still poorly understood. Regarding the vascular involvement in FD patients, the analysis of factors related to thromboembolic events could be useful to improving our understanding of the disease. The aim of this study was to evaluate plasma Hcy and other parameters involved in the methionine cycle, as well as oxidative stress markers. The sample consisted of a group of 10 male FD patients and a control group of 8 healthy individuals, paired by age. Venous blood was...

Parental exposure to the herbicide diuron results in oxidative DNA damage to germinal cells of the Pacific oyster Crassostrea gigas ArchiMer
Barranger, Audrey; Heude-berthelin, Clothilde; Rouxel, Julien; Adeline, Beatrice; Benabdelmouna, Abdellah; Burgeot, Thierry; Akcha, Farida.
Chemical pollution by pesticides has been identified as a possible contributing factor to the massive mortality outbreaks observed in Crassostrea gigas for several years. A previous study demonstrated the vertical transmission of DNA damage by subjecting oyster genitors to the herbicide diuron at environmental concentrations during gametogenesis. This trans-generational effect occurs through damage to genitor-exposed gametes, as measured by the comet-assay. The presence of DNA damage in gametes could be linked to the formation of DNA damage in other germ cells. In order to explore this question, the levels and cell distribution of the oxidized base lesion 8-oxodGuo were studied in the gonads of exposed genitors. High-performance liquid chromatography...

Cross talk: Two way allelopathic interactions between toxic Microcystis and Daphnia ArchiMer
Bojadzija Savic, Gorenka; Bormans, Myriam; Edwards, Christine; Lawton, Linda; Briand, Enora; Wiegand, Claudia.
Due to eutrophication, freshwater ecosystems frequently experience cyanobacterial blooms, many of which produce bioactive metabolites that can affect vertebrates and invertebrates life traits. Zooplankton are able to develop tolerance as a physiological response to cyanobacteria and their bioactive compounds, however, this comes with energetic cost that in turn influence Daphnia life traits and may impair populations. Vice versa, it has been suggested that Daphnia are able to reduce cyanobacterial dominance until a certain cyanobacterial density; it remains unclear whether Daphnia metabolites alone influence the physiological state and bioactive metabolites production of cyanobacteria. Hence, this study investigates mutual physiological reactions of toxic...

Comparative embryotoxicity and genotoxicity of the herbicide diuron and its metabolites in early life stages of Crassostrea gigas: implication of reactive oxygen species production ArchiMer
Behrens, Daphne; Rouxel, Julien; Burgeot, Thierry; Akcha, Farida.
Herbicides are one of the major classes of pollutants contaminating coastal waters over the world. Among them, diuron (3-(3,4-dichlorophenyl)-1,1-dimethylurea) is a phenylurea herbicide frequently detected in oyster-producing area, known to be toxic for this important exploited non-target species. With the aim to investigate the mechanisms by which diuron displays its toxicity in oyster, the implication of both biotransformation and oxygen reactive species (ROS) production was studied considering embryotoxicity and genotoxicity as endpoints. Comparative embryotoxicity and genotoxicity of diuron and its main metabolites (DCPMU, DCPU and 3,4-DCA) were thus studied on oyster larvae by the embryo-larval bioassay on D larvae and the comet assay on trochophore...

Effect of dietary probiotic Pediococcus acidilactici on antioxidant defences and oxidative stress status of shrimp Litopenaeus stylirostris ArchiMer
Castex, Mathieu; Lemaire, Pierrette; Wabete, Nelly; Chim, Liet.
in this study we evaluated the effects of the probiotic Pediococcus acidilactici MA18/5M on shrimp, Litopenaeus stylirostris (also called Penaeus stylirostris), first on antioxidant defences and secondly on the oxidative stress status in the shrimps' haemolymph and digestive gland. We conducted two experiments with the same protocol in which shrimps were fed two diets for three weeks: a control diet and a probiotic diet containing 1 g of live P. acidilactici MA18/5M kg(-1). In the first experiment, the shrimps were found to be healthy over the trial period; no mortalities and non-detectable signs of infection were recorded. These resulted in high final survival rates (above 90% in both treatments). On the other hand, during the second trial, carried out at...

Variation in the link between oxygen consumption and ATP production, and its relevance for animal performance ArchiMer
Salin, Karine; Auer, Sonya K.; Rey, Benjamin; Selman, Colin; Metcalfe, Neil B..
It is often assumed that an animal's metabolic rate can be estimated through measuring the whole-organism oxygen consumption rate. However, oxygen consumption alone is unlikely to be a sufficient marker of energy metabolism in many situations. This is due to the inherent variability in the link between oxidation and phosphorylation; that is, the amount of adenosine triphosphate (ATP) generated per molecule of oxygen consumed by mitochondria (P/O ratio). In this article, we describe how the P/O ratio can vary within and among individuals, and in response to a number of environmental parameters, including diet and temperature. As the P/O ratio affects the efficiency of cellular energy production, its variability may have significant consequences for animal...

Influence de l'environnement trophique de l'élevage en biofloc sur les performances physiologiques de la crevette Litopenaeus stylirostris : Étude de paramètres de la nutrition, de l'immunité et de la reproduction ArchiMer
Cardona, Emilie.
Biofloc is an intensive rearing system with zero or minimal water exchange where a diverse population of microorganisms (microalgae, zooplankton and bacteria) develops in association with organic matter to form the floc particles. These particles play the double role of biological filter and dietary supplement. This dissertation aims to better understand the process of this rearing system and its interactions with the Litopenaeus stylirostris shrimp. Two specific objectives were integrated within the framework of this general objective: (i) to measure the production gain from biofloc rearing and (ii) to study the interaction between biofloc environment and shrimp and to assess its role on production performances of shrimps. Thus, our results show...

Microarray-Based Identification of Gonad Transcripts Differentially Expressed Between Lines of Pacific Oyster Selected to Be Resistant or Susceptible to Summer Mortality ArchiMer
Fleury, Elodie; Moal, Jeanne; Boulo, Viviane; Daniel, Jean-yves; Mazurais, David; Henaut, Alain; Corporeau, Charlotte; Boudry, Pierre; Favrel, Pascal; Huvet, Arnaud.
Summer mortality of the Pacific oyster Crassostrea gigas is the result of a complex interaction between oysters, their environment, and pathogens. Heredity appears to be a major factor determining the sensitivity of oysters to summer mortality, allowing resistant (R) and susceptible (S) lines to be produced. We conducted genome-wide expression profiling of R and S gonads during the 3-month period preceding a summer mortality event, using a cDNA microarray that we designed. ANOVA analysis revealed that 34 genes were differentially expressed between R and S lines on four dates preceding the mortality event. Annotation of some of these genes highlights reproduction and its allocation and antioxidant defenses as the main pathways that operate differentially...
