Bats are considered important reservoirs of rabies virus, which is paramount in the study of the pathogenesis of this zoonosis disease, through the use of sensitive systems. The presence of viral antigen in these species shows that non-neuronal viral spread is efficient in different organs that participate effectively in the elimination of rabies virus, such as, salivary glands and bladder. Molecular techniques have been used increasingly as tools for the diagnosis by detecting the rabies virus genome. This study aimed to detect the presence of rabies virus in the wash of skull and in different organs of the genus Artibeus bats using the hemi-nested RT-PCR (hnRT-PCR) and Real Time RT-PCR molecular techniques. The identification of animal species that transmit pathogens such as the rabies virus is of the utmost importance for public health and the natural history of infectious and contagious diseases. Diagnostic laboratories very often receive mauled or decomposing animal carcasses, particularly of bats, rendering morphometric identification unviable. The São Paulo State Rabies Control Program, coordinated by the Pasteur Institute of São Paulo, has been involved in endeavors to improve the epidemiological surveillance of rabies in dogs, cats and bats, particularly in urban areas. Since 1998, when the last case of canine rabies due to antigenic variant 2 occurred in the state, the Institute has received 66,352 canine central nervous system samples, 17,796 samples from cats and 38,133 from bat specimens. The aim of this study was to develop a new mechanism for antiviral therapy against rabies based on the introduction by transfection with a cationic reagent (lipofectamine 2000) of antibodies into neuronal cells infected with the rabies virus. N2A cells were infected using 96-well plates and different viral concentrations (0.1, 1.0, 10 and 100 TCID50) of three lineages of rabies virus circulating in Brazil (dog, Desmodus rotundus and Eptesicus furinalis). Human rabies is still an important public health problem in some Brazilian regions. Usually, ante-mortem diagnosis of rabies is made by demonstration of virus antigen by direct immunofluorescence in corneal or conjunctival smears and skin biopsies; however, this technique has a low sensitivity. Recently, molecular techniques such as the reverse transcriptase polymerase chain reaction (RT-PCR) and nucleic acid sequence based amplification assay (NASBA) have been developed to improve the sensitivity and specificity of ante as well as post-mortem diagnosis of rabies. The aim of this study was to detect the rabies virus (RABV) presence in different organs of frugivorous bats using molecular techniques such as RT‐PCR, hnRT‐PCR, and the Real Time RT‐PCR. Thirty bats of the genus Artibeus were selected and resulted as positive by the DFA test and N2A‐cells inoculation test using brain tissue in both tests. Samples of salivary gland tissue, urinary bladder tissue, kidney tissue, lung tissue, stool, and skull lavage were collected for molecular assays.
