Molecular confirmation of Maize rayado fino virus as the Brazilian corn streak virus Scientia Agricola

Maize rayado fino virus (MRFV), present in various countries in Latin America, has shown similarities to corn streak virus that occurs in Brazil, regarding pathogenic, serological and histological characteristics. In the current report both virus were molecularly compared to confirm the similarities between them. MRFV was identified by nucleic acid hybridization in samples of maize tissues exhibiting symptoms of "corn stunt" disease, collected from two Brazilian States - São Paulo and Minas Gerais. Phytoplasma associated with shoot proliferation in begonia Scientia Agricola

Begonia is a very appreciated genus of ornamental plants, of economic relevancy, having species of flowers and foliage. In commercial croppings, plants exhibiting characteristic symptoms of phytoplasma infection have been observed, such as shoot proliferation, reduced plant, size small leaves and flowers, and phyllody. Leaves were sampled and total DNA was extracted to be used in nested Polymerase Chain Reaction (PCR), in order to detect and identify an expected phytoplasma. Sesame phyllody associated with a 16SrI-B phytoplasma, a ‘Candidatus Phytoplasma asteris’-related strain, in Paraguay Scientia Agricola

ABSTRACT: Sesame (Sesamum indicum L.) plants exhibiting symptoms of phyllody disease were observed in commercial fields in Paraguay. The symptoms were indicative of infection by phytoplasmas. Thus, the present study investigated the association between affected plants and phytoplasma, which was later analyzed using molecular and phylogenetic methods. Total DNA was extracted from symptomatic and asymptomatic plants and used in nested PCR assays using primers R16SN910601/R16SN011119 and R16F2n/16R2. Molecular characterization of a phytoplasma associated with a commercial variety of Momordica charantia Scientia Agricola

ABSTRACT: Momordica charantia (bitter melon) presents two distinct types or varieties, known as wild type and commercial type. Plants of the wild type are hosts of a phytoplasma of the group 16SrIII-J, which is associated with a disease known as witches’ broom. However, this disease has not yet been reported in commercial bitter melon. Thus, symptomatic plants of the commercial type were analyzed in order to demonstrate the association between phytoplasmas and disease. Efeito inibitório de extratos vegetais sobre Colletotrichum gloeosporioides - agente causal da podridão de frutos de mamoeiro Scientia Agricola

Extratos aquosos foram obtidos a partir de bulbilhos de alho, folhas de hortelã e mamona e frutos de pimenta. Após a incorporação destes extratos em BDA, obtendo-se concentrações de 100, 200, 500, 1000, 5000 e 10000 ppm, foi avaliado o crescimento e a produção de conídios de um isolado de Colletotrichum gloeosporioides, agente de podridão em frutos de mamoeiro. Os resultados demonstraram o efeito inibitório dos diferentes extratos a partir da concentração de 200 ppm. O extrato de alho inibiu o crescimento micelial, em porcentagens variáveis de 5,3 a 67,6%, porém não atuou de modo expressivo sobre a produção de conídios. Control of anthracnose in green pepper involves the use of resistant varieties and/or fungicides. The selection of varieties and efficient products demands great amounts of conidia as inoculum. It is thus necessary to optimize the production of Colletotrichum gloeosporioides conidia in the laboratory, establishing the best conditions for fungus development. Bico-de-papagaio é uma espécie ornamental, encontrada tanto na forma de arbustos, presente em jardins residenciais e públicos, como comercializada em vasos para decoração de interiores. Com o objetivo de detectar a presença de fitoplasma em plantas mostrando sintomas de proliferação de ramos, amostras foram obtidas de plantas sintomáticas e assintomáticas cultivadas em jardins e de plantas comerciais sintomáticas envasadas. A técnica de duplo PCR, utilizando os iniciadores R16mF2/mR1 e R16F2n/R2, foi empregada para detecção do fitoplasma, a partir de DNA extraído de folhas.
