An alkaline single-cell gel electrophoresis (comet) assay for environmental biomonitoring with native rodents Genet. Mol. Biol.
Silva,Juliana da; Freitas,Thales R.O. de; Marinho,Jorge R.; Speit,Günter; Erdtmann,Bernardo.
The main advantages of single-cell gel electrophoresis (SCG) are its applicability to any eukaryotic organism and cell type, its low cost and the short time required to obtain results. These properties make the SCG assay particularly useful in screening for environmental genotoxicity. The present study describes a modified version of this technique for use in field work with native rodents and examines some factors which influence the outcome of the assay. Wild rodents (Ctenomys torquatus, "tuco-tuco") from a region close to a strip coal mine and from a region with no coal mines were used. Animals from the coal mining region had significantly more DNA damage than those from the control area. The use of this SCG technique for direct sampling in the field...

Evaluation of occupational genotoxic risk in a Brazilian hospital Genet. Mol. Biol.
Maluf,Sharbel Weidner; Erdtmann,Bernardo.
Many therapeutic, diagnostic and prophylactic procedures used in hospitals are of potential genetic risk. An evaluation was made of genotoxic occupational risk in 42 workers from the Hospital de Clínicas de Porto Alegre, RS, Brazil, who had been occupationally exposed to lead (solder), ethylene oxide (sterilization area), antineoplastic drugs (nurses and pharmacists) or ionizing radiation. They were compared with 42 unexposed individuals. There was an increase in the frequency of binucleated cytochalasin-blocked lymphocytes with micronuclei, though it was not significant (P = 0.058). The groups exposed to antineoplastic drugs and radiation had a significant increase in micronuclei frequency (P = 0.038 and P = 0.022, respectively). The high frequencies of...

Genotoxicity of Nicotiana tabacum leaves on Helix aspersa Genet. Mol. Biol.
Silva,Fernanda R. da; Erdtmann,Bernardo; Dalpiaz,Tiago; Nunes,Emilene; Ferraz,Alexandre; Martins,Tales L.C.; Dias,Johny F.; Rosa,Darlan P. da; Porawskie,Marilene; Bona,Silvia; Silva,Juliana da.
Tobacco farmers are routinely exposed to complex mixtures of inorganic and organic chemicals present in tobacco leaves. In this study, we examined the genotoxicity of tobacco leaves in the snail Helix aspersa as a measure of the risk to human health. DNA damage was evaluated using the micronucleus test and the Comet assay and the concentration of cytochrome P450 enzymes was estimated. Two groups of snails were studied: one fed on tobacco leaves and one fed on lettuce (Lactuca sativa L) leaves (control group). All of the snails received leaves (tobacco and lettuce leaves were the only food provided) and water ad libitum. Hemolymph cells were collected after 0, 24, 48 and 72 h. The Comet assay and micronucleus test showed that exposure to tobacco leaves for...

Cashew (Anacardium occidentale) apple juice lowers mutagenicity of aflatoxin B1 in S. typhimurium TA102 Genet. Mol. Biol.
Cavalcante,Ana Amélia Melo; Rübensam,Gabriel; Erdtmann,Bernardo; Brendel,Martin; Henriques,João A.P..
Cashew (Anacardium occidentale) is a medicinal plant native to Brazil and also yields a nutritious fruit juice. Its large pulpy pseudo-fruit, referred to as the cashew apple, contains high concentrations of vitamin C, carotenoids, phenolic compounds and minerals. Natural and processed cashew apple juice (CAJ/cajuina) are amongst the most popular juices in Brazil, especially in the north-east. Both juices have antioxidant potential and suppress mutagenicity of hydrogen peroxide. In the present study we evaluated the inhibitory effects of CAJ/cajuina on Aflatoxin B1(AFB1)-induced mutation, using the Salmonella/microsome assay with the experimental approaches of pre-, co- and post-treatments. Both CAJ/cajuina suppress AFB1-induced mutagenesis in strain TA102...

Estudos epidemiológicos entre populações indígenas da Amazônia. II. Prevalências da microfilaremia de Mansonella ozzardi: comparação de dois métodos de diagnóstico () Acta Amazonica
Lawrence,Dale N.; Erdtmann,Bernardo; Peet,Jennifer W.; Mello,José A. Nunes de; Healy,George R.; Neel,James V.; Salzano,Francisco M..

Clastogenic activity of integerrimine determined in mouse micronucleus assays Braz. J. Genet.
Gimmler-Luz,Maria Clara; Erdtmann,Bernardo.
The pyrrolizidine alkaloid integerrimine, obtained from Senecio brasiliensis, was tested by acute dosing at two concentrations (18.75 and 37.5 mg/kg), and at different times, to establish its ability to induce micronuclei in mouse erythrocytes. This alkaloid was able to increase the frequency of micronucleated polychromatic erythrocytes in both, bone marrow and peripheral blood erythrocytes
