Production of an extracellular keratinase from Chryseobacterium sp. growing on raw feathers Electron. J. Biotechnol.

The strain Chryseobacterium sp. kr6 shown to be useful for biotechnological purposes such as hydrolysis of poultry feathers and de-hairing of bovine pelts. The effect of temperature, initial pH and media composition on protease production by this keratinolytic strain was studied. The enzyme was produced between 25 and 37ºC, with maximum activity and yield at 30ºC. When protease production was tested in media with different initial pH, maximum activity was observed when cultivation was carried out at 30ºC and initial pH ranging from 6.0 to 8.0. Higher activity was observed when feathers or feather meal were used as growth substrates, followed by soybean meal. The presence of sboA and spaS genes and antimicrobial peptides subtilosin A and subtilin among Bacillus strains of the Amazon basin Genet. Mol. Biol.

This report demonstrates the usefulness of PCR for the genes spaS and sboA as a means of identifying Bacillus strains with a potential to produce subtilin and subtilosin A. One collection strain and five Bacillus spp. isolated from aquatic environments in the Amazon basin were screened by PCR using primers for sboA and spaS designed specifically for this study. The sequences of the PCR products showed elevated homology with previously described spaS and sboA genes. Antimicrobial peptides were isolated from culture supernatants and analyzed by mass spectrometry. Efeito do hidrorresfriamento na qualidade de pêssegos 'Chimarrita' e 'Chiripá' Rev. Bras. Frutic.

No presente trabalho, foram avaliados os efeitos do hidrorresfriamento para manutenção da qualidade em pêssegos 'Chimarrita' e 'Chiripá'. O hidrorresfriamento foi realizado por imersão em água com gelo a 5ºC, sendo o abaixamento da temperatura monitorado por 3 minutos. O tratamento-controle foi transferência para a câmara fria imediatamente após a colheita dos pêssegos. Os frutos foram armazenados a 0ºC ('Chiripá') e 0,5ºC ('Chimarrita') por 7; 14 e 21 dias e avaliados na colheita, nas saídas de frio, seguidos de 2 a 3 dias a 20ºC para o amadurecimento. Efficacy of modified atmosphere packaging to control Sitophilus spp. in organic maize grain BABT

The effect of modified atmosphere packaging on the mortality of Sitophilus spp. in organic maize grain was studied. Maize grains were packed with five different atmospheres consisting of 20% O2, or 20, 40, 60, 80% CO2 with remaining N2. The packages were stored for 30 days at 26°C. CO2 and O2 concentrations were monitored inside the packages during the storage. The moisture content, titratable acidity and pH were analysed on the first and thirtieth day. At 5, 15 and 30 days of storage no significant differences were observed in the percentages of dead insects when CO2 was used. Production of yeast extract from whey using Kluyveromyces marxianus BABT

The yeast Kluyveromyces marxianus CBS 6556 was grown on whey to produce nucleotide-rich yeast extracts. Thermal treatments of cells at 35 or 50ºC for 15-30h resulted in yeast extracts containing about 20 g/L protein, with only the second treatment resulting in the presence of small amounts of RNA. In contrast, autolysis in buffered solution was the unique treatment that resulted in release of high amounts of intracellular RNA, being, therefore, the better procedure to produce 5'-nucletide rich extract with K. marxianus.

Production of bacteriocin-like substances by lactic acid bacteria isolated from regional ovine cheese BJM

Lactic acid bacteria (LAB) were isolated from ovine milk and cheeses manufactured in the South Region of Brazil. Among 112 bacterial isolates investigated, 59 were chosen through a screening for LAB. Among these 59 strains of LAB, 21% showed antimicrobial, proteolytic and lipolytic activities. Based on this screening, Lactobacillus plantarum LCN 17 and Lactobacillus rhamnosus LCN 43 were selected and tested for the production of bacteriocin-like substances (BLS). The BLS produced by both isolates showed antimicrobial activity against Listeria monocytogenes, whereas that produced by L. plantarum LCN 17 presented higher stability to different temperature, pH and enzyme treatments. Screening for antimicrobial activity among bacteria isolated from the Amazon Basin BJM

Bacteria producing antimicrobial activity were identified among 86 isolates from aquatic environments of Brazilian Amazon Basin. Antimicrobial activity against at least one indicator strain was detected for 59 isolates (68.6%). Inhibitory activity was mostly against Gram-positive bacteria, such as Listeria monocytogenes and Bacillus cereus. The antimicrobial substances produced by 19 strains that showed higher inhibitory activity were partially characterized. These antimicrobial substances showed thermal resistance for up to 100°C and partial resistance to proteolytic treatment. P34 is an antimicrobial peptide produced by a Bacillus sp. strain isolated from the intestinal contents of a fish in the Brazilian Amazon basin with reported antibacterial activity. The aim of this work was to evaluate the peptide P34 for its in vitro antiviral properties against canine adenovirus type 2 (CAV-2), canine coronavirus (CCoV), canine distemper virus (CDV), canine parvovirus type 2 (CPV-2), equine arteritis virus (EAV), equine influenza virus (EIV), feline calicivirus (FCV) and feline herpesvirus type 1 (FHV-1). Antimicrobial susceptibility, hemolysis, and hemagglutination among Aeromonas spp. isolated from water of a bovine abattoir BJM

The presence of Aeromonas spp. in food has been demonstrated. They are often introduced from water, which is the natural habitat of many species and is thought to be the main source of contamination. The occurrence of Aeromonas spp. was investigated in 70 water samples of a bovine abattoir. Aeromonas spp. were present in 21.4% of water samples. A. hydrophila was isolated from 11.4% of supplying water samples and from 25.7% of the water drained from carcasses, whereas A. sobria was sole isolated from 5.7% of supplying water samples. Phenotypic and genotypic characterization of Salmonella Enteritidis isolates BJM

In order to study the epidemiology of Salmonella Enteritidis outbreaks and determine the source of contamination so that a recurrence can be avoided, detailed characterization is necessary. Thus, the purpose of this study was to verify whether rep-PCR was able to discriminate among Salmonella Enteritidis isolates. Inhibition of listeria monocytogenes in minas frescal cheese by free and nanovesicle-encapsulated nisin BJM

The effectiveness of free and nanovesicle-encapsulated nisin to control Listeria monocytogenes in Minas Frescal cheese was investigated. Commercial nisin was encapsulated into liposomes of partially purified soy lecithin. Free (0.1 mg/mL and 0.25 mg/mL) and nanovesicle-encapsulated nisin (0.25 mg/mL) were applied onto the surface of cheese samples, and L. monocytogenes was inoculated before incubation at 6-8°C for 28 days. A bactericidal effect was observed with 0.25 mg/mL free nisin; a bacteriostatic effect was observed for liposome-encapsulated nisin and 0.1 mg/mL free nisin. Hydrolytic potential of a psychrotrophic Pseudomonas isolated from refrigerated raw milk BJM

The production of extracellular hydrolases by a psychrotrophic bacterium isolated from refrigerated raw milk, and identified as a Pseudomonas sp. belonging to the Pseudomonas jenssenii group, was studied. This bacterium produced proteolytic and lipolytic enzymes in all media investigated (skim milk, cheese whey, casein broth, and tryptone soy broth). High levels of α-glucosidase were produced in skim milk broth. Hydrolytic enzymes detected in skim milk broth are of particular concern, indicating that these enzymes could be produced by Pseudomonas sp. during the cold storage of raw milk, contributing to the spoilage problem in milk and dairy products.

Detection of virulence genes in Salmonella Enteritidis isolated from different sources BJM

The presence of three virulence genes, invA, spvR, and spvC, was determined in Salmonella Enteritidis isolated from poultry, pigs, humans and food. All isolates were positive for the invA gene, with 91.2% being positive for spvR and 90.2% for spvC. There was no significant difference in the prevalence of the virulence genes between isolates from different sources. The results indicate that there is a putative high virulence potential for the S. Enteritidis isolates characterized.

Antibacterial activity of bacteriocin-like substance P34 on Listeria monocytogenes in chicken sausage BJM

The antimicrobial activity of the bacteriocin-like substance (BLS) P34 against Listeria monocytogenes was investigated in chicken sausage. The BLS was applied to chicken sausages (256 AU g-1) previously inoculated with a suspension of 10² cfu g-1 of L. monocytogenes. BLS P34 inhibited the indicator microorganism in situ in all incubation times for up to 10 days at 5 °C. The effectiveness of BLS P34 was increased when it was added in combination with nisin. The bacteriocin was also tested in natural eatable natural bovine wrapping (salty semi-dried tripe) against the same indicator microorganism, also showing inhibitory capability in vitro. BLS P34 showed potential to control L. monocytogenes in refrigerated meat products.

Adhesion and biocides inactivation of Salmonella on stainless steel and polyethylene BJM

The adhesion of Salmonella (S.) strains to stainless steel and polyethylene and their inactivation by biocides used in food industry was investigated. Coupons of stainless steel and polyethylene were immersed in bacterial suspensions of S. Enteritidis, S. Typhimurium, and S. Bredeney during 15, 30, and 60 minutes, and submitted to different concentrations of peracetic acid (PAA), sodium hypochlorite (NaOCl), and quaternary ammonium (Quat) sanitizers. Keratinolytic bacteria isolated from feather waste BJM

The aim of this study was to characterize keratinolytic bacteria isolated from feather waste. Four isolates were selected after growth on solid medium with feather meal as sole carbon and nitrogen source and screened for proteolytic activity on milk agar plates. Three isolates were Gram-negative (belonged to the genera Burkholderia, Chryseobacterium and Pseudomonas) and one was Gram-positive (Microbacterium sp.). Production of crystallized fruit from watermelon rind Ciencia e Investigación Agraria

The aim of this work was to produce crystallized fruit from watermelon rind. The following procedure was developed: the outer peel was removed; the material was sliced into 7 mm cubes, blanched for, 0, 5 and 10 minutes, and then treated with 10% sodium chloride solution. This product was treated with solutions of sucrose (30 to 72 °Brix), and dried in a hot air dryer at different temperatures (40, 60 and 80 °C). Products were then analyzed by a sensory panel. The experimental design used was randomized blocks and the results were analyzed by the Tukey's test. The best acceptance of the sensory panel was for the product obtained by 5 minutes blanching followed by drying at 60 °C, whose intensity values for appearance, flavor and gummosis were between 6 and 7.

Activity of the antimicrobial peptide P34 against bovine alphaherpesvirus type 1 Ciência Rural

ABSTRACT: Previous studies have demonstrated the antimicrobial activity of the peptide P34. In this study, the antiviral potential of P34 and the in vitro mechanism of action were investigated against bovine alphaherpesvirus type 1 (BoHV1). P34 exhibited low toxicity, a high selectivity index (22.9) and a percentage of inhibition of up to 100% in MDBK cells. Results from antiviral assays indicated that P34 did not interact with cell receptors, but it was able to inhibit the viral penetration immediately after pre-adsorption. In addition, BoHV1 growth curve in MDBK cells in the presence of P34 revealed a significant reduction in virus titer only 8h post-infection, also suggesting an important role at late stages of the replicative cycle. Influence of post-mortem aging in tenderness of chicken breast fillets Ciência Rural

In this work, analysis of chicken breast fillets was performed for shear values and sensory evaluation after different times of aging. Aging process was carried out by holding carcasses in covered plastic boxes, in a cold room, at 3 +/- 1ºC; groups of 5 carcasses were stored and classified according to the period of storage in cold room as 0 (control), 2, 4, 8, 12 and 24 hours. The variation in pH of chicken carcasses during rigor decreased from 6.4 to 5.9. However, during the process of aging, the pH values increased up to 6.2 in the first 12h, probably as the result of proteolysis.
