Co-evolution model of Colletotrichum lindemuthianum (melanconiaceae, melanconiales) races that occur in some Brazilian regions Genet. Mol. Biol.

Colletotrichum lindemuthianum, the causal agent of anthracnose in the common bean (Phaseolus vulgaris L.), displays a high level of virulence diversity, which explains the large number of existing pathotypes. Several lines of evidence indicate that such diversity is, at least in part, due to plant and pathogen co-evolution. A co-evolution model based on the binary classification of 25 races identified in Brazil by inoculation of differential cultivars and random amplified polymorphic DNA (RAPD) data is proposed. In this model, races 8 and 64 that infected bean cultivar Cornell 49-242 (Are gene) and Mexico 222 (Mexico I gene) are considered to be sources of two important evolutionary routes. Inferences about undescribed races from Brazil could be made.

Transferability and characterization of microsatellite markers in two Neotropical Ficus species Genet. Mol. Biol.

Microsatellite markers were transferred and characterized for two Neotropical fig tree species, Ficus citrifolia and Ficus eximia. Our study demonstrated that microsatellite markers developed from different subgenera of Ficus can be transferred to related species. In the present case, 12 of the 15 primer pairs tested (80%) were successfully transferred to both of the above species. Eleven loci were polymorphic when tested across 60 F. citrifolia and 60 F. eximia individuals. For F. citrifolia, there were 4 to 15 alleles per locus, whereas expected heterozygosities ranged from 0.31 to 0.91. In the case of F. eximia, this was 2 to 12 alleles per locus and expected heterozygosities from 0.42 to 0.87.

Genetic characterization of 12 heterologous microsatellite markers for the giant tropical tree Cariniana legalis (Lecythidaceae) Genet. Mol. Biol.

Twelve microsatellite loci previously developed in the tropical tree Cariniana estrellensis were genetically characterized in Cariniana legalis. Polymorphisms were assessed in 28 C. legalis individuals found between the Pardo and Mogi-Guaçu River basins in the state of São Paulo, Brazil. Of the 12 loci, 10 were polymorphic and exhibited Mendelian inheritance. The allelic richness at each locus ranged from 2-11, with an average of 7 alleles per locus, and the expected heterozygosity ranged from 0.07-0.88. These loci showed a high probability of paternity exclusion. The characteristics of these heterologous microsatellite markers indicate that they are suitable tools for investigating questions concerning population genetics in C. legalis.

Identification of a RAPD marker linked to the Co-6 anthracnose resistant gene in common bean cultivar AB 136 Genet. Mol. Biol.

The pathogenic variability of the fungus Colletotrichum lindemuthianum represents an obstacle for the creation of resistant common bean (Phaseolus vulgaris L.) varieties. Gene pyramiding is an alternative strategy for the development of varieties with durable resistance. RAPD markers have been proposed as a means to facilitate pyramiding of resistance genes without the need for multiple inoculations of the pathogens. Diversidade genética, endogamia e fluxo gênico em pequena população fragmentada de Copaifera langsdorffii Rev. Bras. Bot.

O objetivo deste trabalho foi investigar a taxa de imigração e a distância de dispersão de pólen e sementes em uma pequena população fragmentada de Copaifera langsdorffii Desf., com base no polimorfismo de oito locos microssatélites. Foram mapeadas e genotipadas todas as 47 árvores adultas e 65 subadultas existentes no fragmento. Adultos tinham um total número menor de alelos (142) do que subadultos (164), sendo 20 alelos exclusivos aos adultos e 41 aos subadultos. Adultos tinham significativamente menor diversidade genética e maior endogamia (Â = 17,75 ± 0,51; Âe = 8,97 ± 0,27; ^F = 0,191 ± 0,017) do que subadultos (Â = 20,5 ± 0,65; Âe = 10,86 ± 0,29; ^F = 0,119 ± 0,013). Genetic variability and pedigree analysis of Brazilian common bean elite genotypes Scientia Agricola

Genetic diversity is essential for any breeding program. However, breeders tend to concentrate on specific genotypes, which combine traits of interest and may be used as progenitors in several breeding programs. Common bean (Phaseolus vulgaris L.) breeding programs are not different in this sense. In this study, the genetic diversity of 21 common bean elite lines from the Bean Regional Trials conducted by the Embrapa Rice and Bean Research Center was evaluated using the Random Amplified Polymorphic DNA (RAPD) and pedigree analyses. ABSTRACT: Stylosanthes capitata Vogel and Stylosanthes macrocephala M.B.Ferreira & Sousa Costa are two forage leguminous species of agronomic importance for animal husbandry in tropical environments. The physical mixture of both species (80 % S. capitata and 20 % S. macrocephala) comprises the commercial cultivar “Estilosantes Campo Grande”. However, proximity of fields for seed production may contaminate seed lots, compromising seeds quality. A transferência de alelos de resistência a doenças em plantas pode ser facilitada pelo uso de marcadores moleculares do DNA. Se proximamente ligados a alelos de resistência, eles podem ser usados na seleção assistida por marcadores (S.A.M.). Uma aplicação concreta dos marcadores na S.A.M. é durante o processo de piramidação de alelos de resistência. An efficient and rapid DNA minipreparation procedure suitable for PCR/SSR and RAPD analyses in tropical forest tree species BABT

An efficient and rapid DNA minipreparation modified method for frozen samples was developed for five tropical tree species: Copaifera langsdorffii, Hymenaea courbaril, Eugenia uniflora, Tabebuia roseo alba and Cariniana estrellensis. This procedure that dispenses the use of liquid nitrogen, phenol and the addition of proteinase K, is an adaptation of the CTAB-based DNA extraction method. The modifications included the use of PVP to eliminate the polyphenols, only one chloroform-isoamyl alcohol step and the addition of RNase immediately after extraction with chloroform. Use of RAPD-PCR to identify true hybrid plants from crosses between closely related progenitors Braz. J. Genet.

RAPD-PCR molecular markers were used to identify common bean and soybean hybrid plants derived from crosses between closely related progenitors, with no apparent phenotypic differences. Primers OP-F12 and OP-0O3 were used to identify true hybrids derived from crosses between common bean cultivars Rudá (A 285) and AN 910408, and soybean cultivars Cristalina and Bossier, respectively. Each primer generated one polymorphic DNA band which was present in the male progenitor and absent in the female progenitor. As RAPD bands are normally inherited as dominant characters, the presence of these bands in the F1 plants confirmed their status.
