The effect of combinations of the crude methanolic extract of the leaves of Helichrysum pedunculatum and eight first-line antibiotics were investigated by time kill assays against a panel of bacterial strains that have been implicated in wound infections. The plant extract showed appreciable antibacterial activities against the test bacteria with zones of inhibition ranging between 18 and 27 mm, and minimum inhibitory concentrations (MICs) varying between 0.1 and 5.0 mg/ml. The MICs of the test antibiotics range between 0.001 and 0.412 mg/ml, and combination of the plant extract and the antibiotics resulted in reduction of bacterial counts by between 0 and 6.63 Log10 cfu/ml. At V2 MIC, 56.81% synergy; 43.19% indifference and no antagonism were observed,... |

ABSTRACT The antifungal activity of tacrolimus in combination with antifungal agents against different fungal species has been previously reported. Here we report the in vitro interactions between tacrolimus and amphotericin B, fluconazole, itraconazole, and caspofungin against 30 clinical isolates of both fluconazole-susceptible and fluconazole-resistant Trichosporon asahii. For these analyses, we used the broth microdilution method based on the M27-A3 technique and checkerboard microdilution method. Tacrolimus showed no activity against T. asahii strains (minimal inhibitory concentrations, MICs > 64.0 µg mL−1). However, a larger synergistic interaction was observed by the combinations tacrolimus + amphotericin B (96.67%) and tacrolimus + caspofungin... |

This paper describes the effect of dipyridamole (DIP) on the cytotoxicity of cisplatin in HEp-2 human larynx cancer cells in vitro and the nature of the interaction between cisplatin and dipyridamole. Cytotoxic assays were performed to obtain the IC50 for cisplatin. The cells were treated with 0, 20, 40, 80, 120 or 200 µM cisplatin, with or without a single concentration of DIP and incubated for 60 min at 37ºC and 5% CO2 for 3 days and then counted with a hemocytometer. The accumulation of cisplatin in the cells was measured by atomic absorption and fluorescence was used to determine the membrane binding constant of DIP. In the presence of 10, 20 and 30 µM DIP, the IC50 of cisplatin was reduced by 25, 60 and 82% in HEp-2 cells. Combination index analysis... |

Diclofenac sodium, an antiinflammatory agent, exhibited remarkable inhibitory action against both drug sensitive and drug resistant clinical isolates of Mycobacterium tuberculosis, as well as other mycobacteria. This drug was tested in vitro against 45 different strains of mycobacteria, most of which were inhibited by the drug at 10-25 µg/ml concentration. When tested in vivo, diclofenac, injected at 10 µg/g body weight of a Swiss strain of white mice, could significantly protect them when challenged with 50 median lethal dose of M. tuberculosis H37 Rv 102. According to chi2 test, the in vivo data were highly significant (p<0.01). Diclofenac was further tested for synergism with the conventional antimycobacterial drug streptomycin against M. smegmatis... |

The treatment of brucellosis is still problematic, because of high rates of treatment failure or relapses. As the microorganism is an intracellular pathogen, treatment requires combined regimens. However, limited existing data on in vitro combinations are avaliable for Brucellae. The aim of this study was to investigate the in vitro efficacy of various traditional and new antibiotic combinations against 16 Brucella melitensis strains. The combination effect of antimicrobial agents was evaluated by E-test synergy method to obtain a fractional inhibitory concentration (FIC) index. Co-Trimoxazole (SXT) and moxifloxocin (MXF) exhibited the lowest MIC, while Rifampin (RIF) had the highest MIC in the study. Combinations with RIF showed the best synergistic... |

Jatropha curcas L. is a versatile crop since all its plant material is brought to use either as energy source, industrial or medicinal purpose. Several studies are ongoing in different parts of the world to optimize, enhance and exploit the growth, fruiting cycle and different developmental stages of the plant so that the economic yield of the plants can be utilized to the fullest limits. Foliar application of plant growth regulators such as ethrel (an ethylene releasing compound), indole acetic acid (IAA) and naphthalene acetic acid (NAA) at 50, 100 and 150 ppm, was found to influence different morpho-physiological characters in Jatropha curcas L, such as plant height, collar diameter, tree spread, flower initiation, number of inflorescence per plant,... |

ABSTRACT: Escherichia coli O157:H7 is a toxigenic serotype of E. coli and has been associated with foodborne outbreaks involving meat products, vegetables and fresh produces worldwide. Salts for curing are usually employed as antimicrobials in the production of pork sausages. However, salts do not have a significant inhibitory effect on enterobacteria. Due to the growing demand for natural foods, the use of essential oils has been proposed as natural antimicrobials in food. This study aimed to evaluate the effects of garlic essential oil (GO) and allyl isothiocyanate (AITC) against E. coli O157:H7 in vitro and in pork sausage. The Minimal Inhibitory Concentration (MIC) and Minimal Bactericidal Concentration (MBC) of these oils, alone and in combination,... |

ABSTRACT Thysanoptera species can transmit virus to Solanaceae family vegetables, including tomato plants for industrial processing, causing the disease known as Tomato Spotted Wilt Virus (TSWV). Thrips resistance to insecticides indicate the urgent need of techniques adequate for Integrate Pest Management practices. The objective was to evaluate the control efficiency (CE%) against Frankliniella schultzei (Thysanoptera: Thripidae) of the fungus Beauveria bassiana (Bb), the resistance inducer potassium silicate (KSil) and the chemical insecticide profenophos + cypermethrin (PC) isolated, or in binary mixtures. The treatments were foliar spraying on tomato plants with Bb (T1), KSil (T2) and PC (T3) isolated or in mixtures of Bb+KSil (T4), PC+KSil (T5) and... |

Peptaibols are small linear fungal peptides which are produced in the marine environment. They exhibit neurotoxicity by forming pores in neuronal membranes. This work describes their combine effect with domoic acid, a neurotoxic phycotoxin, on Diptera larvae. The Acute toxicity bioassay on this biological model was tested with a panel of different toxins (microbial, algal or fungal). It allowed the discrimination of neurotoxins and non-neurotoxic toxins, and an evaluation of the toxicity level (MED and ED50) which were correlated with published LD50 in mice for neurotoxins tested. The highest activities on this test were found for Na+ channel blockers tetrodotoxin (ED50 = 0.026 mg/kg) and saxitoxin (ED50 = 0.18 mg/kg). Domoic acid was less active with an... |

The marine ecosystem off British Columbia (BC), Canada, has experienced various changes in the last two decades, including reduced lipid-rich zooplankton biomass, increased marine mammals, and deteriorated commercial fisheries, particularly those targeting pelagic species such as Pacific Herring (Clupea pallasii). Understanding how stressors interactively and cumulatively affect commercially important fish species is key to moving toward ecosystem-based fisheries management. Because it is challenging to assess the cumulative effects of multiple stressors by using empirical data alone, a dynamic, individual-based spatially explicit ecosystem modeling platform such as Object-oriented Simulator of Marine Ecosystems (OSMOSE) represents a valuable tool to... |

There is an increasing need to understand community-level or whole-ecosystem responses to multiple stressors since the impacts of multiple stressors on marine systems depend not only on species- level responses, but also on species interactions and ecosystem structure. In this study, we used a multi-model ecosystem simulation approach to explore the combined effects of fishing and primary productivity on different components of the food-web across a suite of ecosystems and a range of model types. Simulations were carried out under different levels of primary productivity and various fishing scenarios (targeting different trophic levels). Previous work exploring the effects of multiple stressors often assumed that the combined effects of stressors are... |

Propolis is a natural resinous substance collected by bees from vegetal sources and its therapeutic properties have been investigated. In this work, we evaluated the inhibitory activity of ethanolic extracts of propolis (EEP) from the Southeast and South of Brazil on coagulase-negative Staphylococcus (CNS) growth as well as the EEP in vitro synergism with antimicrobial drugs by using the diffusion method (E-test). The EEP chemical characteristics (dry weight, pH, flavonoid and phenolic compounds) were determined. Seven drugs were tested, and synergism was observed between three drugs and Southeast EEP, six drugs and South EEP, and one drug and ethanol control. Ethanolic extracts of propolis from the South of Brazil presented the greatest flavonoid content... |
