Wildfire and Spatial Patterns in Forests in Northwestern Mexico: The United States Wishes It Had Similar Fire Problems Ecology and Society
Stephens, Scott L; ESPM Department University of California, Berkeley; stephens@nature.berkeley.edu; Fry, Danny L.; ESPM Department University of California, Berkeley; dfry@nature.berkeley.edu.
Knowledge of the ecological effect of wildfire is important to resource managers, especially from forests in which past anthropogenic influences, e.g., fire suppression and timber harvesting, have been limited. Changes to forest structure and regeneration patterns were documented in a relatively unique old-growth Jeffrey pine-mixed conifer forest in northwestern Mexico after a July 2003 wildfire. This forested area has never been harvested and fire suppression did not begin until the 1970s. Fire effects were moderate especially considering that the wildfire occurred at the end of a severe, multi-year (1999-2003) drought. Shrub consumption was an important factor in tree mortality and the dominance of Jeffrey pine increased after fire. The Baja California...

The contribution of fire damage to restricting high mountain Polylepis australis forests to ravines: Insights from an un-replicated comparison Ecología austral
Alinari,Julieta; von Müller,Axel; Renison,Daniel.
High mountain forests are often restricted to ravines and much debate has existed on the explanations of this distribution, with arguments ranging from abiotic site conditions being more favorable in ravines to there being reduced herbivory or reduced fre frequency and damage in ravines. We aim at understanding the contribution of fre damage and provide data to help test the hypothesis that fires are less damaging and that trees recover faster in ravines as compared to ridge sites. We evaluated crown damage, post-fire survival, regeneration by resprouts and seeds, and browsed stems in burned and unburned control Polylepis australis trees situated in a ravine and in a ridge forest in the west slope of the mountains of Central Argentina. The proportion of...

Induction of in vitro flowering in Capsicum frutescens under the influence of silver nitrate and cobalt chloride and pollen transformation Electron. J. Biotechnol.
Sharma,Ashwani; Kumar,Vinod; Giridhar,Parvatam; Ravishankar,Gokare Aswathanarayana.
The influence of silver nitrate (AgNO3) and cobalt chloride (CoCl2) on shoot multiplication and in vitro flowering in Capsicum frutescens Mill. was investigated. Exogenous administration of AgNO3 and CoCl2 at a concentration of 30 µM resulted in the maximum tissue response in terms of shoot length and number of shoots after 45 days culturing on MS medium. Both silver nitrate (40 µM) and cobalt chloride (30 µM) influenced in vitro flowering after 25 and 45 days respectively. This is the first report on in vitro flowering in C. frutescens. The study also demonstrated successful transformation of pollen obtained from the in vitro flowers. Since capsicum is highly recalcitrant to in vitro plant regeneration, the results of the study may be highly useful in...

In vitro culture response of barley (Hordeum vulgare) ethylene synthesis mutant MC 169 Electron. J. Biotechnol.
Biagioli,Carolina; Rios,Raúl; Basso,Alicia; Franzone,Pascual; Prina,Alberto.
Although it is generally accepted that plant in vitro culture response is influenced by the donor genotype, the genetic and molecular bases of this phenomenon are barely known. As a consequence, the optimization of tissue culture protocols is mainly empirically done. Researchers of the IGEAF studied the genetic basis of the in vitro regeneration of various plant species, including the tissue culture response of artificially induced barley mutants. One barley mutant, MC 169, carries a nuclear gene, recently described controlling the root growth in hydroponic cultivation. Under this condition, the roots of MC 169 mutant plants were longer than those of the original wild type line MC 182, a fact that was associated with a reduced ethylene biosynthesis. On the...

Expression of SOD transgene in pepper confer stress tolerance and improve shoot regeneration Electron. J. Biotechnol.
Chatzidimitriadou,Kiriaki; Nianiou-Obeidat,Irini; Madesis,Panagiotis; Perl-Treves,Rafael; Tsaftaris,Athanasios.
The objective of this work was to study the stress tolerance and regeneration capability of transgenic pepper plants carrying a sod gene, encoding a tomato chloroplast-localized Cu/Zn SOD protein. The expression of the sod gene was confirmed by enzymatic staining following polyacrylamide gel electrophoresis (PAGE), revealing a ‘novel’ band, which could represent a heterodimeric enzyme. Transgenic T1 and T2 progeny plants were exposed to different oxidative stresses including Methyl viologen (MV) and drought and found to have an increased resistance to oxidative damage. Furthermore, the SOD carrying transgenic pepper plants showed increased levels of regeneration efficiency compared to the wild type pepper plants. Pepper is a...

Optimization in Agrobacterium-mediated transformation of Anthurium andraeanum using GFP as a reporter Electron. J. Biotechnol.
Zhao,Qing; Jing,Ji; Wang,Gang; Wang,Jie Hua; Feng,Yuan Yuan; Xing,Han Wen; Guan,Chun Feng.
Although Agrobacterium-mediated transformation protocols for many economically important plant species have been well established, protocol for a number of flowering plants including Anthurium andraeanum remains challenging. In this study, we report success in generating transgenic Anthurium andraeanum cv Arizona using Agrobacterium GV3101 strain harboring a binary vector carrying gfp as a reporter gene. The possibility of facilitating the screening process for transgenic plants expressing functional proteins using gfp marker was explored. In order to realize high transformation efficiency, different explant sources including undifferentiated callus pieces and petioles were compared for their regeneration efficiency and susceptibility to...

Somatic embryogenesis and plant regeneration capacity in Argentinean maize (Zea mays L.) inbred lines Electron. J. Biotechnol.
González,Germán Andrés; Pacheco,María Gabriela; Oneto,Cecilia Décima; Etchart,Valeria J; Kandus,Mariana V; Salerno,Juan Carlos; Eyherabide,Guillermo; Presello,Daniel; Lewi,Dalia Marcela.
Somatic embryogenesis, which is still the method of choice for tissue culture, regeneration and transformation of maize, is largely considered highly genotype-dependent. The Hi II, a highly embryogenic genotype, has been extensively used in transformation protocols. However, this is not an inbred line; instead, it has a proportion of the undesirable A-188 background, and the progeny segregates for phenotypic characteristics and shows poor agronomic performance. In an effort to identify genotypes that combine a high somatic embryogenic response and good agronomic performance, we evaluated 48 advanced inbred lines developed at INTA. Callus development and somatic embryogenesis capacity were measured in 200 immature embryos per line. Embryogenic capacity [EC...

Isolation and regeneration of transiently transformed protoplasts from gametophytic blades of the marine red alga Porphyra yezoensis Electron. J. Biotechnol.
Takahashi,Megumu; Uji,Toshiki; Saga,Naotsune; Mikami,Koji.
Despite the recent progress of transient gene expression systems in a red alga Porphyra yezoensis by particle bombardment, a stable transformation system has yet to establish in any marine red macrophytes. One of the reasons of the difficulty in genetic transformation in red algae is the lack of systems to select and isolate transformed cells from gametophytic blades. Thus, toward the establishment of the stable transformation system in P. yezoensis, we have developed a procedure by which transiently transformed gametophytic cells were prepared from particle bombarded-gametophytic blade as regeneratable protoplasts. Using mixture of marine bacterial enzymes, yield of protoplasts was high as reported elsewhere; however, these protoplasts did not develop. In...

Response of root explants to in vitro cultivation of marketable garlic cultivars Horticultura Brasileira
Scotton,Danielle C; Benedito,Vagner Augusto; Molfetta,Jeanne B de; Rodrigues,Benedita Inês FP; Tulmann-Neto,Augusto; Figueira,Antonio.
Garlic cultivars are sexually sterile under standard growth conditions, with direct implications for commercial production costs as well as breeding programs. Garlic is propagated commercially via bulblets, which facilitates disease transmission and virus load accumulation over vegetative generations. Tissue culture produces virus-free clones that are more productive, while keeping the desired traits of the cultivar. Consequently, this technique allows studies of garlic genetics as well as guarantees genetic conservation of varieties. We aimed at analyzing the in vitro regeneration of eight marketable cultivars of garlic using root segments as explants. For each genotype, bulblet-derived explants were isolated and introduced into MS medium supplemented...

Shoot tip culture and thermotherapy for recovering virus-free plants of garlic Horticultura Brasileira
Torres,Antonio Carlos; Fajardo,Thor Vinícius; Dusi,André Nepomuceno; Resende,Renato de Oliveira; Buso,José Amauri.
Garlic shoot tip culture associated with dry heat thermotherapy (cloves exposed to 37°C for 35 days) were essential for recovering virus free plants of the cv Amarante. In this condition 70% of the explants developed in vitro and produced plants. A total of 77% of those plants was virus free when indexed by ISEM, which resulted in a final index of 54% of virus free plants from treated cloves. The percentage of regeneration decreased to 20% as the temperature increased up to 40°C. However 90% of those plants were virus free, leading to a final index of 18% virus free plants out of treated cloves.

Total mercury in the water column near the shelf edge of the European continental margin ArchiMer
Cossa, Daniel; Cotte Krief, Marie-hélène; Mason, Robert P.; Bretaudeau-sanjuan, Jane.
The purpose of this paper is to investigate the distribution and the relative importance of processes that affect mercury distribution in the water column at the shelf edge of the Celtic Sea, on the western European continental margin. The water column, down to 4500 m, was sampled during two cruises, one in winter 1994 and the other in June 1995, on eight stations. Total mercury concentrations ranged from 0.29 to 9.37 pM with the extreme values in the first 200 m. The highly variable concentrations in the surface layers are attributed to intense atmospheric-sea water exchange processes. The low concentrations encountered in the shelf water prelude any significant influence of continental waters in this area. In the mixed layer, biological uptake and...

Absorption et régénération de l'azote dans le système brassé de la Manche : productions nouvelle et régénérée ArchiMer
Maguer, Jf; L'Helguen, S; Madec, C; Le Corre, P.
Nitrate and ammonium uptake rates were measured in spring and summer in deep and shallow well-mixed waters of the English Channel during different cruises between 1986 and 1994. In the deep waters, nitrate uptake was relatively low during phytoplankton development and ammonium uptake represented more than 70 % of the total uptake irrespective of the season. In the shallow waters, nitrate uptake during spring phytoplankton growth was high and represented about 75 % of the total uptake. Ammonium uptake became substantial towards the end of spring and summer. The high contribution of ammonium to the nitrogenous nutrition of phytoplankton over the whole of the well-mixed waters is related to a high rate of recycling of nitrogen in the water column. Ammonium...

MatrigelBD, A Biocompatible Scaffold that Improves Gingival Mesenchymal Stem Cells Proliferation International Journal of Morphology
Brizuela,Claudia; SaintJean,Nicole; Garchitorena,Nicolas; Rodriguez,Ismael; Inostroza,Carolina.
MatrigelBD is a hydrogel scaffold with three-dimensional intercrossed networks of hydrophilic polymers with high water content. Human gingival tissue might represent a better source of MSCs, allowing these cells to be easily obtained in a relatively non-invasive way. The objective of this study was to evaluate the biocompatibility of MatrigelBD with GMSCs in vitro. Gingival connective tissue samples were obtained from healthy donors. Fresh tissue was minced and cultured during two weeks, after which cells at passage fourth were analyzed for their immune phenotype by flow cytometry. Differentiation into osteogenic, chondrogenic, and adipogenic lineages was induced and evaluated by culture staining. The "construct" was made of MatrigelBD with GMSC. To assess...
