Experimental antiviral therapy against different rabies virus lineages using transfection with anti-rabies antibodies MV&Z

The aim of this study was to develop a new mechanism for antiviral therapy against rabies based on the introduction by transfection with a cationic reagent (lipofectamine 2000) of antibodies into neuronal cells infected with the rabies virus. N2A cells were infected using 96-well plates and different viral concentrations (0.1, 1.0, 10 and 100 TCID50) of three lineages of rabies virus circulating in Brazil (dog, Desmodus rotundus and Eptesicus furinalis). The diversity of bat species in Brazil is great, and there are 172 species distributed among nine families. The Phyllostomidae family is the most numerous, followed by Vespertilionidae and Molossidae. According to feeding habits, the majority of bats are insectivorous, followed by frugivorous, nectarivorous, carnivorous and hematophagous. The identification of animal species that transmit pathogens such as the rabies virus is of the utmost importance for public health and the natural history of infectious and contagious diseases. The São Paulo State Rabies Control Program, coordinated by the Pasteur Institute of São Paulo, has been involved in endeavors to improve the epidemiological surveillance of rabies in dogs, cats and bats, particularly in urban areas. Since 1998, when the last case of canine rabies due to antigenic variant 2 occurred in the state, the Institute has received 66,352 canine central nervous system samples, 17,796 samples from cats and 38,133 from bat specimens. Using conventional techniques for rabies diagnosis, 11 dogs, 8 cats and 554 bats were found to be positive for this disease. Bats are considered important reservoirs of rabies virus, which is paramount in the study of the pathogenesis of this zoonosis disease, through the use of sensitive systems. The presence of viral antigen in these species shows that non-neuronal viral spread is efficient in different organs that participate effectively in the elimination of rabies virus, such as, salivary glands and bladder. Rabies is an enzootic disease in Brazil, including Rio de Janeiro state, especially rabies transmitted by vampire bats. All years are accounted large losses of livestock due to this disease in the State. The aim of this study was perform a phylogeographic analysis of rabies virus in the Rio de Janeiro state. It was analyzed a dataset of 58 genetic sequences from the coding region of the N gene. We isolated 35 samples in the Rio de Janeiro between 2001 and 2006 from vampire bats and domestic herbivores in 18 cities of the State. Human rabies is still an important public health problem in some Brazilian regions. Usually, ante-mortem diagnosis of rabies is made by demonstration of virus antigen by direct immunofluorescence in corneal or conjunctival smears and skin biopsies; however, this technique has a low sensitivity. Recently, molecular techniques such as the reverse transcriptase polymerase chain reaction (RT-PCR) and nucleic acid sequence based amplification assay (NASBA) have been developed to improve the sensitivity and specificity of ante as well as post-mortem diagnosis of rabies. O Estado de São Paulo (ESP) coordena a Campanha de Vacinação contra a raiva em cães e gatos desde 1975. No inicio da década de 80, atingiu-se 80% de cobertura vacinal (1 cão para 10 pessoas), com a participação de todos os municípios. A vacina utilizada no período 1975 a 2009 foi tipo Fuenzalida & Palácios. O imunobiológico utilizado nos estados é fornecido pelo Ministério da Saúde (MS), que decidiu em 2010 substituir a vacina por uma produzida em culturas de células BHK, que não deveria causar reação adversa neurológica e propiciaria uma melhor resposta imunológica.
