ABSTRACT Poincianella pyramidalis (Tul.) L.P. Queiroz, is an arboreal species endemic to Brazil’s Caatinga biome and an important source of lumber and also of medicinal substances. This study aimed to evaluate the effect of the osmotic agents sucrose, sorbitol and mannitol and the growth retardant paclobutrazol (PBZ) on the induction of slow growth in vitro of P. pyramidalis, seeking to establish alternative techniques for ex situ conservation of the species. BSA and molecular markers screening for salt stress tolerant mutant of Petunia obtained in in vitro culture Ciência Rural

ABSTRACT: In this study, we performed BSA to identify genetic markers linked to salt tolerance. We tested the genetic diversity among four bulked DNA samples of EMS induced mutant clones and one bulked DNA sample of non-mutated clone of Petunia for salt tolerance in in vitro callus cultures using RAPD and ISSR markers. Out of the 36 RAPD and 16 ISSR primers identified, 25 and 13 were effectively used to amplify genomic DNA of all the five bulked samples, respectively. In total, 114 RAPD amplifications products were obtained, of which 28% were polymorphic and 2% were genotype-specific bands. Micropropagation of two species of foliage anthurium by direct organogenesis Ciência Rural

ABSTRACT: The objective of the present research was to develop a protocol for micropropagation of Anthurium bonplandii and Anthurium maricense by direct organogenesis. Nodal segments, with two or three nodes, were used as explants. The cultures were kept in a growth chamber at a temperature of 25±2ºC, under a photoperiod of 16 hours and a luminosity of 30μmol m-2 s-1. At 60 days, the number of regenerated buds per explant (NBE) was counted. The experiment was carried out in an entirely randomised design consisting of six treatments for six different concentrations of 6-benzylaminopurine (6-BA) added to the P2 (Pierik) medium (0.0, 1.11, 2.22, 3.33, 4.44, and 5.55µM). Abstract: The objective of this work was to evaluate the effects of temperature and osmotic agents on the in vitro conservation of Cape gooseberry (Physalis peruviana). Temperatures at 18 and 25°C, as well as the osmotic agents sucrose, mannitol, and sorbitol were tested. A short-term in vitro conservation of Cape gooseberry can be achieved at 18°C, using 30 g L-1 sucrose.
