Although viruses have been associated with mortality outbreaks in different mollusc species, little information is available on their exact affiliation and taxonomic position. Herpes viruses have attracted particular attention because of their economic and ecological impact on both farmed and wild marine molluscs over the past 20 years. Investigations based on molecular approaches have been carried out on the herpes virus infecting Pacific oysters in France. The monitoring network IFREMER /REMORA is assessing on a yearly basis, from February to December, mortality, growth and quality criteria of two oyster batches ("juveniles" = one-year class ; and "adults" = two-years class), deployed among various stations of the main French oyster areas. This network is a tool for trend estimations (geographic and chronological, on the national scale) ; it must not be considered as an accurate estimation of professional oyster ongrowings. In April 2010, the sampling of juvenile Pacific cupped oysters originating from France was undertaken from a farm located off the coast of the Marche region in Italy. Samples were sent to the national reference laboratory for mollusc diseases for bacteriological, histological, electron microscopical and molecular analysis. Classical and Real-Time PCR indicated the presence of the microvariant (OsHV-1 mu var) of Type 1 Ostreid Herpes virus (OsHV-1) despite the absence of clinical and pathological signs normally associated with the presence of this variant. Virus-induced genes were identified using suppression subtractive hybridisation (SSH) from Pacific cupped oyster, Crassostrea gigas, haemocytes challenged by OsHV-1. A total of 304 clones from SSH forward library were sequenced. Among these sequences, some homologues corresponded to (i) immune related genes (macrophage express protein, IK cytokine, interferon-induced protein 44 or multicopper oxidase), (ii) apoptosis related genes (Bcl-2) and (iii) cell signalling and virus receptor genes (glypican).
