Pathogenic threats and probiotic use in larviculture of the scallop, Pecten maximus ArchiMer

During a series of experiments, bacteriological elements in scallop larval rearing were investigated: larvae susceptibility to pathogens as a function of their age, and the use of probiotic bacteria during larviculture. Younger larvae (d5 PF) were highly more susceptible to pathogenic-challenge than their older siblings, which were challenged at an older age (d15 PF). A challenge with 104 CFU mL−1 of V. pectenicida killed 100% of d5 PF larvae 7 days following challenge, yet killed only 9% of d15 PF larvae 9 days following challenge. Larval mortalities occurring in molluscan hatcheries have often been associated with bacterial contamination. Although batches of oyster and clam larvae have been routinely reared in the hatchery of Argenton without antibiotics, high larval mortalities have been recorded with Pecten maxirnus under similar conditions. For this species, an addition of chloramphenicol was found necessary. However, this chemical has now been banned in Europe. Thus either substitition products or an improvement in the rearing procedures is essential. Caracteristiques ultrastructurales du spermatozoide de la coquille Saint-Jacques, Pecten maximus (Mollusque, Bivalve) ArchiMer

The spermatozoon of Pecten maximus is of the primitive type usually associated with species having external fertilization or emitting the sperm in the water. The size of the nucleus is reduced. The middle piece is similar to that reported for several Bivalves. It contains glycogen. But the spermatozoon of P. maximus exhibits its own ultrastructural characteristics: the short acrosome differs in size, shape, position and ultrastructure or by a combination of these from that of many bivalves. No axial rods was observed in contrast to several species of Mytilidae, Veneridae and Ostreidae. These originalities could go to the genus level.

SELEDRAG : Etude comparative de la sélectivité des dragues à coquilles Saint-Jacques Pecten maximus ArchiMer

The SELEDRAG project was conducted in French territorial waters in May 2019. It was a short project, lead by the CNPMEM (French National Committee for Marine Fisheries), with Ifremer as scientific partner, and funded by “France Filière Pêche”. The context of the project is the French King Scallop fishery in the Eastern Channel, where 720 fishing boats are directly concerned by this species. Despite its importance, there is very little management for this species at the European level, but several binding rules for French fishermen in order to protect the stock, are not applicable to other countries. A coupled biophysical model for the distribution of the great scallop Pecten maximus in the English Channel ArchiMer

In this paper we used a modelling approach integrating both physical and biological constraints to understand the biogeographical distribution of the great scallop Pecten maximus in the English Channel during its whole life cycle. A 3D bio-hydrodynamical model (ECO-MARS3D) providing environmental conditions was coupled to (i) a population dynamics model and (ii) an individual ecophysiological model (Dynamic Energy Budget model). L'analyse de ces données permet de faire quelques constatations sur le cycle sexuel de la coquille Saint-Jacques dans cette zone. - Le cycle sexuel de la population est très étalé dans le temps. This work is part of the program, "recruitment determinism in scallops" initiated in 1992 by the "Contrat de Baie". The aim of this research was to explain interannual abundance fluctuations observed for Pecten maximus, in the Bay of Brest. To this end an analytical approach to the life cycle of the species was undertaken to determine its critical phases. A phase may be considered as "critical" if the mortality rate at its level varies from one year to the other. Ontogenic changes in the contents of dopamine, norepinephrine and serotonin in larvae and postlarvae of the bivalve Pecten maximus ArchiMer

In the present study, norepinephrine (NE), epinephrine (E), dopamine (DA) and 5-hydroxy-tryptamine (5HT) have been quantified by high performance liquid chromatography (HPLC) in Pecten maximus during larval and post-larval development. With average values ranging from 2 to 5 pg mug(-1) of protein, NE remained low through the whole larval life, while epinephrine (E) was undetected. DA and 5HT were the most abundant monoamines with significant variations between larval, pre-metamorphic and post-larval stages. Nous disposons aujourd'hui d'o utils diagnostiques sensibles et spécifiques (PCR, HIS ... ) et d'un protocole de transmission de l' infection qui permettent d'assurer un suivi des productions larvaires en écloserie de bivalves pour les espèces considérées comme sensibles aux infections à herpèsvirus, mais également pour de nouvelles espèces. En effet, des mortalités massives (100 %) ont été rapportées dans une écloserie de coquille St-Jacques en Bretagne et ont pu être associées à la détection de OsHV -1. Les résultats obtenus concernant cette espèce, Pecten maximus, complètent et reflètent l'ensemble des travaux réalisés.

Le transfert en mer de post-larves de coquille Saint-Jacques depuis la nurserie, à terre, jusqu'à la station de prégrossissement, en mer, provoque de faibles survies : 20 à 50 % L'étude du passage en mer montre 2 stress élémentaires principaux : la dé-fixation des post-larves et leur émersion pendant le transport (l'effet des chocs thermiques n'a pas été étudié ici). Quelques comparaisons de lots de post-larves dé-fixées ou émergées, montrent que la cause principale de mortalité serait la dé-fixation. The king scallop (Pecten maximus L.) has been the object of some in-depth studies since 1973 with the double aim of natural resources management and extensive aquaculture in mind. The results obtained during the first ten years enabled the launch of a pluri-annual 1983-1987 program which aims to demonstrate the re-populating of the roadsted of Brest and the introduction of new management methods in the Bay of Saint-Brieuc. The technical results of the resource management and of the aquaculture operations are described. They enabled the stabilisation of the annual production of the Saint-Brieuc deposit as well as to foresee a new biomass of 80-100 tonnes of spawners per year in the roadsted of Brest. Effect of the probiotic strain Phaeobacter gallaeciensis after bacterial challenge on the complete larval development of Pecten maximus ArchiMer

The lipid content of spawned oocytes of Pecten maximus was studied by image analysis of semi -thin histological sections and a lipid index was defined. The female gametes were obtained by thermic stimulations of mature adults scallops from different populations after out of season conditioning or sampled in situ during the spawning season. After fertilization, the larval development was followed. The results reveal a large variability in lipid content of normal oocytes. The lipid content can be regarded as homogeneous inside a spawning but the heterogeneity is greater for wild scallops than for conditioned ones. Since 1973 some pectinides spat collection experiments have been conducted by the Fishery team of the Brittany Oceanology Centre, on two deposits exploited by commercial fishing: the roadsted of Brest and especially the St-Brieuc Bay The experiment carried out in 1973 was the first French attempt on this group of species and the results have been very encouraging: although the technology used was rudimentary, the best collectors enabled the collection of 30 to 50 king scallops (Pecten maximus) each and of 6,000 to 8,000 scallops (Chlamys varia and Chlamys opercularis). Toxicity to bivalve hemocytes of pathogenic Vibrio cytoplasmic extract ArchiMer

Using a chemiluminescence (CL) test, it had been previously demonstrated that Vibrio pectenicida, which is pathogenic to Pecten maximus larvae, was able to inhibit completely the CL activity of P, maximus hemocytes and partially inhibit those of Crassostrea gigas, Conversely, a Vibrio sp, strain, S322, pathogenic to C,gigas larvae was more active in reducing the CL activity of oyster hemocytes than of scallop hemocytes, Using this same CL biotest, V, pectenicida and S322 cytoplasmic extracts were shown to reproduce CL inhibition while the cytoplasmic extract of a nonpathogenic strain (U1, Pseudoalteromonas) was without effect. The assessment of the King scallop stock of the Bay of Seine (Pecten maximus) was carried out in July 2018, in the Bay of Seine sensu stricto, located in French territorial waters (from the Normandy coast to the south to the limit of 12 nautical miles to the North), as well as the bordering zone lying north of this 12-mile limit and up to a latitude of 49°48N to the North, called "Extérieur Baie de Seine". For this, a scientific survey, based on a stratified random sampling plan, was conducted aboard the F/R Thalia, a coastal research vessel of the French Oceanographic Fleet. To date, it has been impossible to fertilize the oocytes of ripe Pecten maximus obtained by gonad stripping as it is routinely done in hatchery with Crassostrea gigas . The events occurring between artificial induction of spawning and normal fertilization were studied experimentally in order to explain this difference. Fertilization and rearing trials were performed on oocytes obtained from the gonads of both species by scarification, by aspiration from genital ducts (of P. maximus only) and after spawning.
