Pressure and cold shock induction of meiotic gynogenesis and triploidy in the European sea bass, Dicentrarchus labrax L.: relative efficiency of methods and parental variability ArchiMer

The optimal conditions for the retention of the second polar body in sea bass eggs were investigated by altering the timing, intensity and duration of application of pressure and cold shocks. Treatment optima for cold shocks were 0-1 degrees C for 15-20 min at 5 min after fertilisation (a.f.) and 8500 psi for 2 min at 6 min a.f. for pressure shocks. Meiogenesis was obtained by fertilising eggs with UV-irradiated homologous sperm (32,000 erg mm(-2)) and pressure or cold shocking eggs as above. 100% triploidy was induced following definition of liable periods for the disruption of the meiotic spindle obtained in gynogenesis. Measuring individual feed efficiency and its correlations with performance traits in Nile tilapia, Oreochromis niloticus ArchiMer

Estimating individual feed intake of fish held in groups has long been a challenge precluding precise knowledge of the individual variation of feed efficiency (FE) in fish. In this study, counts of the number of feed pellets (1.63 mg on average) eaten by individual tilapia (Oreochromis niloticus) held in 8 mixed sex groups of 15 fish were measured from video recordings made over a period of 10 days where fish were fed twice daily to achieve compensatory growth after 10 days of fasting. The initial body weight of the fish was 9.77 ± 2.03 g. Accumulated measures of feed intake (FI) over 11 meals were found to achieve 95% repeatability and a high accuracy of estimation of FI. Heritability of Boldness and Hypoxia Avoidance in European Seabass, Dicentrarchus labrax ArchiMer

To understand the genetic basis of coping style in European seabass, fish from a full factorial mating (10 females x 50 males) were reared in common garden and individually tagged. Individuals coping style was characterized through behavior tests at four different ages, categorizing fish into proactive or reactive: a hypoxia avoidance test (at 255 days post hatching, dph) and 3 risk-taking tests (at 276, 286 and 304 dph). We observed significant heritability of the coping style, higher for the average of risk-taking scores (h2 = 0.45 ± 0.14) than for the hypoxia avoidance test (h2 = 0.19 ± 0.10). Application of DNA parentage assignment for commercial selection in aquaculture is still rare. An experiment was performed to quantify and to minimize maternal effects on growth in rainbow trout. Six hundred families were reared until 198 day post fertilization (dpf) according to two different procedures. In the first procedure (NORM), all families were pooled at eyed stage. Little information is available on whether swimbladder inflation has a genetic basis for its expression too. In this work, 24 full-sibling sea bass families from a 4 dams x 6 sires factorial crossing were reared under communal conditions. Feed conversion ratio (FCR), the ratio between feed intake and body weight gain, is of major interest for improving aquaculture sustainability through reduced feed costs and environmental impacts. Demonstrating whether FCR measured in juvenile fish is an accurate predictor of their performance during the whole rearing period is critical to developing genetic improvement programs for this trait. Heritabilities and GxE interactions for growth in the European sea bass (Dicentrarchus labrax L.) using a marker-based pedigree ArchiMer

253 full-sib families from 33 males and 23 females of European seabass were produced in a partly factorial mating design. All fish were reared in the same tank during 14 months, then 7000 of them were dispatched in four farms to different locations (France, Israel, Italy, Portugal) representing a wide variety of environmental conditions. Around 400 g mean weight, 1177 to 1667 fish in each site were weighed and length was measured. Condition factor (K) was calculated. Combining Individual Phenotypes of Feed Intake With Genomic Data to Improve Feed Efficiency in Sea Bass ArchiMer

Measuring individual feed intake of fish in farms is complex and precludes selective breeding for feed conversion ratio (FCR). Here, we estimated the individual FCR of 588 sea bass using individual rearing under restricted feeding. These fish were also phenotyped for their weight loss at fasting and muscle fat content that were possibly linked to FCR. The 588 fish were derived from a full factorial mating between parental lines divergently selected for high (F+) or low (F–) weight loss at fasting. It has been achieved with variable success in a number of freshwater species and can be attained by artificial fertilization of genetically inactivated eggs following exposure to gamma (gamma), X-ray or UV irradiation (haploid androgenesis) and by restoration of diploidy by suppression of mitosis using a pressure or thermal shock.
