A cinomose é uma doença viral altamente contagiosa que afeta o sistema respiratório, o sistema gastrointestinal e o sistema nervoso central (SNC) dos caninos e felinos. Devido à grande diversidade de sinais clínicos observados nos animais afetados, a confirmação laboratorial é fundamental para o diagnóstico definitivo da doença. Este trabalho teve por objetivo comparar as técnicas utilizadas no diagnóstico laboratorial da cinomose canina. Para tanto foram selecionadas 20 amostras de SNC de caninos domésticos, enviados ao Laboratório de Virologia da Seção de Diagnóstico da Raiva do Instituto Pasteur.  Destas amostras, 7 foram provenientes de animais sem histórico clinico de  doença e 13 foram provenientes de animais com manifestação de sinais clínicos ...

The aim of this study was to develop a new mechanism for antiviral therapy against rabies based on the introduction by transfection with a cationic reagent (lipofectamine 2000) of antibodies into neuronal cells infected with the rabies virus. N2A cells were infected using 96-well plates and different viral concentrations (0.1, 1.0, 10 and 100 TCID50) of three lineages of rabies virus circulating in Brazil (dog, Desmodus rotundus and Eptesicus furinalis). This study describes the comparison of in vivo and in vitro transfection of antibodies to use as a new mechanism for antiviral therapy against rabies. The transfection was made in vitro with neuroblastoma murine cell line (Neuro‐2a) and in vivo with 21 years old mice. Both cells and mice were infected with different infectious doses (0.1; 1.0; 10 and 100 ID50) of the rabies virus. The identification of animal species that transmit pathogens such as the rabies virus is of the utmost importance for public health and the natural history of infectious and contagious diseases. The São Paulo State Rabies Control Program, coordinated by the Pasteur Institute of São Paulo, has been involved in endeavors to improve the epidemiological surveillance of rabies in dogs, cats and bats, particularly in urban areas. Since 1998, when the last case of canine rabies due to antigenic variant 2 occurred in the state, the Institute has received 66,352 canine central nervous system samples, 17,796 samples from cats and 38,133 from bat specimens. Using conventional techniques for rabies diagnosis, 11 dogs, 8 cats and 554 bats were found to be positive for this disease. Bats are considered important reservoirs of rabies virus, which is paramount in the study of the pathogenesis of this zoonosis disease, through the use of sensitive systems. The presence of viral antigen in these species shows that non-neuronal viral spread is efficient in different organs that participate effectively in the elimination of rabies virus, such as, salivary glands and bladder. Antigenic variant 2 of the Rabies Virus (RABV), typically found in dogs in Brazil, still circulates in the North and Northeast of Brazil in spite of mass vaccination campaigns against rabies in domestic animals. Previous studies have shown that this variant is made up of two main genetic lineages: one found in dogs (Canis familiaris) and another typically found in wild canids, particularly the crab-eating fox (Cerdocyon thous), which is known in Brazil as cachorro do mato. Rabies is an enzootic disease in Brazil, including Rio de Janeiro state, especially rabies transmitted by vampire bats. All years are accounted large losses of livestock due to this disease in the State. The aim of this study was perform a phylogeographic analysis of rabies virus in the Rio de Janeiro state. It was analyzed a dataset of 58 genetic sequences from the coding region of the N gene. Rabies is a worldwide zoonotic disease caused by rabies virus (RABV). According to the World Health Organization, more than 55,000 people still die of rabies every year. The domestic dogs are the major responsible for transmission of the virus for people.The human rabies transmitted by dog is considered a neglected disease.
