Anomalous somatic embryos in Acca sellowiana (O. Berg) Burret (Myrtaceae) Rev. Bras. Bot.

Somatic embryogenesis represents a valuable tool for the studies on the basic aspects of plant embryo development. Today this process is used as a potencial technique for large-scale plant micropropagation although, so far, it has been applied to only a small number of species. However, when somatic embryos are malformed they are considered economically useless. In Acca sellowiana (O. Berg) Burret, an important fruit-producing crop, large amounts of anomalous somatic embryos (76.3%) were found just after 40 days of culture of explants in a 2,4-D containing medium. Among the anomalous forms found in the cotiledonary stage, 12.2% consisted of fused embryos, 40.4% displayed fused cotyledons, 13.0% presented supernumerary cotyledons, and 10.7% showed absence...

Anatomical and ultrastructural aspects of leaf galls in Ficus microcarpa L.f. (Moraceae) induced by Gynaikothrips ficorum Marchal (Thysanoptera) Acta Botanica

Several species of Ficus present leaf galls and the goal of this research is to study the structural alterations involved in the formation of leaf galls caused by Gynaikothrips ficorum on F. microcarpa, an ornamental plant. The galls of young and mature leaves were separated into two developmental stages based on the presence of lesions on leaf lamina and the degree of leaf folding. Swellings of the lamina were observed in young and mature leaves during gall development which coincided with the areas of cellular hypertrophy and tissue hyperplasia. Swellings were detected in a greater amount and more precociously on young leaves when compared to mature ones. Métodos de coloração de Roeser (1972): modificado - e Kropp (1972) visando a substituição do azul de astra por azul de alcião 8GS ou 8GX Acta Botanica

Em folhas de Lychnophora ericoides, Coffea arabica e Nymphaea mexicana foram testados métodos de coloração baseados em Roeser (1972) modificado e Kropp (1972), visando a substituição do corante azul de astra por azul de alcião 8GS ou 8GX. As amostras foram fixadas em FAA, desidratadas em série butílica terciária e incluídas em parafina. Os cortes histológicos transversais foram corados segundo diferentes baterias de coloração, modificadas quanto ao tipo de corante usado, diferenciador e série de desidratação. As lâminas permanentes foram preparadas com bálsamo-do-canadá sintético. Os resultados obtidos evidenciaram que o método de Roeser (1972) modificado é melhor que o de Kropp (1972), nas condições deste experimento. O azul de astra pode ser substituído...

Structural aspects of the zygotic embryogenesis of Acca sellowiana (O. Berg) Burret (Myrtaceae) Acta Botanica

Acca sellowiana has anatropous, bitegmic and crassinucellate ovules. The outer and inner integuments are double-layered except in the micropyle, where they are composed of more layers; the micropyle is zig-zag shaped. The egg apparatus lies at the micropylar pole, and the zynergids present a conspicuous filiform apparatus. The antipodal cells are present in the chalazal region, persisting before the occurrence of double fertilization. The zygote is visible 21 days after pollination; nuclear endosperm is already present. The first mitotic division of the zygote occurs at 24th day. Endogenous auxin and cytokinin contents associated with shoot formation in leaves of pineapple cultured in vitro Braz. J. Plant Physiol.

The in vitro culture of pineapple leaves on a shoot induction medium (SIM) results in the formation of protuberances and further development in shoots, and plantlets. The contents of endogenous indoleacetic acid (IAA) and five cytokinins (Cks), N6(2-isopentenyl)adenine (iP), N6(2-isopentenyl)adenosine (iPR), zeatin (Z), zeatin riboside (ZR) and N6-benzyladenine (BA), present in the basal portion of those leaves, were correlated to the organogenic response that occurs over 15 days of culture. The endogenous auxin/cytokinins ratio was lowest on the 3rd day, mainly due to a strong increase in the iP level.
