Evaluation of gene expression profiling in a mouse model of L-gulonolactone oxidase gene deficiency Genet. Mol. Biol.
Yan,Jian; Jiao,Yan; Li,Xinmin; Jiao,Feng; Beamer,Wesley G.; Rosen,Cliff J.; Gu,Weikuan.
Humans and guinea pigs are species which are unable to synthesize ascorbic acid (vitamin C) because, unlike rodents, they lack the enzyme L-gulonolactone oxidase (Gulo). Although the phenotype of lacking vitamin C in humans, named scurvy, has long been well known, information on the impact of lacking Gulo on the gene expression profiles of different tissues is still missing. This knowledge could improve our understanding of molecular pathways in which Gulo may be involved. Recently, we discovered a deletion that includes all 12 exons in the gene for Gulo in the sfx mouse, characterized by spontaneous bone fractures. We report here the initial analysis of the impact of the Gulo gene deletion on the murine gene expression profiles in the liver, femur and...

Differential gene expression between wild-type and Gulo-deficient mice supplied with vitamin C Genet. Mol. Biol.
Jiao,Yan; Zhang,Jifei; Yan,Jian; Stuart,John; Gibson,Griffin; Lu,Lu; Willaims,Robert; Wang,Yong Jun; Gu,Weikuan.
The aim of this study was to test the hypothesis that hepatic vitamin C (VC) levels in VC deficient mice rescued with high doses of VC supplements still do not reach the optimal levels present in wild-type mice. For this, we used a mouse scurvy model (sfx) in which the L-gulonolactone oxidase gene (Gulo) is deleted. Six age- (6 weeks old) and gender- (female) matched wild-type (WT) and sfx mice (rescued by administering 500 mg of VC/L) were used as the control (WT) and treatment (MT) groups (n = 3 for each group), respectively. Total hepatic RNA was used in triplicate microarray assays for each group. EDGE software was used to identify differentially expressed genes and transcriptomic analysis was used to assess the potential genetic regulation of Gulo...

Antioxidant characterization of different italian broccoli landraces Horticultura Brasileira
Nicoletto,Carlo; Santagata,Silvia; Pino,Silvio; Sambo,Paolo.
ABSTRACT: The Brassicaceae plants are among the most consumed vegetables in the world. They feature a large biodiversity, in which landraces and primitive cultivars still play a major role on the cultivation systems of many countries. Brassicas and especially broccoli are closely linked to antioxidant compounds that play a key role for human health. This experiment aimed to characterize some Italian unknown broccoli landraces from the antioxidant point of view. Five landraces named BF, BF4, BFT, BB and BS were evaluated. The total antioxidant properties of broccoli were significantly different among ecotypes and the higher values were recorded in BS and BF4. Total phenols showed the higher values in BF4 and concerning the ascorbic acid content found in the...

Baudin Laurencin, F; Messager, J; Stephan, Gwendal.
Symptoms recalling a nutritional pathology and more precisely a process of lipid peroxydation were observed during the last years in seabass Dicentrarchus labrax and other fishes cultured in tropical marine condition, Ocyurus chrysurus and Lutjanus analis . They included dark coloration, skin ulceration, lethargy, anorexy, emaciation. Histological examinations showed hepatic fatty degenerative lesions, pancreatitis, muscular degeneration and retinal atrophy. Additional vitamins E and C in the food suppressed that pathological symptoms. The previously evoked clinical or histological signs were not achieved in spite of an increase of the level of hepatic, muscular or blood malondialdehyde and of the conjugated dienes of the perivisceral fat. In the same...

Vitamin C Deficiency Fails to Protect Mice from Malaria OAK
HERBAS, Maria Shirley; SUZUKI, Hiroshi; 鈴木, 宏志.
Nutritional deficiencies are frequent in malaria-endemic areas. It seems that micronutrient antioxidants play an important role in malaria parasite’s proliferation. Thus, the effect of vitamin C deficiency on malaria infection was examined in mice. When vitamin C deficient mice, L-gulono-γ-lactone oxidase gene knockout mice which are unable to synthesize ascorbic acid, were infected with a lethal dose of Plasmodium berghei NK65-infected red blood cells, the knockout mice showed similar parasitemia kinetics and survival rates as wild-type mice. The results indicate that deficiency of vitamin C might not affect the development of the malaria parasite in mice.

Variation in egg and larval quality in various fish and crustacean species OMA
Lavens, P.; Sorgeloos, P..
It is generally accepted, that a major constraint in the further development of marine fish and crustacean aquaculture, is the variable quality of eggs and larvae used for the hatchery-production of fry. Nonetheless, valid criteria to evaluate optimal egg quality have not been identified yet. Instead, particular indicators, are used, e.g. the fertilization and hatching rate, overall culture success during the first rearing period, etc., but these are not totally objective because hatchery-specific conditions may interfere. The biochemical composition of the egg may be one of the possible determinants of egg quality, which can be evaluated on a more objectie basis. In this respect, studied have been initiated to analyze in freshly-released eggs, the levels...

Experiences on importance of diet for shrimp postlarval quality OMA
Lavens, P.; Sorgeloos, P..
This paper gives an overview of own experiences with respect to the impact of dietary conditions on the quality of postlarval penaeid shirmp. Direct nutrient-specific effects are documented for highly unsaturated fatty acid (HUFA), phosphatidyl choline (PC) and ascorbic acid (AA). Specific immunostimulants may also contribute to an improved stress resistance of the shrimp fry. Last, but not least, some emphasis is placed on the role of the microflora during the hatchery rearing, the possible reduction of bacteria in the live food adminstered, and the use of probionts through the diet.

The effect of supplemental ascorbic acid in enriched live food for <i>Clarias gariepinus</i> larvae at startfeeding OMA
Merchie, G.; Lavens, P.; Verreth, J.; Ollevier, F.P.; Nelis, H.; De Leenheer, A.P.; Storch, V.; Sorgeloos, P..
The effect of three dietary ascorbic acid (AA) concentrations, each applied via two feed types, on production characteristics and physiological condition of African catfish (<i>Clarius gariepinus</i>) larvae has been assessed in two 10-day culture trials. Three treatments received only <i>Artemia</i> nauplii enriched with an experimental emulsion containing 0, 10 or 20% ascorbyl palmitate (AP) and yielding 530, 1200 and 1600 µg AA g-1 DW <i>Artemia</i> , respectively; the other three treatments were fed the same <i>Artemia</i> diets which were partially substituted by an artificial diet containing no vitamin C (ratio 20:80). No differences in survival could be observed: however, from...

Optimization of dietary vitamin C in fish and crustacean larvae: a review OMA
Merchie, G.; Lavens, P.; Sorgeloos, P..
HPLC techniques were adapted and standardized for quantification of ascorbic acid (AA) and its derivates in both diets and target organisms. To assess the dietary needs for AA at start of exogenous feeding, the AA content in the various live diets currently used in aquaculture (algae, rotifers, <i>Artemia</i> ) was analyzed. Application of techniques for boosting vitamin C using ascorbyl palmitate as the source enabled the transfer of elevated levels (up to 2,500 µg AA/g DW) of bioactive vitamin C. Larvae of fish (<i>Clarias gariepinus, Dicentrarchus labrax, Scophthalmus maximus</i>), white shrimp (<i>Penaeus vannamei</i>) and prawn (<i>Macrobrachium rosenbergii</i>) were enriched via the live food chain....
