Mammary carcinosarcoma in cat: a case report 45

A case of mammary carcinosarcoma is reported in a 13-year-old, mixed breed female cat, which was not spayed and had not received contraceptives. The patient presented extensive and coalescent nodules in all mammary glands. Based on the histological and immunohistochemical findings, the diagnosis of mammary carcinosarcoma was confirmed.

Pleomorphic lobular carcinoma of the canine mammary gland: histopathologic and immunohistochemical features 45

This report describes the morphological and immunohistochemical findings of a case of mammary gland pleomorphic lobular carcinoma occurring in the canine species. Histologically, it was characterized by the presence of tumor cells loosely dispersed in the stroma or arranged in a linear pattern showing relatively abundant eosinophilic cytoplasm with an eccentric nuclei. These morphological features, characteristic of pleomorphic mammary gland lobular carcinoma in woman, were not previously described in dogs.

Epidemiological, clinical and pathological evaluation of overall survival in canines with mammary neoplasms 45

ABSTRACT Canine mammary neoplasms (CMNs) are the most frequent lesions and in female dogs. However, studies correlating pathological criteria with clinical evolution in female dogs with mammary neoplasms are scarce. The present study aims to present epidemiological, clinical-pathological and overall survival data to help establish the prognosis and understand the biological behavior of CMNs. A total of 1539 cases were included (85% malignant and 13% benign). Tumor size was an important prognostic factor and was associated with overall patient survival (P< 0.0001). Association of nucleolar organizing regions and Ki-67 expression with recurrence rate of hair follicle tumor in dogs 45

Mitotic index, nuclear diameter, number of nucleolar organizing regions, and Ki-67 expression, in hair follicle tumors of 82 dogs were evaluated. Tissue specimens were used to prepare sections for histological staining for number of nucleolar organizing region and immunohistochemical staining for Ki-67. Tumors were classified as trichoblastoma (n=32), benign trichoepithelioma (n=30), pilomatricoma (n=7), malignant trichoepithelioma (n=6), infundibular keratinizing acanthoma (n=5), and tricholemmoma (n=2). Subcutaneous administration of ketoprofen delays Ehrlich solid tumor growth in mice 45

Ketoprofen, a nonsteroidal anti-inflammatory drug (NSAID) has proven to exert anti-inflammatory, anti-proliferative and anti-angiogenic activities in both neoplastic and non-neoplastic conditions. We investigated the effects of this compound on tumor development in Swiss mice previously inoculated with Ehrlich tumor cells. To carry out this study the solid tumor was obtained from cells of the ascites fluid of Ehrlich tumor re-suspended in physiological saline to give 2.5x106cells in 0.05mL. After tumor inoculation, the animals were separated into two groups (n = 10). A 3-year-old female Pinscher dog was presented with a history of lameness and pain in hind limbs for one week which acutely progressed to non-ambulatory paraparesis. The dog had been apathetic and anorexic during the six days prior to presentation. The neurological findings were compatible with upper motor neuron signs to the left hind limb due to a spinal cord compressive mass on the twelfth thoracic vertebral body. Protocol for the anatomopathological examination of canine mammary tumors 45

Foi elaborado um protocolo para exame anatomopatológico de tumores de mama em cães, constituído de três partes: requisição, exame clínico e laudo histopatológico. O exame clínico contém dados sobre a descrição macroscópica da lesão. O laudo histopatológico constituiu-se de campos para descrição microscópica pormenorizada das lesões e classificação da principal lesão observada. A elaboração do protocolo tem como objetivo estabelecer critérios para estudos e pesquisas sobre neoplasias mamárias em animais e auxiliar no diagnóstico e prognóstico de lesões mamárias.

DNA flow cytometry of canine mammary tumors: comparative aspects with human breast tumors 45

Flow cytometric analysis of DNA content was performed on 28 samples of canine mammary tumors. Nine of them were benign and 19 were malignant. All benign tumors and 11 malignant tumors (57.9%) were diploid (P<0.05). Form the aneuploid tumors, five (26.3%) were hyperdiploid, one (5.3%) hypodiploid, one (5.3%) near triploid and one (5.3%) multiploid. The analysis of the expression of the markers PR and CD31 revealed a significant difference between diploid and aneuploid tumors (P<0.05). The immunoreactivity of PR was higher in diploid tumors, while the immunoreactivity of CD31 was stronger in aneuploid tumors. Uma chimpanzé de 22 anos de idade foi necropsiada com histórico clínico de anorexia, vômitos freqüentes e desidratação conseqüentes à gastrite iatrogênica. Macroscopicamente, o útero apresentava-se aumentado de volume, com o lúmen totalmente ocluído por nódulos firmes, esbranquiçados e coalescentes que se estendiam para o miométrio. Histologicamente, os nódulos eram constituídos por leiomiócitos bem diferenciados dispostos em várias direções e com coloração característica pelo tricrômio de Gomori e Masson. Twenty BALB/c mice were inoculated with cell suspension of Ehrlich tumor. Ten mice were inoculated in the cushion plant (solid form) and the other 10 in the peritoneum (ascitic form). Animals were euthanized on different times (7 and 14 days). Cytological and histological slides, immunohistochemical (PCNA) analysis and NORs silver impregnation technique were performed. The results showed more proliferation on the 7th day in the ascitic form and on 14th day in the solid form, using both analyses (PCNA and AgNORs). Estudou-se atividade antineoplásica de um produto natural isolado de Alomia myriadenia (miriadenolídeo) no modelo do tumor de Ehrlich em camundongos. Cell proliferation markers in the transplanted canine transmissible venereal tumor 45

Adult male mongrel dogs were subcutaneously transplanted with the canine transmissible venereal tumor (TVT) on the hypogastric region. Twelve specimens of tumors were collected, half during the proliferative phase and the other half during the regressive phase. Fragments of the tumor were fixed in 10% buffered formalin and routinely processed for light microscopy. Sections of 4µm were stained by Schorr or AgNOR or either immunostained for MIB1 (Ki67). Schorr stain, AgNOR and MIB1 showed an increased proliferative activity through mitotic index, nuclear argyrophilic protein stain and cycling tumoral cells in the growing tumors, respectively. Spindle cell sarcoma in Bothrops leucurus 45

O presente relato descreve os achados anatomopatológicos de uma neoplasia maligna em jararaca (Bothrops leucurus), mantida em cativeiro durante sete anos. O animal apresentava massa nodular subcutânea localizada no lado direito do terço anterior. O exame histológico revelou tratar-se de neoplasia mesenquimal maligna, permitindo o diagnóstico de sarcoma fusocelular.

A new methodology for the improvement of diagnostic immunohistochemistry in canine veterinary pathology: automated system using human monoclonal and polyclonal antibodies 45

The authors describe their experience with an automated immunohistochemical system applied to canine tissue samples. Twenty human cellular markers specific monoclonal and polyclonal antibodies and two different antigen retrieval methods were used in normal and neoplastic breast tissue, as well as skin samples obtained from female dogs of pure and mixed breeds. The antibodies tested were the most frequently used in human and veterinary medicine studies, employed with diagnostic purposes in breast pathology, as well as in cancer research. Cystadenocarcinoma simulating hemangiosarcoma of the salivary gland in dog: case report 45

Cystadenocarcinoma is regarded as a rare adenocarcinoma variant in animals. This work reports the case of an 8-year-old female Poodle dog with salivary gland cystadenocarcinoma with morphological characteristics similar to a hemangiosarcoma. Histopathological analysis showed a tumor mass with cystic formations containing a large amount of red blood cells. In order to distinguish these two entities, periodic acid Schiff (PAS) staining and immunohistochemical analysis were carried out with the use of cytokeratin AE1/AE3 (CK) and CD31-specific antibodies. Neoplastic cells were PAS-negative, CK-positive and CD31-negative confirming their epithelial origin. Based on the findings, the diagnosis of high grade cystadenocarcinoma was established.

Invasive micropapillary carcinoma of the dog mammary gland: a case report 45

This report describes the morphological and immunohistochemical findings of two cases of breast invasive micropapillary carcinoma occurring in dogs. Histologically, the tumors are characterized by the presence of numerous irregular cystic formations filled out with nests of epithelial cells that exhibit a micropapillary pattern. These morphological features are characteristic of invasive micropapillary carcinoma in woman, a breast tumor not previously described in dogs.

Assessment of cell proliferation and prognostic factors in canine mammary gland tumors 45

Three methods for the analysis of cell proliferation, mitotic index/10 high-power fields (10 HPF), mitotic index/four sets of 10 HPF (40 HPF), and MIB-1 index were evaluated in a series of canine mammary gland tumors, as well as the possible correlation between them. Fifty-six canine mammary gland tumors, including 23 benign and 33 malignant, were studied. Model of induction of thyroid dysfunctions in adult female mice 45

It is described the elaboration of a protocol to induce hyperthyroidism and hypothyroidism in mice by administrating thyroxin and propylthiouracil, respectively, in the drinking water. The drugs were administered to adult female mice of the Swiss strain for 30 days in order to obtain a systemic status of thyroid dysfunction. The induction of hyperthyroidism and hypothyroidism in the animals was confirmed by the histomorphological analysis of the thyroid in the end of the experiment, when the state of gland dysfunction in the animals submitted to the treatment was observed.
