Comparação de três técnicas utilizadas no diagnóstico laboratorial de cinomose canina MV&Z
Gamon, T. H. M.; Batista, H. B. C. R.; Cruz, F. P. N.; Peixoto, Z. M. P.; Carnieli Jr, P.; Oliveira, R. N.; Nasruai, A. C. R.; Castilho, J. G..

Experimental antiviral therapy against different rabies virus lineages using transfection with anti-rabies antibodies MV&Z
Castilho, J. G.; Batista, H. B. C. R.; Rodrigues, A. C.; Carnieli Jr, P.; Oliveira, R. N.; Silva, A. C. R.; Caporale, G. M. M.; Carrieri, M. L.; Kotait, I.; Brandão, P. E..
The aim of this study was to develop a new mechanism for antiviral therapy against rabies based on the introduction by transfection with a cationic reagent (lipofectamine 2000) of antibodies into neuronal cells infected with the rabies virus. N2A cells were infected using 96-well plates and different viral concentrations (0.1, 1.0, 10 and 100 TCID50) of three lineages of rabies virus circulating in Brazil (dog, Desmodus rotundus and Eptesicus furinalis). After incubation for 24 h, the cells were transfected with antirabies- virus polyclonal antibodies and lipofectamine 2000. These cells made up the treatment group (TG). The cells in the negative control group (CG) were treated with only Minimum Essential Medium. After 11 hours, the plates were fixed with...

Canine and feline rabies in the state of São Paulo – data from the Pasteur Institute of São Paulo, 1999-2012 MV&Z
Carrieri, M. L.; Achkar, S. M.; Castilho, J. G.; Oliveira, R. N.; Carnieli Jr, P.; Macedo, C. I.; Omoto, T. M.; Takaoka, N. Y.; Kotait, I..
The São Paulo State Rabies Control Program, coordinated by the Pasteur Institute of São Paulo, has been involved in endeavors to improve the epidemiological surveillance of rabies in dogs, cats and bats, particularly in urban areas. Since 1998, when the last case of canine rabies due to antigenic variant 2 occurred in the state, the Institute has received 66,352 canine central nervous system samples, 17,796 samples from cats and 38,133 from bat specimens. Using conventional techniques for rabies diagnosis, 11 dogs, 8 cats and 554 bats were found to be positive for this disease. When molecular techniques, such as antigenic typing and genetic sequencing, were used, antigenic variant 3, which is associated with Desmodus rotundus, was detected in 11 dogs and 7...

Phylogeography of the rabies virus isolated from canids in the North and Northeast of Brazil MV&Z
Carnieli Jr, P.; Oliveira, R. N.; Castilho, J. G.; Vieira, L. F. P..
Antigenic variant 2 of the Rabies Virus (RABV), typically found in dogs in Brazil, still circulates in the North and Northeast of Brazil in spite of mass vaccination campaigns against rabies in domestic animals. Previous studies have shown that this variant is made up of two main genetic lineages: one found in dogs (Canis familiaris) and another typically found in wild canids, particularly the crab-eating fox (Cerdocyon thous), which is known in Brazil as cachorro do mato. C. thous is currently the only wild land species in which RABV is continually isolated, with an average of 20 cases/year. Although previous studies and phylogenetic analyses of the N and G genes and intergenic G-L region all confirmed the existence of these two genetic lineages, the way...

Development of real time RT-PCR (TAQMAN) for detection and genetic characterization of antemortem human rabies MV&Z
Mori, E.; Scheffer, K. C.; Fahl, W. O.; Oliveira, R. N.; Asano, K. M.; Carnieli Jr, P.; Carrieri, M. L.; Kotait, I..
Human rabies is still an important public health problem in some Brazilian regions. Usually, ante-mortem diagnosis of rabies is made by demonstration of virus antigen by direct immunofluorescence in corneal or conjunctival smears and skin biopsies; however, this technique has a low sensitivity. Recently, molecular techniques such as the reverse transcriptase polymerase chain reaction (RT-PCR) and nucleic acid sequence based amplification assay (NASBA) have been developed to improve the sensitivity and specificity of ante as well as post-mortem diagnosis of rabies. Rapid and accurate diagnosis of ante-mortem human rabies is essential for effective medical management and to ensure appropriate post-exposure prophylaxis of potential contacts with the patient....
